1.The significance of detection of Twist,p53 and E-cadherin in breast cancer

Objective To observe the expression and correlation of Twist,p53 and E-cadherin mRNA in breast cancer.Methods Thirty breast cancer tissue and adjacent noncancerous tissue in patients underwent modified radical surgery,and the normal breast tissue in 10 patients with benign breast lesions,respectively,were detected the expression of Twist,p53 and E-cadherin mRNA by RT-PCR and the relationship with clinicl pathological factors and their interrelationships.Results There was significant correlation between the expression of Twist mRNA and the degree of differentiation[high and middle differentiation 93.33%(14/15),1ow differentiation 53.33%(8/15)],lymphatic invasion[yes 93.75%(15/16),no 50.00%(7/14)]and TNM staging[Ⅰ-Ⅱ?stage 46.15%(6/13),III-Ⅳ?stage 94.12%(16/17)]in breast cancer tissue(P<0.05),while there was no obvious correlation between the expression of Twist mRNA and the age,tumor diameter,adventitial infiltration,estrogen receptor,progesterone receptor(P>0.05).The expression of Twist and p53 mRNA in breast cancer tissue were significantly higher than those in adjacent noncancerous tissue and normal breast tissue[(112.36±12.53)% vs.(101.37±9.12)% and(98.03±9.04)%,(101.73±13.32)% vs.(91.43±7.43)% and(90.83±6.58)%,P<0.01],while the expression of E-cadherin mRNA in breast cancer tissue was significantly lower than that in adjacent noncancerous tissue and normal breast tissue[(55.43±12.49)% vs.(69.39±5.63)% and(72.73±7.93)%,P<0.01],there was no significant difference between adjacent noncancerous tissue and normal breast tissue(P>0.05).The expression of Twist mRNA in breast cancer tissue was positively correlated to the expression of p53 mRNA(r=0.526,P=0.004),and negatively correlated to the expression of E-cadherin mRNA(r=-0.500,P=0.000).Conclusions Twist and p53 in breast cancer tissue show higher expression,while E-cadherin shows lower expression.Twist can promote invasion,metastasis of breast cancer by up expression of p53 and down expression of E-cadherin,and it plays an important role.
2.Clinical Comparison of Incidence of Diarrhea between Cerebral Hemorrhage and Cerebral Infarction

Objective To analyze and compare the incidence of diarrhea between cerebral hemorrhage and infarction. Method We observed and compared the time when diarrhea occurred in both 147 cases of cerebral hemorrhage patients and 142 cases of cerebral infarction patients by using statistical methods. Result The incidence of diarrhea in former group was 36.05%, and that of latter was 16.91%. Conclusions The incidence in patients with diarrhea of cerebral hemorrhage was markedly higher than that of patients with cerebral infarction. This may attribute to the higher intracranial pressure in cerebral hemorrhage patients than that of cerebral infarction ones. The complications of reduction of immune function,intestinal infection and hypoproteinemia may be the other factors resulting in the higher incidence of diarrhea in cerebral hemorrhage patients than that of cerebral infarction ones.
3.Color stability of carious incipient lesions in the enamel treated with resin infiltration

BACKGROUND:The resin infiltration technique for masking white spot lesions has obtained a result superior to the conventional remineralization. Up to now, few studies have evaluated the color of carious incipient lesions treated with the resin infiltration.
OBJECTIVE:To assess the color change of caries incipient lesions in the enamel treated with resin infiltration.
METHODS: Forty-eight extracted teeth were exposed to demineralizing solution for 4 weeks to establish artificial caries models. Then artificial caries models were treated with resin infiltration and randomized into four groups: these specimens were immersed in red wine, coffee, tea and artificial saliva for 2 and 4 weeks, respectively. The base line color was evaluated by spectrophotometer. The variation in color (ΔE) was analyzed by one-way analysis of variance and Tukey’s tests.
RESULTS AND CONCLUSION:Al specimens underwent color and lightness changes, irrespective of immersion media. After 2 weeks of immersion, ΔE values were ranked as folows: red wine > coffee > tea > artificial saliva, and there were significant differences between four groups (P < 0.05). After 4 weeks of immersion,ΔE values were ranked as folows: red wine≈ coffee > tea > artificial saliva, and there were significant differences between four groups (P < 0.05) but not between the red wine and coffee groups (P > 0.05). Significant changes in the color were found in the red wine and coffee groups at different time after immersion (P < 0.05). Immersed in red wine, coffee and tea, the?E value was > 3.7, which is not acceptable in clinic. These findings indicate that the color stability of carious incipient lesions located in the enamel treated with resin infiltration is associated with the soak solution and soak time. In addition, immersion in red wine and coffee results in significantly increased color alteration (ΔE) compared with the artificial saliva.
4.Expression of midkine protein in non-small-cell lung cancer and its clinical significance

Purpose:To investigate the features of midkine( MK) protein expression and the correlation between MK expression with angiogenes is and prognosis in non-small-cell lung cancer(NSCLC). Methods:Immunohistochemistry analysis for MK and microvessel de nsity(MVD) were performed on 44 samples of tumor tissue. Results:No positivity expression was detected in either the lun g tissue close to tumor or normal lung tissue,but 26 of 44 NSCLC tissue showed a positive reaction to MK immunostain. Significant correlation between MK express ion with MVD enhancement and lymph metastasis in NSCLC was found,and the surviva l terms in NSCLC with MK expression were significantly shorter than those in NSC LC without MK expression.The enhanced MVD was related to lymph metastasis signif icantly,and the survival terms in NSCLC with enhanced MVD were significantly sho rter than those in NSCLC without enhancement. Conclusions:In human non-small-cell lung cancer, MK overexpre ssion at the protein level may very well be closely related to angiogenesis and metastasis. MK protein may be one of the predictors for prognosis in NSCLC.
5.Research on Life Quality Scale for Patients with Idiopathic Pulmonary Fibrosis.

OBJECTIVETo develop a life quality scale suitable for idiopathic pulmonary fibrosis (IPF) patients, objectively reflecting its changes.
METHODSAuthors first put forward a theoretical structure model of a scale according to patient-reported outcome (PRO) scale formulation principle by combining basic theories of Chinese medicine (CM). Then authors developed an initial scale on the basis of various life quality scales for respiratory disease patients by using structural decision making. Totally 34 patients with confirmed diagnosis of IPF were tested by questionnaire. Items were screened using expert importance scoring method, factor analysis, correlation coefficient method, Cronbach's alpha coefficient method. IPF patient reported outcomes (IPF PRO, IP) were finally defined.
RESULTSA new IP scale was developed covering three areas and 38 items. Pearson correlation coefficient for correlation analysis of clinical symptom scores in ST-George Respiratory Questionnaire and IP scale was 0.828 (P < 0.01). Pearson correlation coefficient for correlation analysis of activity ability scores was 0.929 (P < 0.01). Pearson correlation coefficient for correlation analysis of total scores was 0.862 (P < 0.01). By reliability of IP scale itself (reliability) analysis, Cronbach's alpha coefficient was 0.713. By using factor analysis method for data analysis, KMO statistics was 0.902.
CONCLUSIONIP scale fully reflected the connotation of IPF patients' quality of life, so it could be used as CM clinical therapeutic effect evaluation tool.

6.Advance in studies on long-circulating tumor targeted compound preparations.

Tumor has long been a hard-nut problem in the world medical field. The effect of the conventional drugs is very limited because of the intervention of multiple micro-environmental factors during the occurrence and progression of tumors. With the characteristics of high efficiency, low toxicity and multi-targets synergistic effect, the long-circulating tumor targeted compound preparations show its unique advantages in improving tumor microenvironment and enhancing the therapeutic effect of treatment, thus it has gradually become a hotspot of studies both at home and abroad. Through consulting a great number of professional literatures at home and abroad in recent years, the authors summarized the current studies in vitro and in vive on long-circulating tumor targeted compound preparations in different carriers, in the expectation of providing new ideas and methods for the development of long-circulating tumor targeted compound preparations.

8.The determination of proteasomal activities in circulating white blood cells in patients with advanced gastric carcinoma and their clinical significance

ObjectiveTo investigate the clinical significance of determination of 20S proteasomal activities changes in circulating white blood cells (WBC) in patients with advanced gastric carcinoma.MethodsThe 20S proteasomal activities in circulating WBC separated from 80 patients with advanced gastric carcinoma were determined by 20S proteasome assay kit; the concentrations of interleukin-6(IL-6) in plasma were determined by radioimmunoassay method.The result was compared with that of 11 healthy volunteers.ResultsThe proteasomal activities in circulating WBC[(38.26 ±9.23),(101.85 ±27.63)pmol AMC/ ( s *mg pro) ] and concentrations of IL-6 [ (23.89 ± 6.76),(72.52 ± 20.71 ) ng/L ] of healthy volunteers and the patients with advanced gastric carcinoma had statistical significance (P < 0.01 ).The proteasomal activities and the concentration of IL-6 were correlated with age,pathological type,weight changes,Karnofsky score and disease severe degree(P < 0.01 );while they had no relationship with sex(P > 0.05).Cox's proportional hazard regression model demonstrated that pathological type,Karnofsky score and proteasomal activities were the independent risk factors affecting the survival of patients with advanced gastric carcinoma (P <0.01 or <0.05).20S proteasomal activities and the concentration of IL-6 had positive correlation (r =0.691,P =0.000).ConclusionsThe proteasomal activites in circulating WBC and the concentration of IL-6 in plasma increase and are correlated with age,pathological type,weight changes,Karnofsky score and disease severe degree.Proteasomal activities in circulating WBC can be used as the prognostic reference factor of advanced gastric carcinoma.20S proteasomal activities and the concentration of IL-6 have positive correlation.
