1.Effect of glucose regulated protein 78 on autophagy and apoptosis in ovarian epithelial carcinoma

Objective To explore the effect and mechanism of glucose regulated protein 78 (GRP78) on autophagy and apoptosis in ovarian carcinoma, and to investigate the influence on the growth and sensitivity to cisplatin on the ovarian cancer cells.Methods The human ovarian cancer cell line SKOV3 were treated by the GRP78 regulator BAPTA-AM and A23187, which were used to decrease or increase the expression levels of GRP78, respectively.The experiment were divided into three groups.Cells in the group of BAPTA-AM were treated by BAPTA-AM at the final concerntration of 40 μ mol/L for 1 hour.Cells in the group of A23187 were treated by A23187 at the final concerntration of 4 μmol/L for 24 hours.While, cells in the control group were treated by culture medium without any GRP78 regulator for 24 hours.The expressions of GRP78, beclin1, Bcl-2 and CHOP mRNA and protein were detected by reverse transcription (RT)-PCR and western blot.The autophagy levels was observed by green fluorescent protein-microtubule-associated protein 1 light chain 3-Ⅱ (GFP-LC3-Ⅱ) fluorescence staining.The flow cytometry was used to analyse the apoptosis rates of cells.The effect on cell growth and the sensitivity to cisplatin of SKOV3 were accessed by methyl thiazolyl tetrazolium (MTT).Results (1)The mRNA expressions of GRP78, beclin1, Bcl-2 and CHOP in the group of BAPTA-AM were 0.583±0.025, 0.860± 0.055, 0.714±0.032 and 0.811±0.004, respectively.The mRNA expressions of GRP78, beclin1, Bcl-2 and CHOP in the group of A23187 were 0.840± 0.044, 0.654 ± 0.065, 0.908 ± 0.047 and 0.620 ± 0.062, respectively.The mRNA expressions of GRP78, beclin1, Bcl-2 and CHOP in the control group were 0.687± 0.032, 0.772 ±0.029, 0.845 ±0.018, 0.712 ± 0.077, respectively.While the protein expressions of GRP78, beclin 1, Bcl-2 and CHOP in the group of BAPTA-AM were 0.423±0.035, 0.952±0.022, 0.385±0.032, 0.681± 0.095, respectively.The protein expressions of GRP78, beclin1, Bcl-2 and CHOP in the group of A23187 were 0.743 ±0.032, 0.638±0.025, 0.596±0.029, 0.431 ±0.095, respectively.The protein expressions of GRP78, beclin1, Bcl-2 and CHOP in the control group were 0.617±0.031, 0.789±0.083, 0.492±0.036, 0.531 ± 0.003, respectively.The mRNA and protein expressions of beclin and CHOP in the group of BAPTA-AM were both higher than those in the control group (P<0.05).While, the mRNA and protein expressions of beclin and CHOP in the group of A23187 were both lower than those in the control group (P< 0.05).(2) The autophagy fluorescence of SKOV3 in the group of BAPTA-AM, A23187 and the control group were 706±117, 473±128, 595± 126, respectively, in which there were significant differences among three groups (P<0.05).(3) The apoptosis rate of SKOV3 in the group of BAPTA-AM was (27.4±2.2)%, which was higher than that in the control group [(19.6± 1.4)%, P<0.05].The apoptosis rate of SKOV3 in the group of A23187 was (12.2± 1.9)%, which was lower than that in the control group (P<0.05).(4) The comparison of the sensitivity to cisplatin in 3 groups of SKOV3.The 50% inhibition concentration (IC50) of SKOV3 to cisplatin was (3.02±0.62) mg/L.After treated by BAPTA-AM + cisplatin, the IC50 was (2.00±0.17) mg/L and the sensitivity of SKOV3 to cisplatin was increased by 33.8%, and there was significant difference (P<0.05), compared with the control group.And after treated by A23187 + cisplatin, the IC50 was (4.91±2.52) mg/L and the sensitivity of SKOV3 to cisplatin was decreasd by 62.6%;and there was significant difference (P<0.05), compared with the control group.Conclusion GRP78 could regulate autophagy and apoptosis of ovarian cancer cells by regulating the expressions of beclin1, Bcl-2 and CHOP, thereby affecting the sensitivity to cisplatin in ovarian carcinoma, which may be a new method for the treatment and improvement of the sensitivity to cisplatin in ovarian carcinoma.

3.Nursing care of patients with nasal packing silicone ventilation tube after functional endoscopic sinus surgery

This paper introduces the nursing care of 1556 patients with nasal packing silicone ventilation tube after func-tional endoscopic sinus surgery. Meticulous nursing care during nasal packing silicone ventilation tube could alleviate patients' discomfort obviously. It was emphasized to suck at exact time according to the patients' demands, adopt effective suction means to alleviate the damage of nasal mucosa,such as suction in the tube or in the bottom of the tube, take measures to promote the cleaning of secretion,which could improve the breath and sleep,as well as shorten the time of recovery.
4.3.0 T MRI findings of femoroacetabular impingement

Objective The purpose of our study is to evaluate high-resolution 3.0 T MRI in the identification of finding in patients with a clinical diagnosis of femoroacetabular impingement (FAI). Methods From June 2008 to May 2010, 54 consecutive patients with clinically diagnostic FAI received an high-resolution 3.0 T MR scan (Siemens AG, Germany), including 38 males and 16 females with an average age of 33.5 years (range, 18-50). All cases were assigned into 3 groups according to the morphology changes of the hip joint: Group Cum, Group Pincer, and Group Normal. The frequency of abnormal signs in 3 groups was calculated. Nonparametric tests by SPSS were used in data analysis. Results Forty-four patients (81.5%, 44/54) showed acetabular labrum disorders, 35 patients (64.8%, 35/54) showed femoral-acetabular cartilage lesions on MRI, 19 patients (35.2%, 19/54) showed arthroedema, 14 patients (25.9%, 14/54) showed femoralacetabular bone disorders, 4 patients (7.4%, 4/54) showed articular capsule and ligaments disorders. There was no statistically significant in the frequency of abnormal sign in MR1 between male and female. The frequency of acetabular labrum disorders, femoral-acetabular cartilage lesions and femoral-acetabular bone lesions in Group Cum was more than that of Group Pincer and Group Normal. The frequency of acetabular labrum disorders was the most disorders (Group Cum 88.5%, Group Pincer 77.8%, Group Normal 70.0%),next high-frequency was acetabular cartilage lesions. Conclusion 3.0 T MRI provides a useful assessment of patients in whom a FAI is clinically diagnosed. A high-resolution, nonarthrographic technique can provide preoperative information. Acetabular labrum disorders and femoral-acetabular cartilage lesions maybe characterized as an earlier period of FAI.
5.Application of motivational interview in education on diabetes in special clinics

Objective To study the effect of motivational interview on health education to patients with diabetes.Methods Fifty patients with diabetes were randomly divided into the control group and the intervention group by using random digit table.The control group was given routine health education and the latter motivational interview.The two groups were compared in terms of self-efficacy and fasting blood sugar and glycated hemoglobin indexes.Result The self-efficacy,fasting blood sugar and glycated hemoglobin indexes in the intervention group were all better than those of the control group(P<0.01).Conclusion Motivational interview can effectively improve the patients’self-efficacy,do good for controling blood sugar level and meanwhile improve the quality of health education.
7.Analysis of hysteroscopy results of 539 patients accept in vitro fertilization-embryo transfer

Objective To evaluate uterine cavity of the patients who accept the treatment in vitro fertilization‐embryo trans‐fer(IVF‐ET)and determine the clinical value of hysteroscopy for the patients .Methods Between January 2012 and May 2014 ,539 cases who accepted the IVF‐ET and hysteroscopy ,were divided into two grops ,group A(258 patients examinated before IVF‐ET cycle) and group B(281 patients after the IVF‐ET failure) ,and retrospective analysis was performed for their uterine cavity and cervix canal data .Results The total incidence of abnormal hysteroscopic findings was 49 .54% ,and group B was significantly high‐er than that in group A(54 .10% vs .44 .57% ,χ2 =4 .5 ;P=0 .043) .The percent of abnormal cervix canal was 18 .74% ,and there wasn′t significant difference between the two groups .Conclusion The incidence of abnormal hysteroscopic findings in IVF‐ET is higher than that in normal patients ,so hysteroscopy has an important clinical value for diagnosis and evalution ,especially for pa‐tients with failure of IVF‐ET .
8.Mechanism of Escherichia coli injury under UV disinfection

Objective To study the inactivation kinetics and injury mechanism of Escherichia coli under UV disinfection in drinking water.Methods The inactivation kinetics of E.coli ATCC 25922 was determined by plate count methods in the UV disinfection experiment.The morphology,cell membrane permeabilization and injury of biological macromolecules were observed under a transmission electron microscope (TEM).The rate of ONPG hydrolysis and changes in activities of antioxidant enzymes were observed Raman spectroscopy.Results the changes of damaged cells involved morphological damage such as loss of the structural integrity of the wall and membrane,condensation of cellular material and leakage of significant amounts of cytoplasmic material,a more than four-fold increase of cell membrane permeabilization and damage to the structure of protein,nucleic acids and phospholipid.Conclusion UV disinfection can lead to a multi-target damage.
9.Effect of local subhypothermia on oxyradicals and inflammatory reaction in patients with acute cerebral infarction

Forty five patients with acute cerebral infarction were randomized to two groups: in treatment group patients received local subhypothermia and conventional therapy, in control group patients received conventional therapy only. Clinical outcome was assessed by the National Institutes of Health Stroke Scale (NIHSS) on admission and at 7, 14 and 30 d after treatment. Serum neuron specific enolase (NSE), nitrogen monoxide ( NO ) , superoxide dismutase (SOD), interleukin-6 (IL-6 ) and intercellular adhesion molecule-1 (ICAM-1) were detected on admission and at 7,14 d after treatment The study showed that NIHSS scores of treatment group on 14, 30 d were lower than those of control group ( P < 0. 05 ). Serum NSE, NO, IL-6 and ICAM-1 levels significantly decrease; while serum SOD levels increased (P < 0. 05). In conclusion, local subhypothermia therapy can inhibit inflammatory reaction, reduce oxygen free radical formation and improve neurological function in patients with acute cerebral infarction.
10.Photodynamic Therapy and Sonodynamic Therapy

Objective: This paper summarized the research status and progress of photodynamic therapy (PDT) and sonodynam-ic therapy (SDT), and we hope this can be a useful reference for clinics or research. Methods: In this review we expound their molecular mechanisms in detail and classify the photosensitizer and sonosensitizer, indicate their applications for the cancer treatment and summarize their new treating modes. PDT and SDT have huge potential for cancer treatment. Results: PDT and SDT have achieved a certain results, whereas there are still many problems, such as tissue oxygen content, new photosensitizer and sonosensitizer, drug dosage and mechanisms of SDT. Conclusions: PDT and SDT, which are new methods for treating cancer, have shown a good application prospect. But PDT has not been universal in clinics and SDT is still in the experiment stage.
