The pathogenesis of barium-induced severe myocardial necrosis in dietary potassum-depleted rats was investigated. In this study four experiments were performed. The results of these experiments suggest that there exist a complex pathogenesis of myocardial lesion produced by barium in condition of potassium deficiency. The main mechanism seems to be the direct stimulating effect of barium on myocardial contractibility. It seems unlikely that barium-induced constriction of coronary arteries, hypertension, hypokalemia, and increased catecholamine secretion are the basic mechanism of the myocardial necrosis. But these factors may play a promoting or aggravating role in the development of the myocardial lesion. Potassium deficiency not only acts as a conditioning factor for barium-induced myocardial necrosis, but also relatively increases the cardiotoxity of barium. This may be the principal pathogenesis of severe myocardial necrosis produced by barium in combination with potassium deficiency.

Objective:To investigate the relationship between poly(ADP-ribose)polymerase(PARP) inhibition and the growth activity of murine colon carcinoma CT26 cell lines in vitro.Methods:The murine colon carcinoma CT26 cell lines were treated with PARP inhibitior 5-AIQ in vitro.MTT assay was used to determine the growth activity of CT26 cells.The expressions of PARP and NF-?B p65 in the nuclei of CT26 cells were investigated by double labelling immunofluorescence and laser scanning cofocal microscope.Results:The inhibitory rates of the growth in 5-AIQ-treated colon carcinoma CT26 cell groups were 17.52%、27.63% and 39.93%,respectively.The inhibitory rate increased with the raising of the 5-AIQ concentration(0.1mmol/L,0.5mmol/L,and1.0mmol/L),and the difference was significant(P

The effects of nitrendipine (Nit) on myocardial contractility, myocardial succinate de-hydrogenase (SDH) and cytochrome oxidase (CCO) activities in streptozotocin (65mg/kg) diabetic rats were studied. Four weeks after the induction of diabetes, the rats were treated with Nit (30mg ?kg -2/day) for 4 weeks. The results showed that ventricular diastolic function was affected after 4 weeks of diabetes, and both ventricular diastolic and systolic functions were obviously involved after 8 weeks. These changes were significantly improved in diabetic rats receiving Nit treatment. The myocardial SDH and CCO activities in diabetic animals were markedly lower as compared to controls. The attenuation of these enzyme activities in diabetic rats was significantly reversed by administration of Nit. These findings suggest that Nit treatment may exhibit some beneficial effects on diabetic cardiomyopathy.

ObjectiveTo investigate the effect of down-regulating epidermal growth factor receptor (EGFR) expression on Bleomycin induced pulmonary fibrosis in mice.MethodsForty 4 ～6 week aged C57BL/c male mice were randomly divided into control,bleomycin,bleomycin plus EGFR RNAi groups and RNAi negative control group.Bleomycin group were treated with bleomycin (3 mg/kg) by endotracheal injection on day 0,control group were treated with PBS.And bleomycin plus EGFR RNAi group were received EGRF siRNA plus bleomycin intratracheal administration.RNAi negative control group received negative EGRF siRNA plus bleomycin intratracheal administration.Mice were sacrificed 10 days after the treatments.Hydroxyproline (HYP) assay was performed in the lung tissue.The lung tissue slides were examined pathologically with H.E.staining,and EGFR mRNA expression was detected by reverse transcriptasepolymerase chain reaction (RT-PCR) technique.Western blot were performed to identify the protein level.of total EGFR and phosphorylated EGFR.ResultsHistological examination of lung specimens demonstrated that EGFR siRNA administration lessened lung fibrosis induced by bleomycin and significantly reduced HYP content (543.00±25.89 vs 900.73±31.77,P＜0.01).EGFR mRNA (0.31±0.05 vs 0.75±0.08,P ＜0.01) and protein expression(1.53±0.47 vs 2.56±0.37,P ＜0.01) in EGFR siRNA-treated mice was significantly decreased.The expression of phosphorylation of EGFR protein (1.78±0.35 vs 2.84±0.51,P ＜0.01) and EGFR protein in RNAi group was less than in bleomycin group.Conclusions EGFR RNAi reduced the BLM-induced lung fibrosis by inhabiting EGFR activation.

Objective To understand the level of tulnor necrosis factor-α(TNF-α)and asymmetric dimethylarginine (ADMA) in serum of healthy adults and primary hypertension patients with different left ventricle,and to explore the role of TNF-α and ADMA in the pathophysiological mechanism of heart damage due to hypertension.Methods Thirty health adults were recruited as control.Based on the left ventricular mass index(LVMI)and relative wall thickness(RWT),66 patients of hypertension were divided into 4 groups:normal left ventricle group with normal LVMI and RWT,concentric remolding group with normal LVMI and incremental RWT,typical concentric hypertrophy group with incremental LVMI and RWT,eccentric left ventricular group incremental LVMI and normal RWT.TNF-α was measured by radioimmunoassay and ADMA was measured through reversed phase high-performance liquid chromatography.Resuits TNF-α and ADMA increased with the severity of the left ventricle.Each left ventricle group was significantly higher than control group (P<0.01).The differences between left ventricle groups were statistically significant (P<0.01).Conclusions TNF-α and ADMA may participate in the development of left ventricle.

BACKGROUND: In association with advanced startover of hypothalamus-pituitary-sexual gland axis, the growth of skeleton is accelerated and epiphyseal fusion occurred earlier in puberty precocity children. In clinic,Chinese drug for nourishing yin and reducing fire retards bone age and the advanced fusion in the disease. At the age of sexual development, Chinese drug for benefiting kidney to supplement essence is replaced to promote sexual development and accelerate skeleton growth. The patients with puberty precocity are the same to that in normal puberty in sexual development and bone growing, just being advanced in time. But, it is still unknown the mechanism of Chinese drugs on bone growth and effects on somatostatin (SS) in bone growth.OBJECTIVE: To explore the regulation of kidney-nournishing Chinese drug on periventricular nucleus (PeVN) SS.DESIGN: Completely randomized group division and control experiment were designed.SETTING: Department of Endocrinology, Children's Hospital Affiliated to Shanghai Jiaotong University MATERIALS: The experiment was performed in Institute of Pediatrics of Children's Hospital Affiliated to Shanghai Fudan University from October 2001 to April 2002, in which, 24 SD white rats were employed, of purebred, female and clean grade. 4 groups were randomized, named the control A, the group with drugs of nourishing yin and reducing fire (experiment group No. 1), the control B and group with drugs of nourishing yin and reducing fire + benefiting kidney to supplement essence (experiment group No.2),6 rats in each group.METHODS: ① Control A:physiological saline was applied for gastricperfusion, 5 mL each time, once per day, totally for 1 month. Experiment No. 1 group: Chinese mixture for nourishing yin and reducing fire was applied for gastric perfusion, composed of shengdihuang (Radix Rehmanniae),zhiguiban (Chinemys reevesii.), huangbai (Cortex Phellodendri) and zhimu (Rhizoma Anemarrhenae) etc, 5 mL each time, once per day, totally for 1 month. Control B: physiological saline wasapplied for gastric perfusion,5 mL each time, once per day, totally for 2 months. Experiment No.2 group:Firstly, Chinese mixture for nourishing yin and reducing fire was applied for gastric perfusion, 5 mL each time, once per day, totally for 1 month.And then Chinese mixture for benefiting kidney to supplement essence was applied for breeding, composed of shudihuang (Radix Rehmanniae Preparata), guibanjiao (Colla Carapacis Testudinis), xianlingpi, lujiaojiao (Colla Cornus Cervi) etc., 5 mL each time, once per day, totally for 1 month.② In situ hybridization staining procedure with floating and routine immunohistochemical technique. Medical imaging analysis software was used to assay PeVN SS mRNA, SS immunoreactive neuron counts and absorption (A value) of positive cell area that reflects the gene expression and protein expression. Number of positive neurons of unit area refers to the positive neuron counts in each square millimeter.③ Singer-factoranalysis of variance was used for the comparison of measurement data. F test was adopted for difference comparison among groups.MAIN OUTCOME MEASURES: Effects of Chinese drugs for nourishing yin and reducing fire and benefiting kidney to supplement essence on PeVN SS gene mRNA and protein expressionRESULTS: 24 rats were all in result analysis. ① PeVN SS mRNA positive neuron counts, positive neuron counts of unit area and A value of positive cell area: Those in experiment No.1 group was higher remarkably than the those of control A (P ＜ 0.05); and those in experiment No.2 group were lower remarkably than those of experiment No. 1 group (P ＜ 0.05). ② PeVN SS positive neuron counts, positive neuron counts of unit area and A value of positive cell area: those in experiment No. 1 group: those in experiment No.1 group were higher remarkably than those of control A (P ＜ 0.05) and those in experiment No.2 group were lower remarkably than those of experiment No. 1 group (P ＜ 0.05).CONCLUSION: By controlling hypothalamus SS gene expression and protein expression, Chinese drugs for nourishing yin and reducing fire and benefiting kidney to supplement essence regulate the synthesis and releasing of pituitary growth hormone.

Oxidative stress plays an important role in the progression of diabetes mellitus. As a potent antioxidant, alpha-lipoic acid is able to clear free radicals and alleviate oxidative damages and therefore has been widely applied in the clinical management of diabetes mellitus. This article summarizes the clinical application of alpha-lipoic acid in alleviating diabetes mellitus-related oxidative damages, protecting vascular lesions, treating diabetic polyneuropathies, and modulating insulin sensitivity.

Parenteral nutrition associated cholestasis (PNAC) mainly occurs in children. Currently identified risk factors include premature infants, lack of enteral feeding, repeated infections, and toxicity or nutrient deficiency of parenteral nutrition solution. Recent studies have shown that nuclear receptor-mediated bile acid transporter may be the key site of PNAC pathogenesis, in which some cytokines play important roles. New anti-cholestatic therapy based on the regulation of expression of these molecules may prevent end-stage liver disease caused by PNAC.

Objective To investigate the differences in some measured paremeters on spino-pelvic sagittal X-ray films in middle-aged and elderly patients between lumbar spondylolysis and isthmic spondylolisthesis,and to explore the predictors that the spondylolysis at lumbar 5 may develope into spondylolisthesis in order to provide imaging details for the clinical treatment of teenage patients with lumbar spondylolysis.Methods Imaging data of 60 patients with spondylolysis at lumbar 5 and other 60 with isthmic spondylolisthesis in middle or elder age in our hospital were analyzed retrospectively,and a control group with 60 normal volunteers were recruited in this study.Some parameters including the angles of pelvic incidence (PI),pelvic tilt (PT),sacral slope (SS),lumbar-sacral angle (LSA)and lumbar lordosis (LL),and the sagittal vertical axis (SVA)were measured in sagittal radiographs of the spine and pelvis. Differences in sagittal parameters among 3 groups were analyzed by one-way variance analysis and SNK-q test.Results The PI,PT, SS and LL were higher,and LSA was lower in patients with isthmic spondylolisthesis than those in the control group and lumbar spondylolysis one (P<0.05).However,there was no significant difference in SVA (P>0.05).No significant differences in all spino-pelvic sagittal parameters were found between lumbar spondylolysis group and the control (P>0.05).Conclusion Such spino-pelvic sagit-tal parameters as PI,PT,SS,LL and LSA on X-ray film can be regarded as predictors that lumbar 5 spondylolysis may develop into spondylolisthesis and may provide imaging reference for the clinical treatment of teen-age patients with lumbar spondylolysis.

Fever model was made by intravenously injected leucocytic pyrogen(LP,EP). Chai Ge Jie Ji Tang CGJJT fluid was perfused into rabbits stomach by mouth. 65 New Zealand white rabbits were divided into four groups: (1) control group; (2) LP group; (3) CGJJT fluid group; (4) LP+ CGJJT fluid group.
