2.Effect of exogenous atrial natriuretic peptide on airway inflammation in mice with allergic asthma and its mechanism

Objective To investigate the effect of exogenous atrial natriuretic peptide on airway inflammation in allergic asthma mice model and its mechanism.Methods A total of 48 SPF 48 BALBc mice were selected and randomly divided into the normal group (equal quantity normal saline intervention),model group (equal quantity normal saline intervention),experimental group A (exogenous atrial natriuretic peptide 0.5 μg/g) and experimental group B(exogenous atrial natriuretic peptide 0.5 μg/g-+-A71915).The model group,experimental group A and B were given the corresponding treatment measures after successfully constructing the model,at 24 after the last provocation test,serum and bronchoalveolar lavage fluid in each group were collected for detecting inflammation factors.The HE staining was used to observe the pathological changes of lung tissue,the expression level of GATA3 protein in lung tissue was detected by Western blot technology.Results The blood EOS,lymphocyte and neutrophil ratio in the model group were higher than those in the normal group (P<0.05),which in the experimental group A were higher than those in the model group and experimental group B (P<0.05);bronchoalveolar lavage fluid EOS,lymphocytes,neutrophil ratio,IL-6,TNF-α levels in the model group were higher than those in the normal group (P<0.05),alveolar lavage fluid EOS,lymphocyte,neutrophil count,IL-6 and TNF-α levels in the experimental group A were higher than those in the model group and experimental group B (P<0.05);the expression level of GATA3 protein in lung tissue of the model group was higher than that in the normal group(P<0.05),the expression level of lung tissue GATA3 protein in the experimental group A was higher than that in the model group and experimental group B (P<0.05).Conclusion Exogenous atrial natriuretic peptide can further aggravate airway inflammation reaction in allergic asthma mice model,which is related to the activation of inflammatory cells,the release of inflammatory cytokines and the increase of GATA3 protein expression in lung tissue.
3.Expression,purification and biological activity assays of global human C1 q and tumor necrosis factor related protein-2

Aim To prepare soluble global human C1 q and tumor necrosis factor related protein-2 in Escherichia coli. Methods Recombinant expression plasmid was transformed into strain BL21-codonplus (DE3),and the recombinant protein of Trx-gH2 was expressed by IPTG induction and then purified by Ni-NTA affinity and gel filtration chromatography.Results The purified recombinant Trx-gH2 was shown to be active under in vi-vo and in vitro assay conditions.Conclusion Active recombi-nant global hCTRP2 is efficiently prepared from Escherichia coli protein expression system.
4.Electrophysiology characteristic of critical illness polyneuromyopathy and its effect on the duration of mechanical ventilation

Objective To analyze the electrophysiologie characteristics of critical illness of polyneuromyopathy ( CIPM ) and its effect on the duration of usage of mechanical ventilation. Methods Denmark Keypoint 4-electromyogram was utilized to record electraneurophysiology on 17 subjects with lung infection and respiratory failure who were treated with mechanical ventilation over 1 week and had difficulty in weaning from the ventilator in intensive care unit. Electrophysiology character of CIPM was described and the duration of mechanical ventilation of CIPM patients was compared with non-CIPM subjects. ResultsOf the 17 cases, 9 subjects were diagnosed with CIPM including 6 patients with critical illness polyneuropathy (CIP), and 3 patients with critical illness myopathy (CIM), the 2 subtype of CIPM. Compound muscle action potential (CMAP) and sympathetic skin response (SSR) in all of CIPM patients were abnormaL Though the amplitude of sensory nerve action potential (SNAP) in patients with CIP was reduced, nerve conduction velocity (NCV) was normal in all of them. Repetitive nerve stimulation, somatosensory evoked potential and blink reflex were normal in all of the patients with CIPM, however, the rate of occurrence of F wave was reduced in 2 patients with CIP and in 2 patients with CIM. The median and quartile range of duration of mechanical ventilation in CIPM patients was 30. 0 days (20. 0--45.0 days), significantly longer than13.5 days (9.5--17.5 days) in non-CIPM patients (U=7.500,P=0.006). ConclusionsThe ehctrophysiology change in patients with CIPM is characterized by reduced amplitude of CMAP and (or) SNAP, normal NCV and abnormal SSR. The duration of mechanical ventilation is significantly prolonged in CIPM patients.
5.Effects of the contact heat evoked potential in evaluating the small fiber impairment of diabetic neuropathy

Objective To investigate a non-invasive and objective measure for patients with diabetic small fiber neuropathy by contact heat evoked potential (CHEP). Methods CHEP was performed in diabetic patients (n=46) and normal subjects (n =40). Thermal stimuli were given at 52 ℃ to 3 body sites: right forearm, right dersum hand and right peroneal area. CHEP were recorded from Cz. The main components of CHEP were observed. Studies on sensory nerve conduction were performed in all the diabetic patients and the normal subjects. Results In 7 patients there were no recordable CHEP on stimulation of the forearm, and in some cases of dorsum hand (n =9) and the leg (n = 16). In contrast, CHEP were recordable in all control subjects. The diabetic group had reduced N-P amplitudes and prolonged N-wave latencies. In patients with normal sensory nerve conductions, amplitudes of CHEP were reduced and latencies of CHEP were prolonged. Twenty-five patients had normal sensory nerve conduction for upper limbs, the amplitudes of CHEP being (34.0±12.6) μV vs (48.4±17.5)μN, Z= -3. 151, P<0.01 for the forearm, (28.2±10. 1) μV vs (43.0±16.6) μV, Z= -3.712, P<0.01 for the dersum hand with a latency of (420.4±27.8) ms vs (407.2±24.6) ms, t =2.015,P=0.048. In 18 patients with normal sensory nerve conductions for lower limbs, the latency of CHEP for the leg was (473.5±46. 6) ms vs (448.6±35.0) ms, t =2. 219 (P =0. 031) and amplitudes (23. 8±7.4) μV vs (41.5 ±18.5) μV, Z = - 3. 855 (P <0.01). Conclusion CHEP provides an objective and non-invasive potential clinical utility in the evaluation of diabetic small fiber neuropathy.
6.Single nucleotide polymorphisms in cell cycle regulator p21 and p27 genes are associated with susceptibility to epithelial ovarian cancer

Objective To investigate the association of single nucleotide polymorphisms(SNP)in p21and p27 genes with the risk of epithelial ovarian cancer(EOC).Methods Genotypes were analyzed by polymerase chain reaction-restrictive fragment length polymorphism(PCR-RFLP)method in 234 patients with EOC and 284 control women in China.Results (1)The frequencies of the p21 in healthy controls were 34.2%.49.6%and 16.2%,while the distribution of the C and T allele was 59.0%and 41.0%,respectively.The p21 C/C(28.2%),C/T(53.0%),T/T(18.8%)distribution in ovarian cancer patients was not significantly different from that in healthy controls(P>0.05).There was no statistic difference in allele distribution between ovarian cancer patients and healthy controls(P>0.05)either.The stratification analysis by tumor histological type did show that the genotype distribution in four types of ovarian cancer patients was significantly different from that in healthy controls(P=0.02).The C/C genotype was likely to reduce the risk of epithelial endometrial cancer.and the adjusted odds ratio was 0.56(95%CI:0.32-0.98).(2)The genotype frequencies of the p27 in healthy controls were 88.4%,10.9%and 0.7%.while the distribution of the V and G allele was 93.8%and 6.2%.respectively.The V/V(93.6%),V/G(5.1%)and G/G(1.3%)distribution in ovarian cancer patients was significantly different from that in healthy controls(P=0.04).There was no statistic difference in allele distributionbetween ovarian cancer patients and healthy controls(P>0.05).Compared with the V/G and G/G genotypes,the V/V genotype increased the risk of EOC,the adjusted odds ratio was 1.92(95%CI:1.02-3.63).Conclusion The C/C genotype of p21 may reduce the risk of epithelial endometrial cancer,and the genotype of p27 V/V may be a potential risk factor for susceptibility to EOC.
7.Effects of Fufang Xiaojingtong Capsule on dysmenorrhea induced by oxytocin in rats

AIM: To investigate the analgesic effect and possible mechanism of Fufang Xiaojingtong Capsule(Radix astragali;Radix paeoniae alba,Radix et Rhizoma ginseng,Radix rehmanniae praeparata;Rhizoma chuanxiong)(FFXJTC) on oxytocin-induced dysmenorrhea in rats. METHODS: Sixty rats were randomly divided into normal control group(control),dysmenorrhea model group(model),high dose(2.0 g/kg),middle dose(1.0 g/kg) and low dose(0.5 g/kg)groups of treatment by FFXJTC,and Tianqi Tongjing Capsule(TQTJC) group(2.0 g/kg).Diethylstilbestrol was continuously subcutaneous injection for 10 d once a day in rats of all groups except rats of control group.FFXJTC was administrated ig for 7 d in rats of all groups except for model group given distillated water.After administration of oxytocin(0.2U) in every rat,the frequency of stretching writhing was recorded within 30 min.The blood was drawn 1 h after administration of oxytocin.The contents of nitric oxide(NO)and malondiadehyde(MDA) and activities of SOD and GSH-PX were respectively determined in serum. The contents of 6-Keto PGF_(1?) and TXB_2 were respectively determined in plasma.Uterus was dissected and homogenated for endothelin(ET-1) measurement. RESULTS: Compared with that of model group,frequency of stretching writhing decreased significantly,the activities of SOD and GSH-PX were markedly enhanced,NO content increased and MDA content decreased in serum,and 6-Keto PGF_(1?) content increased and TXB_2 content decreased in plasma in FFXJTC group. CONCLUSION: FFXJTC has an analgesic effect on dysmenorrhea induced by oxytocin in rats,its mechanism may be related to anti-oxidization,decreasing the contents of TXA_2 and ET-1,increasing the contents of NO and PGI_2.
8.Study on preparation process and formulation optimization of herpetin liposomes.

Herpetin (HPT) is an active monomer constituent isolated from lignanoid in seeds of Herpetospermum caudigerum. HPT shows inhibitory effects in hepatic injury and HBV-DNA and the replication. In the study, we successfully prepare herpetin liposomes by film dispersion method for the first time. The prescription process was optimized, with the entrapment efficiency as the index. According to the optimized prescription, the mass ratio of HPT: phospholipids: cholesterol was 2.44:78.05: 19.51, the hydration and de-molding process was performed with 0.5% F68 solution at 50 degrees C, and the water-bath ultrasonic time was 20 min. The HPT liposomes prepared by this method showed an average entrapment efficiency of (94.50 +/- 2.15)% and a particle size of (119.2 +/- 10.7) nm, which was consistent with the trial expectations and will lay a solid foundation for the hepatic targeting delivery system in future.

9.Recent progress of potential effects and mechanisms of chlorogenic acid and its intestinal metabolites on central nervous system diseases.

Chlorogenic acid displays several important roles in the therapeutic properties of many herbs, such as antioxidant activity, antibacterial, antiviral, scavenging free radicals and exciting central nervous system. Only about one-third of chlorogenic acid was absorbed in its prototype, therefore, its gut metabolites play a more important role in the therapeutic properties of chlorogenic acid. It is necessary to consider not only the bioactivities of chlorogenic acid but also its gut metabolites. This review focuses on the potential activities and mechanisms of chlorogenic acid and its gut metabolites on central nervous system diseases.

10.Effect of the application on improving postoperative upper extremity function rehabilitation in patients with breast cancer

Objective To promote upper extremity function rehabilitation in patients with breast cancer postoperatively.Methods The application of upper extremity function rehabilitation was designed,and was applied in patients with breast cancer after surgery who were selected from June 2015 to September,and we established management and evaluation scheme.Result The application improved patients' functional exercise compliance and upper extremity dysfunction(P<0.001).In the short term,upper extremities edema incidence was 2.17%,which was lower than the national average.The nurses thought that it improved the work efficiency.Conclusion The application is an effective tool to improve postoperative upper extremity function rehabilitation in patients with breast cancer.
