International Eye Science 2015;(1):34-37
Abstract? AlM: To investigate the inhibition effect of bevacizumab on human Tenon capsule fibroblasts ( HTFs ) and discuss the countermeasures of bleb scarringscarring in glaucoma surgery countermeasures.? METHODS: Adopted cell recovery method and followed the aseptic principles, we performed the culture of HTFs which came from the Central Laboratory of Shaanxi People's Hospital cell library. Wound Healing assay:We scraped a cell-free zone on the cell surface when the cells reached confluence at 80%. The control group was added to serum-free DMEM medium. The HTFs of bevacizumab group were stimulated with 1mg/mL concentrations without DEME for 0, 24, 48, and 72h. The scratch width was observed and measured.? RESULTS: HTFs were long fusiform shape under microscope, the nucleus is in the center of the cell with larger nucleus, abundant cytoplasm, were arranged in a whorled growth out of shape, strong ability to proliferate, conform to general forms of fibroblast. The morphological and biological characteristics of cells after cryopreservation and resuscitation remain unchanged. Wound healing assay: 0h, equal to the initial width of the two groups, 24h when the migration distance of the two groups of cells are basically the same, 48h when the control group cell migration distance is greater than that in the bevacizumab processing group, 72h when the control group scratches basichealing, bevacizumab treated cells migrate closer than 48h no significant change, and a lot of cells died.?CONCLUSlON:Cell recovery method can successfully cultured HTFs, which was stability on morphology and biological properties, laying the cellular basis for experimental research. Fibroblast itself has a strong ability to migrate, outside - derived bevacizumab can inhibit HTFs migration evidently and it will cause excessive cell death when. Bevacizumab has certain extent inhibitory effect on HTFs migration, and it is likely to become one of the important drugs for creating bleb scarring after glaucoma surgery in the future.

A study on the optimal dilution of serum to detect anti HBc-IgM with ELISA. was carried out. Samples were diluted to 10-1to 10-7. Among 26 samples of normal individuals, 22 were negative, but two samples of 10-1 dilution, one sample of 10-2, and one sample of 10-1-10-3 were positive though the control wells were all negative. 42 samples of the patients with variours types of hepatitis B were examined. It was found that when serum dilution was 10-1, only 6 cases were positive; when the dilution was 10-2, 20 cases were positive; when the dilutions were 10-3, 5?10-3, and 10-4, high positive rates were obtained and they were 88.1%, 78.5%, and 69% respectively; and when the samples were further diluted, the positive rate decreased. In order to get goodfresults and minimize false results, either false negative or false positive, it is suggested that 5?10-3 be the optimal dilution.

Objective To study the effects of intraperitoneal adherence of pus mosses on the efficacy of open repair(OR)and laparoscopic repair(LR)for perforated peptic ulcers(PPUs). Methods Intraperitonealad-herence of pus mosses was divided into the first, second and third stage, respectively. Surgical duration, volume of peritoneal drainage ,rate of abnormal wound healing ,number of intensive care unit transfer ,recovery time of bowel function ,ambulation time ,length of hospital stay ,and postoperative treatment abandonment rate were compared. Results There were 78 ,46 ,and 29 patients for the three stages in OR group ,and 71 ,39 ,and 15 patients in LR group, respectively. There were significant differences insurgical duration, volume of peritoneal drainage, rate of abnormalwound healing, number of ICU transfer, recovery time of bowel function, ambulation time, length of hospital stay about the first stage between the two surgical methods(P values were 0.000,0.000, 0.015,0.000,0.000,0.000,and 0.043 respectively)except for treatment abandonment rate(P value was 0.209). There were significant differences in volume of peritoneal drainage ,rate of abnormal wound healing ,number of ICU transfer, recovery time of bowel function, ambulation time, and length of hospital stay(P values were 0.000, 0.033,0.000,0.000,0.001,and 0.028 respectively)for the second stage except surgical duration and treatment abandonment rate(P values were 0.196 and 0.358 respectively) between the two procedures. The differences were significant in surgical duration ,volume of peritoneal drainage ,and length of hospital stay between the two methods (P values were 0.039 ,0.003 ,0.024 ,and 0.002 respectively) for the third stage but there were no significant differencesin rate of abnormal wound healing , number of ICU transfer , recovery time of bowel function ,ambulation time ,and treatment abandonment rate (P values were 0.175 ,0.173 ,0.766 ,and 0.757 respectively). Conclusions LR is superior to OR under the first stage of pus moss and still has advantages under the second stage. However,LR may be inferior to OR under the third stage.

Objective To discuss the application value of 18F-FDG PET/CT in diagnosis of colorectal cancer. Methods We retrospectively analyzed the data of 186 patients who were diagnosed as colorectal cancer by 18F-FDG PET/CT. We classifed all cases according PET/CT imaging into position group and type group, and calculated SUV (maximum standard uptake value) of tumor tissue, and compared with normal tissue. Results There were 112 cases of rectal cancer,27 cases of carcinoma of sigmoid, 17 cases of carcinoma of cecum,16 cases of ascending colon carcinoma,14 cases of carcinoma of descending colon according to positions via PET/CT. There were 68 cases of protrude type,57 cases of ulcerative type,34 cases of infiltrating type,27 cases of colloid adenoma according types via PET/CT. The SUV of tumor tissue was higher than normal value, the difference had statistical significance. Conclusion 18F-FDG PET/CT has high application value in colorectal cancer diagnosis, positions and types.

BACKGROUND: The soft tissues would shrink with nasal framework collapse following surgical trauma, which cause aseptic inflammation, lead to parts of cartilage resorption. Accordingly, long-term saddle nose deformity usually accompanied by short nasal columella. Complications such as skin perforation or ulceration would appear if corrected the deformity using medical silicone rubber with large tension.OBJECTIVE: To explore the effectiveness of repairing post-traumatic saddle nose deformity with autologous costal cartilage transplantation.METHODS: A total of 21 cases with post-traumatic saddle nose deformity accompanied by short nasal columella were selected, including 6 males and 15 females, aged 16 45 years. All of the cases had trauma history and agreed with the treatment. The costal cartilage was obtained from the seventh rib and formed to babylon weeping willow leaf shape and columella nasi stent to repair post-traumatic saddle nose deformity. The "V-Y" progradation suture was used in the philtrum introcession and the botton of nasal columella to extend the nasal columella. The recovery of saddle nose deformity after transplantation, discharge of transplanted cartilage, as well as the incision scar status was observed.RESULTS AND CONCLUSION: The results were satisfactory and there were no complications after transplantation. All the cases were followed up from 6 months to 2 years. No case suffered costal cartilage grafts discharge or chondral deformation. The scar was little at the bottom of nasal columella. It is an ideal method for repairing post-traumatic saddle nose deformity using transplantation of autologous costal cartilage with "V-Y" progradation suture.

Objective To introduce a two-stage operative method to treat ma lu nion after the distal femoral fracture. Methods From Aug. 1992 to Aug. 2000, 12 patients underwent osteotomy and secondary fixation due to malunion after the di stal femoral fracture. There were 10 closed fractures and 2 open ones. The prima ry treatment was conservative for 2 patients, and operative for 10. We first per formed the osteotomy along the primary fracture line where the deformity was the most severe. When the roentgenogram confirmed that the deformity had been corre cted satisfactorily, a secondary operation of ORIF (open reduction with internal fixation) was performed. Results On average, all the 12 patients gained bone un ion 5.1 months postoperatively. The mean follow-up duration was 7.2 years (rang e: 3.5 to 11.5 years). None of the patients was afflicted by infection or loosen ing of the internal fixator. According to the Schatzker scoring scale system, al l the patients were evaluated as poor preoperatively, but as excellent in 7, goo d in 4 and fair in 1 at the last follow-up. Conclusion Osteotomy and secondary fixation is not only simple and accessible but also can lead to confirmed clinic al results. However, it is still very important to reduce and fix the acute frac tures stably to avoid malunion.
