1.Role of Haptoglobin in the Immunomodulatory in Patients with Premature Coronary Heart Disease

Objective To analyse the role of haptoglobin in the immunomodulatory in patients with premature coronary heart disease.Methods Clinical data of 60 premature coronary heart disease patients and 40 healthy people was analysed.The general information and serume haptoglobin,hs-CRP and IL-6 were compared.Results There were no statistically difference between the two groups of the general information such as sex,years,BMI,hypertension and diabetes (P>0.05),excepts the smoking history(27/33 vs 9/31,χ2=3.324,P<0.05).Hp,hs-CRP and IL-6 levels in the observer group were signifi-cantly higher than the control group,the difference was statistically significant (97.76±23.08 mg/L vs 65.81±19.42 mg/L,10.43±3.81 mg/L vs 3.05±0.94 mg/L,16.43±4.59 pg/L vs 4.62±1.83 pg/L,P<0.001).Correlation analysis showed that in the observer group Hp and hs-CRP showed a significant positive correlation (r=0.518,P<0.001),and the same results was found between IL-6 and Hp (r= 0.291,P=0.024).Conclusion Haptoglobin level was significantly in-creased in patients with coronary heart disease,which suggested that haptoglobin played a role in the development of coro-nary heart disease,and its level was correlated with IL-6.

3.Forensic Validation of the Goldeneye™ DNA ID 25A Kit.

OBJECTIVE:
To test and estimate the forensic application of Goldeneye™ DNA ID 25A Kit.
METHODS:
The kit was validated by a series of tests for accuracy, sensitivity, consistency, peak height balance, stability, and mixed samples through measured blood samples and other samples in routine casework.
RESULTS:
The peak height balance of the different loci was ≥ 42%. The genotyping results of the positive control DNA was accurate. The complete STR genotyping result could be obtained from 0.125 ng positive control DNA.
CONCLUSION
Goldeneye™ DNA ID 25A Kit is suitable for criminal cases and DNA database in forensic practice.

4.Instant effects of temperature on mechanical properties of single living intact human red blood cell

AIM: To study the instant effect of circumstance temperature on mechanical properties of single living intact human red blood cell. METHODS: The shape, size, membrane bending elastic modulus and shear elastic modulus of single living intact red blood cell was determined with non-invasive, in situ, real time by employing a static micro-image analyzing and a dynamic micro-image analyzing system. RESULTS: The contact area and diameter of red blood cells were decreased as the temperature increased from 35 ℃ to 43 ℃. The membrane bending elastic modulus and shear elastic modulus of single living intact red blood cell were the least at physiological temperature 37 ℃. CONCLUSION: Red blood cells are easy to carry out their physiological functions such as carrying oxygen to all of the body with the best shape and mechanical deformation at 37 ℃.
5.Clinical Study on Acupuncture for Post-stroke Myodystonia

Objective To observe the clinical efficacy of acupuncture plus contralateral needling in treating post-stoke myodystonia. Method Totally 120 patients with post-stroke myodystonia were randomized into group A, B, C and D, 30 in each group. Group A was intervened by yin-reducing and yang-tonifying needling method plus contralateral needling; group B was by ordinary acupuncture plus contralateral needling, group C was by yin-reducing and yang-tonifying needling alone, while group D by ordinary acupuncture. Clinical Spasticity Index (CSI) was observed before and after intervention. Result After treatment, the CSI scores (tendon reflex, muscle tension, episodic spasm scores and total score) were significantly changed in the four groups (P<0.01, P<0.05). The tendon reflex score, muscle tension score and total score in group A were significantly different from that in the other three groups after intervention (P<0.01,P<0.05). There was a significant difference in comparing the episodic spasm score between group A and D after intervention (P<0.01). There were significant differences in comparing the tendon reflex score, muscle tension score and total score between group B and D after intervention (P<0.05). The total score in group C was markedly different from that in group D after intervention (P<0.05). Conclusion Yin-reducing and yang-tonifying needling method plus contralateral needling is an effective method in treating post-stroke myodystonia.
6.Application of the Paediatric Yorkhill Malnutrition Score in nutritional risk screening in children hospitalized in general surgery department

Objective To investigate the status of nutritional risk and clinical outcome in children hospitalized in the general surgical department, which can provide basis for clinical nutritional support and selecting the screening tool. Methods Nutritional risk screening was performed on 506 children hospitalized in the general surgical department by using the Paediatric Yorkhill Malnutrition Score (PYMS). The data on incidence of complications, parenteral nutritional support during hospitalization, length of hospital stay and total hospital expenses were recorded. Results Of the 506 cases, 64.6%(327/506) had low nutritional risk, 27.1%(137/506) had moderate nutritional risk, and 8.3%(42/506) had high nutritional risk. Hepatic and gall diseases were the most common types of moderate nutritional risk (55.2%, 16/29), intestinal obstruction were the most common types of high nutritional risk (23.1%, 6/26). The incidence of complications, parenteral nutritional support during hospitalization, length of hospital stay and total hospital expenses were 10.2%(14/137), 21.9%(30/137), (9.54±4.95) d, (6 899.13±3 281.18) yuan RMB and 21.4%(9/42), 35.7%(15/42), (10.60±7.25) d, (7 521.64±6 026.23) yuan RMB in moderate or high nutritional risk children, and 0.9%(3/327), 3.1%(10/327), (17.17 ± 4.17) d, (4 691.96 ± 3 114.43) yuan RMB in low nutritional risk children, and there were significant differences (χ2/F=18.665-64.554, P<0.05). Conclusions Moderate or high nutritional risk is seen in children hospitalized in the surgical department. High nutritional risk score is correlated with adverse clinical outcome. PYMS can be used for nutritional risk screening in hospitalized children.
7.Application of nutritional risk screening in children hospitalized in general surgery department

Objective To evaluate the application of nutritional risk screening in children hospitalized in general surgery department,in order to provide the basis for selecting the screening tool.Methods Nutritional risk screening was performed on 123 children hospitalized in the general surgical department by using the Assessment of Malnutrition in Paediatrics (STAMP) and Paediatric Yorkhill Malnutrition Score (PYMS).The malnutrition prevalence based on the World Health Organization (WHO) child growth standards(WHO standards) was evaluated.The data of hospital stay and total hospital expenses were recorded and analyzed.Results By using the STAMP and PYMS,76.4% (94/123 cases) and 85.4% (105/123 cases) of the patients had low-moderate nutritional risk,23.6% (29/123 cases) of the patients and 14.6% (18/123 cases) had high nutritional risk,respectively.The malnutrition prevalence was 17.9% (22/123 cases) based on WHO standards.Children with high nutritional risk screened by STAMP,PYMS and malnutrition evaluated by WHO standards had significantly longer hospital stay(days) and higher total hospital expenses(yuan) compared with those in low-moderate nutritional risk or non-malnutrition patients,and the differences were statictically significant (hospital stay:STAMP:M =8.50 > 7.00,Z =-2.608,P =0.008;PYMS:M =8.00 >6.25,Z =-2.761,P =0.006;WHO standards:M =8.25 >6.75,Z =-2.812,P =0.005.Total hospital expenses:STAMP:M =4 970.46 > 2 939.07,Z =-2.913,P =0.004;PYMS:M =5 486.57 > 3 294.96,Z =-3.024,P =0.002;WHO standards:M =5 515.87 > 3 170.04,Z =-3.098,P =0.002).According to WHO standards,the sensitivity,specificity,Youden index,positive predictive value and negative predictive value of STAMP and PYMS were 68.2% vs.77.3%,86.1% vs.99.0%,54.3% vs.76.3%,51.7% vs.94.4%,92.6% vs.95.2% respectively.Conclusion Both STAMP and PYMS can be used for nutritional risk screening in hospitalized children in general surgery department,while sensitivity and specificity of PYMS are better than those of STAMP.
8.A new alkaloid from Salsola collina

Salsola collina is widely distributed in droughty and semi-droughty area, which is used as a kind of folk remedy in traditional Chinese medicine for treatment of hypertension. The study is on the chemical constituents of this herb from its aerial parts to obtain its active constituents. Dried and crushed aerial parts of this herb were extracted three times with 95% EtOH at reflux. The ethanol extracts were combined and concentrated under reduced pressure at 70 ℃ to yield residue, which was suspended in water and successively partitioned with light petroleum, chloroform and n-butanol. The chloroform and n-butanol fractions were treated by various chromatographic techniques, such as silica gel, C18 reversed-phase silica gel and macroporous resin column chromatography. Compounds were elucidated by their physicochemical properties and spectroscopic analysis. In the course of our study on searching biological active components from this herb, a new alkaloid together with three known alkaloids were isolated and identified as N-transferuloyl-3-methyldopamine (1), 3-[4-(β-D-glucopyranosyloxy)-3- methoxyphenyl]-N-[2-(4-hydroxyl-3-methoxyphenyl)ethyl]-2-propenamide (2), salsoline A (3), salsoline B (4). Compound 4 is a new compound and named as salsoline B, while compound 2 was obtained in Salsola collina for the first time.

10.Efficient transient expression to analyze miRNA targets in rice protoplasts.

Compared with the transgenic approach, transient assays provide a convenient alternative to analyze gene expression. To analyze the relationship between miRNAs and their target genes, a rice protoplast system to detect target gene activity was established. The MIRNA and GFP-fused target sequence (or GFP-fused mutated sequence as a non-target control) were constructed into the same plasmid, and then delivered into rice protoplasts. The GFP expression level decreased significantly when the protoplasts were transfected with the plasmid containing GFP-fused target compared to that of the plasmid with non-target sequence either by fluorescence microscopy or qRT-PCR method. Two microRNA genes, osaMIR156 and osaMIR397, and their target sequences were used to prove the feasibility of the rice protoplast transient assay system. This method will facilitate large-scale screening of rice miRNA target in vivo, and may be suitable for functional analysis of miRNAs of other monocot plants that might share the evolutionarily conserved small RNA processing system with rice.
