Objective To investigate the effects of up?regulated miR?206/miR?1 on the proliferation of breast cancer stem cells and the effect mech?anism. Methods Breast cancer stem cells(BCSCs)were isolated from breast cancer cell line MCF?7 by fluorescence?activated cell sorting. Cells in the experiment were divided into the blank control group, the negative control group, the miR?206 group and the miR?1 group. The BCSCs were transfected by negative control mimic,hsa?miR?206mimic and hsa?miR?1mimic in all groups except the blank control group. MiR?206and miR?1 expression levels as well as the transcription factor EVI?1 gene were detected by real time PCR. The expression levels of the transcription factor EVI?1 protein were detected by Western blot. MTT method was used to detect the effects of miR?206 and miR?1 on the proliferation of BCSCs. Results The BCSCs(CD44+/CD24-/low cells)isolated from MCF?7 cell lines were successfully cultured in serum?free medium for subsequent studies. After transfection of hsa?miR?206mimic and hsa?miR?1mimic for 48 hours,miR?206and miR?1relative expression levels increased. EVI?1mRNA ex?pression levels significantly decreased. The results of Western blot and MTT showed that up?regulated expression levels of miR?206 and miR?1 could significantly reduce the expression of EVI?1 protein and inhibited the proliferation of BCSCs. The differences in levels of miR?206,miR?1 and EVI?1 protein were statistically significant(P<0.05). Conclusion Up?regulated miR?206 and miR?1 expression can inhibit the proliferation ability of BCSCs,which may be related to the down?regulation of EVI?1.

Objective To observe the effect of acupuncture on the expression of nitricoxide synthase (NOS) in gastrointestinal tissues after operation for incomplete intestinal obstruction in rats, and to explore the mechanism of acupuncture in promoting the recovery of gastrointestinal motility. Methods Forty Sprague-Dawley (SD) rats were randomized into treatment group 1, treatment group 2, model group 1, model group 2, and a normal control group, 8 rats in each group. The incomplete intestinal obstruction models were developed by using single ligation method in treatment groups and model groups. Afterwards, the treatment groups were intervened by acupuncture at Zusanli (ST 36) and Yanglingquan (GB 34), and rats in treatment group 1 and 2 were respectively dissected after 1-day and 2-day treatment. The rats in model group 1 and 2 were respectively dissected 1 d and 2 d after modeling. The morphological observation was performed on rat’s intestinal tissues, and the average diameter of the upper part of the obstructed intestine and the expression of NOS in gastrointestinal tissues were compared. Results The average ileocecal diameter after modeling was significantly different from that in the normal control group (P<0.05). There was a significant difference in comparing the ileocecal diameter between model group 1 and 2 (P<0.05). The ileocecal diameter in treatment group 1 was significantly different from that in treatment group 2 and model groups (P<0.05). The ileocecal diameter in treatment group 2 was insignificantly different from that in the normal control group (P>0.05). The NOS content in model groups was significantly different from that in the normal group (P<0.05). The NOS content in treatment group 1 was markedly different from that in model groups and treatment group 2 (P<0.05) after intervention. There were no significant inter-group differences in comparing NOS content (P>0.05). Conclusions Acupuncture can promote the recovery of gastrointestinal motility by regulating intestinal NOS content in rats after operation for incomplete intestinal obstruction.

Objective To investigate the incidence of osteoporosis (OP) in patients with ankylosing spondylitis and the relationship between OP and the clinical data. Methods Serum levels of bone alkaline phosphatase (BALP) and tartrate resistant acid phosphatase 5b (TRACP5b) were detected by enzyme linked immunosorbent assay ( ELISA ) in 60 cases with ankylosing spondylitis, and it was compared with normal controls. Bone mineral density (BMD) was measured through dual-energy X-ray absorptiometry ( DXA), including lumbar ( L2 - L4), bilateral femoral neck and greater trochanter. Some clinical data was collected and analyzed at the same time. Results The incidence of OP in AS patients was 35%, and the incidence of OP in the femoral proximal end was higher than that in lumbar. Compared with normal controls[ ( 1.06 ±0. 18 )U/L ], the levels of serum TRACP5b in AS[ (1.31 ± 0. 82 )U/L] patients was significantly higher ( P <0. 05 ). The levels of serum BLAP in OP combined AS group[ ( 21.65 ± 5.41 ) U/L]were significantly lower than non-OP group[ (32. 37 ± 16. 5 ) U/L] ( P <0. 05 ). The disease duration was negatively correlated with the BMD of femoral neck ( P < 0. 01 ). Conclusions There was higher incidence of OP in AS patients, which were related with the abnormality of bone metabolism and the disease duration.Multiple factors participated in the regulation of bone metabolism of AS.
