Chongqing Medicine 2017;46(13):1753-1755
Objective To observe the protective effect of GSPB2 for delaying the human endothelial cells apoptosis with H2O2 induced human endothelial cells apoptosis as the model and to investigate its mechanism. Methods The human endothelial cells were pre-treated by 0.5 mmol H2O2,then different concentrations of GSPB2(5.0,10.0,20.0 μmol/L) was added for protecting cells. The cellular apoptosis was detected by TUNEL method;the apoptosis related molecules, such as Caspase-3,Bax,Bcl-2,and influencing apoptosis related molecules such as PI3K,p-Akt and Akt protein expression were detected by Western bl0t;Transwell chamber was used to detect cell migration situation. Results 0.5 mmol H2O2 induced obvious increase of human endothelial cells (P<0.01).GSPB2 could significantly alleviate the H2O2 induced apoptosis of human endothelial cells(P<0.05).10 μmol/mL GSPB2 could be close to return to H2O2 untreated control group level.Further found that Caspase-3,Bax protein expression were up-regulaed,Bcl-2 expression was down-regulated and the expression of PI3K,p-Akt and Akt wa sup-regulated.Conclusion The different concentrations of GSPB2 has a protective effect on vascular endothelial cells induced by H2O2,moreover which manifested by the concentration and time dependence. Its mechanism is related with the related molecules PI3K,p-Akt and Akt
2.Concentration of hyaluronic acid in peritoneal fluid in patients with gastric carcinoma and its clinical significance
Academic Journal of Second Military Medical University 1981;0(03):-
Objective: To study the relationship between the level of hyaluronic acid(HA) in peritoneal fluid in patients with gastric cancer and the invasion or diffusion of the cancer. Methods: The concentration of peritoneal fluid hyaluronic acid (PFHA) was determined by RIA in 48 patients with gastric cancer prior to and after operation. The PFHA from 23 non carcinoma patients were taken as control. The peritoneal fluid was obtained by the implantable Infuse Port. Results: The average PFHA level of gastric cancer group[(913.01?170.69) ?g/?g] was significantly higher than that of the non carcinoma control [(362.39?168.82) ?g/?g]. The PFHA concentration were significantly higher in groups of pTNM Ⅲ+Ⅳ [(1 094.42?127.65) ?g/?g] than that in groups of pTNM Ⅰ+Ⅱ [(814.03?232.22) ?g/?g]. The PFHA level in the radical resection group was significantly reduced a month after operation, and there were no significantly changes in patients received palliative resection. Conclusion: These findings suggest that the elevation of PFHA may be related to the peritoneal invasion and diffusion potential of gastric cancer. The PFHA level shows a decreasing tendency in patients received radical resection.

To study the status of homozygous deletions of MTS1/p16 gene in squamous cell carcinoma of the larynx (LSCC) and to discuss the relations of homozygous deletions to the pathological grade and clinical stage of the tumor,Fifty-nine primary LSCC were examined for homozygous deletions of the MTS1/p16 gene by comparative PCR.Three specimens were eliminated for being incapable of comparison with others.Homozygous deletions of the MTS1/p16 gene were found in 9 of 56 tumors (16.07%).The rates of homozygous deletions of MTS1/p16 gene in well, medially, poorly differentiated tumors were 17.24%(5/29),11.76%(2/17),and 20.00%(2/10) respectively. There had no significant difference in various pathological grades of tumors.Homozygous deletions rates of tumors in stagesⅠ~Ⅱand in stagesⅢ~Ⅳwere 5.41% (2/37) and 36.84%(7/19) respectively. The rate of homozygous deletions in stages Ⅲ~Ⅳtumors was significantly higher than that in stagesⅠ~Ⅱ(P<0.01).Homozygous deletions of the MTS1/p16 gene were correlated well with clinical stages. Our data suggested that homozygous deletions of MTS1/p16 gene might be one of the genetic events in the development of the tumor and might play a role in malignant progression of LSCC.

Journal of Applied Clinical Pediatrics 1993;0(03):-
Objective To explore the clinical and laboratory characteristics of langerhans cell histiocytosis (LCH) in children, so as to improve diagnosis level and decrease misdiagnosis rate. Methods Twenty-five cases of LCH from Jan.1996 to Feb.2006 were analyzed by retrospective study. The clinical data were collected and abstracted for information regarding clinical symptom, physical sign, laboratory examination, imaging, pathology, diagnosis and treatment. Results Some laboratory findings in hemogram, bone marrow examination and chest X-ray were non-specific.The X-ray characteristic of skeleton was osteolysis. Computerized tomography and magnetic resonance imaging were important in defining the extent of lesion in fundus cranii and sella.Seven cases were examined for anti-Epstein-Barr virus(EBV) IgM, 3 cases were positive;5 and 3 cases out of 10 cases showed humoral and cellular immunity abnormality, respectively. The misdiagnosis rate was 52%,1 case had been misdiagnosed for 7 years. Chemotherapy was effective in the future. Conclusions The clinical manifestations of LCH varies widely, leading to high rate of misdiagnosis. The etiology of LCH is unclear, and some of our patients show the evidence of EBV infection or immunity abnormality. Definitive diagnosis of LCH is based on pathology. Ultrastructure and immunophenotype should be done to improve diagnosis level.
