1.E-cadherin promoter methylation and demethylation in epithelial ovarian carcinoma cells

Objective To investigate the cytidylyl phosphate guanosine(CpG) islands methylation status of E-cadherin (E-cad) promoter region in human ovarian carcinoma cell lines (ES-2,3 AO, SKOV3 ), and the effect of 5-azacytidine-2 '-deoxycytidines (5-Aza-CdR ) on the cell proliferative ability, invasion and the expression of E-cad protein. Methods Methylation specific PCR(MSP) was used to detect CpG islands methylation status of E-cad promoter region in ES-2,3AO and SKOV3 cell lines. After treated with different concentrations of 5-Aza-CdR, morphological changes of cell lines were observed under microscope. The proliferative ability was evaluated by methyl thiazolyl tetrazolium(MTT) assay. E-cad protein expression was detected by western-blot and cellular invasion was investigated by 24-well matrigel invasion chambers. Results Hypermethylatian status of CpG islands of E-cad promoter region was observed in ES-2 and SKOV3 cell lines, but not in 3AO cell lines. After treated with 5-Aza-CdR (0.1,1.0,10.0 μmol/L), ES-2 and SKOV3 cell lines displayed morphological evidence of differentiation. 5-Aza-CdR was found to decrease proliferation as evidenced by cell growth curve , to increase the level of E-cad protein expression (P < 0.01 ), and effectively inhibit the ability of cell invasion(P <0.01 ). Conclusions CpG hypermethylation is an important mechanism of E-cad gene inactivation in ES-2 and SKOV3 cell lines. 5-Aza-CdR be found to inhibit proliferation and invasion, and increase the expression of E-cad probably by the inhibition of hypermethylation.
2.Perioperative ALB,PA and hs-CRP changes in surgical patients

objective To evaluate the clinical implications of perioperative serum albumin (ALB),pre-albumin(PA)and high-sensitivity C-reactive protein(hs-CRP)changes in surgical patients.Methods This study included 103 surgical cases undergoing major,moderate or minor surgery respectively.Perioperative serum ALB.PA and hs-CRP levels were measured.Results were compared with 56 normal controls. Results Pre-and postoperative ALB was(43.6±5.1)g/L and(35.8±5.3)g/L in major surgery patients compared with that of(45.0±3.7)g/L and(38.3±5.7)g/L in moderate or minor surgery patients,both were significantly lower than that of(46.7±2.9)g/L in control group(F=54.07,P<0.05);Pre-and postoperative PA level was(246±68)mg/L in major surgery patients while that was (270±53)mg/L and(207±70)mg/L respectively in moderate or minor surgery group,that in both groups were lower than that(279±40)mg/L in control group(F=41.18,P<0.05),Pre-and postoperative hsCRP level was(5.3±5.1)mg/L and(12.7±4.2)mg/L in major surgery patients compared with that of (4.5±3.6)mg/L and(11.1±4.3)mg/L in moderate or minor surgery patients,again both that in the two groups were much higher than that of(1.0±0.7)mg/L in control group(F=69.87,P<0.05).Conclusions Combined detection of serum ALB,PA and hs-CRP levels is a measure of surgical trauma especially for these patients suffering from postoperative infections.
3.Advances in YAP on stemness maintenance of cancer stem cell Properties

Yes-associated protein (YAP) is a multifunctional protein that can interact with different transcription factors to acti-vate gene expression. YAP, as the key transcription protein of Hippo pathway, has made important contribution to the control of the or-gan size, stem cell biology, and tissue-specific stem cell self-renewal. In the Hippo pathway, MST1/2 kinases, together with the adaptor protein SAV, phosphorylate LATS1/2 kinases. YAP is phosphorylated and inhibited by the activated LATS1/2, a key component of the Hippo tumor suppressor pathway. In recent years, numerous studies have shown that the high expression of YAP in the embryonic stem cells, neural stem cells, and hematopoietic stem cells can maintain the cancer stem cell;and YAP has become the marker of cancer stem cells. Meanwhile, more and more studies indicate that the disorder of Hippo-YAP signaling pathway may be associated with cancer stem cells. In this review, we mainly discuss the role of the Hippo pathway in the stem cell biology and its potential implications in tis-sue homeostasis and cancer.
4.The diagnostic Value of Three Methods Among autoimmune diseases

Objective To study more significant detective combination by comparing three methods of antinuclear antibody,extractable nuclear antigen and antinuclear antibodies 3.Methods Three methods were adopted as follows:detecting antinuclear antibody with HEP-2 and prime hepar as antigen media,determining extractable nuclear antigen by EUROASSAY including nRNP/Sm、Sm、SSA、SSB、Scl-70 and Jo-1,using crithidia iuciliae as antigen,to detect ds-DNA antibody with indirect immunofluorescence assay,and to study 15 autoantibodies by EUROLINE.Results To compare positive rates of three dilution(1:10,1:80,1:100)in ANA detection,there existed significant difference(P<0.01)between two dilution of 1:10 and 1:80 only,.In the detection of multiple nuclear types(cytoplasmic pattern,centromere pattern and homegeneous pattern etc),the positive rate of antinucleatr antibodies 3 was higer than that of ENA.IIF was prior to the EUROLINE in the detection of ds-DNA.Conclusion During detecting ANA with IIF,the dilution of 1:80 has advantages in raising positive rate,and ANA3 of 15 antigens provides facilities for clinical diagnosis.
5.EXPERIENCES OF RESCUE OF CHILDREN SUFFERING FROM SEVERE HEMOLYTIC UREMIC SYNDROME

In order to improve the diagnosis and treatment of severe hemolytic uremic syndrome (SHUS),we summarized the rescuing experiences in 3 children with SHUS.Therefore,close observation of gastro intestinal symptoms,collaborated with repeated examinations of the blood,renal function,urinary output,blood pressure,and manifestations of the central nervous system could help predict to certain extent the occurrence of further hemolysis and renal failure in these patients.Energetic treatment of malignant hypertension and relieving the symptoms of center nervous system could successfully improve renal functions.If urinary output decreased quickly,creatinine (Cr) and blood urea nitrogen (BUN) increased,or severe infection occurred in SHUS patients,hemodialysis should be undertaken as early as possible to prevent caute renal failure and multiple organ failure syndrome (MOFS).Therefore,the treatment of SHUS in children has its peculiarity and complexity.
6.Mouse nerve growth factor and sub-hypothermia for neural protection in severe traumatic brain injury and its mechanism

Objective To investigate the mechanism of neural protection of mouse nerve growth factor combined with sub-hypothermia in the treatment of patients with severe traumatic brain injury. Methods 90 cases of severe traumatic brain injury were randomly divided into study group and control group with 45 cases of each group, the control group were given routine treatment; the study group were given on the basis of routine treatment of mouse nerve growth factor combined with sub-hypothermia treatment, with 2 weeks treatment, the clinical indicators and corresponding nerve injury, inflammation, oxidative stress indexes, clinical effect and complications were compared after 2 weeks treatment. Results Compared with before treatment or control group, scores of Glasgow coma scale (GCS) and Glasgow outcome scale (GOS) and montreal cognitive assessment (MoCA) in study group after the treatment increased, National Institute of Health stroke scale (NIHSS) score decreased(P<0.05), neuronspecific enolase (NSE), myelin basic protein (MBP) and S100 beta levels decreased(P<0.05), the serum tumor necrosis factor-α (TNF-α),interleukin-6 (IL-6), IL-10 levels decreased (P<0.05), the malondialdehyde (MDA) decreased, the glutathione peroxidase (GPx) and oxidation resistance (AOA) levels increased (P<0.05). The control group efficiency was 73.33%, the study group efficiency was 91.11%, there was significant difference (P<0.05). All patients were followed up, no case off, there was no significant difference in adverse drug reaction rate between two groups. Conclusion Mouse nerve growth factor and sub-hypothermia has the significant neural protection for patients with severe traumatic brain injury, and its mechanism may be related to reduce nerve injury indicators and improve inflammatory factor and oxidative stress response.
7.Early evaluation of the single-fiber conduction studies of motor nerve damage in carpal tunnel syndrome

Objective To evaluate the function status of motor nerve in patients with carpal tunnel syndrome ( CTS) sensory neuropathy by single-fiber conduction studies ( SF-CS).Methods Forty patients with CTS were divided into two groups according to the nerve conduction abnormality of median nerve .The sensory conduction abnormalities ( SCA) group included 20 patients with abnormal sensory conduction , while the sensory motor conduction abnormalities ( SMCA) group included 20 patients with abnormal motor sensory conduction.The Keypoint.net electromyogram device was used to detect the wrist to the abductor pollicis brevis latency , using the saddle shaped stimulating electrodes for stimulating and the single fiber electromyography electrode for recording.The control group included 20 healthy people.Results The latency of single-fiber conduction of median nerve in SCA group , SMCA group and control group was (3.92 ±0.28) ms, (4.71 ±0.49) ms and (3.41 ±0.31) ms (F=63.829, P=0.000), respectively.The area under the receiver operating characteristic curve was 0.920 for the latency of single-fiber conduction of median nerve , and its specificity was 85%in cut off point.Conclusions There is clinical motor nerve damage in patients with CTS sensory neuropathy.The SF-CS may become an effective means for early evaluation of motor nerve function state.
8.The relationship between circulating tumor cells and clinicopathologic characteristics of primary lung cancer patients

Objective To investigate the relationship between the presence of circulating tumor cells (CTC) and the clinicopatho-logic characteristics of primary lung cancer patients .Methods Samples of peripheral blood(7 .5 mL) from 65 cases of patients with lung cancer(lung cancer group) and 10 cases of patients with lung benign diseases and 10 healthy volunteers (control group) ,and then those CTC were enriched by negative immunomagnetic separation with anti-CD45 antibody and then dectected by immunocyto-chemistry with anti-pan-cytokeratin antibody .The relationship between the positive rate of CTC and clinicopathologic characteristics of lung cancer patients were analyzed .Results The positive rate of CTC in lung cancer patients was 69 .23% (45/65) ,while CTC was not detected in any patients with lung benign disease or healthy volunteers .The positive rate of CTC in patients at TNM stageⅠ - Ⅱ ,Ⅲ and Ⅳ was 23 .08% (3/13) ,50 .00% (6/12) and 90 .00% (36/40) ,respectively .There was significant difference in posi-tive rate of CTC between patients at different TNM stages (P= 0 .000) .The positive rate of CTC in metastatic (stage M1 ) lung cancer patients(83 .33% ,35/42) was significantly higher than that in non-metastatic(stage M0 ) lung cancer patients(43 .48% ,10/23)(P=0 .002) .With the increasing number(0 ,1 ,2 ,3) of metastatic sites ,the positive rate of CTC increased (53 .48% ,83 .33% , 87 .50% ,75 .00% ) ,and there was significant difference in different positive rates of CTC (P=0 .024) .There was statistically sig-nificant difference in the positive rate of CTC between Cyfra 21-1 normal group(53 .33% ,16/30) and high value group(92 .00% ,23/25)(P=0 .021) ,the same with LDH normal group(55 .00% ,22/40) and high value group(82 .86% ,29/35)(P=0 .004) .Conclu-sion CTC were presented in some lung cancer patients .The presence of CTC in peripheral blood is correlated positively with the patients′clinical stage ,M stage ,the number of metastatic sites ,Cyfra21-1 value and LDH value .CTC detection is useful for more accurate risk assessment of patients with lung cancer .
10.Nonpharmacological therapy of gastroesophageal reflux in preterm infants

Gastroesophageal reflux (GER) was very common among preterm infants, which could cause several complications and severely affects the prognosis. The standardization of diagnosis and treatment of gastroesophageal reflux was very important for preterm infants. Due to less adverse reactions, nonpharmacological therapy was the most advisable choice in preterm infants with GER. In this paper, nonpharmacological therapies including body positioning, feeding frequency and modality, changing food patterns, gastric tube, and nonnutritive sucking were reviewed. Body positioning was the safest and most effective nonpharmacological therapy. Other nonpharmacological strategies such as changes of feeding frequency and modality might also be useful. The role of some strategies was still controversial, thereby needing to be further investigated. Selection of what kind of specific nonpharmacological therapy should be based on the specific conditions.
