2.Study on Immunoregulation Effect of Exopolysaccharide Produced by Bifidobacterium spp

The immunoregulation effect to polysaccharides from Bifidobacterium spp. was investigated on the base of functional assessment standards of health food. Effects of the EPS on immunity were investigated by promoted the proliferation of spleen lymph cells, delayed type hypersensitivity reaction, the HC50 value and macrophage function assay in mice. Data showed that the EPS could obviously increase the ratio of swallowed chicken red blood cell by macrophage and the HC50 value in mice. However, no significant effect was found on the delayed type hypersensitive induced by sheep red blood cell, for only the low dose of 100 mg/(kg?d) EPS promoted the proliferation of spleen lymph cells. Bifidobacterium spp. EPS can certain immunomodulating function.
3.Reflection on the Selection and Evaluation of Essential Drug List

OBJECTIVE:To review the selection process and methods of WHO Essential Drug List and to analyze the problem and challenge of the selection and evaluation of National Essential Drug List in order to provide suggestion for the selection and evaluation of essential drug list in China. METHODS:The selection process of previous National Essential Drug Lists were compared with that of National Essential Drug Lists(2009 edition)and WHO Essential Drug List. The problem and limit of National Essential Drug List were found out to put forward countermeasure. RESULTS:The prominent problem of the selection and evaluation of National Essential Drug List were as follows:the categories of drugs were changed significantly and including of some drugs was not supported by scientific evidence. There was no indirection and standard for the evidencebased selection and evaluation of essential drug. CONCLUSION:Aiming at the problem of the selection and evaluation of National Essential Drug List,evidencebased evaluation research is conducted to establish indirection and standard for the selection of essential drug list and database. Professionals and institutions for evidencebased evaluation and selection of essential drug list should be established to promote evidencebased evaluation and selection of National Essential Drug List and make more scientific decision.
4.Determination of Content and Entrapment Efficiency of Transferrin Modified Vincristine-Tetrandrine Lipo-somes

OBJECTIVE:To establish the method for the determination of main components content and entrapment efficiency of transferrin modified vincristine-tetrandrine liposomes. METHODS:HPLC method was adopted to determine the content of vin-cristine. The determination was performed on ELITE C18 column with mobile phase consisted of methanol-15%triethylamine (70∶30)at a flow rate of 1.0 ml/min. The determination wavelength was set at 297 nm and column temperature was 30 ℃. The sample size was 20 μl. Free drug was isolated from liposomes by dextran gel column chromatography,and entrapment efficiency was deter-mined. RESULTS:The linear range of vincristine was 160-1 600 μg/ml (r=0.999 8,n=5) with average recovery of 99.20%(RSD=0.26%,n=9). RSD of precision test was 0.070%(n=5). The average content of vincristine in liposomes was 0.790 mg/ml(RSD=0.15%,n=3),and the average entrapment efficiency was 85.94%(RSD=2.08%,n=3). CONCLUSIONS:The method is accurate,reliable,simple and rapid,and can be used for the determination of the content and entrapment efficiency of vincristine in transferrin modified vincristine-tetrandrine liposomes.
5.Effects of JNK inhibition on insulin signaling pathway in HepG2 cells

Objective To investigate the effects of JNK inhibition on insulin signaling pathway. Methods HepG2 cells were treated with different kinds of JNK inhibitors for 12 h, and then the cells were treated with 10 nmol/L insulin for 5 min to stimulate insulin signaling pathway. Mitogen-activated protein kinases ( MAPK ) and insulin signaling pathways were analyzed by Western blot using the total cell lysates. Results JNK activity was significantly inhibited by JNK inhibitor JNKi-Ⅷand results showed that JNKi-Ⅷtreatment could reduce insulin signaling pathway in a dose-dependent manner. Furthermore, other JNK inhibitors including JNKi-Ⅴ, JNKi-Ⅲ, and SP600125 blocked JNK activity in HepG2 cells. Similar to JNKi-Ⅷ, these JNK inhibitors also impaired insulin signaling transduction in a dose-dependent manner. Conclusion In HepG2 hepatocytes, JNK activity inhibition blocks insulin signaling transduction.
6.Determination of the Content of Vinorelbine in Liposomes by HPLC

The study is aimed to establish a method to determine the content of Vinorelbine in liposomes by HPLC.The experiment was carried out on Kromasil C18(4.6 mm×250 mm,5 μm) colum with the mobile phase consistedof acetonitrile-0.06 mol·L-1 KDP buffer(pH 3.0,35:65) at a flow rate of 1.0 mL·min-1.The detection wavelength was at 215 nm and the column temperature was at 30℃.The result showed that the linear range of Vinorelbine was from 2.08 μg·mL-1 to 20.8 μg·mL-1(r=0.9999).The average recovery rate was 98.64%(RSD =1.09%).It is con cluded that the method used in this analysis is simple,precise and replicable with high recovery and accuracy.It can be used to determine the content of total Vinorelbine in liposomes.
7.Association of physical activities,sedentary behaviors with overweight/obesity in 9 -11 year-old Chinese primary school students

Objective:To explore the association of physical activities,sedentary behaviors with over-weight/obesity in Chinese children aged 9 -1 1 years,analyze the gender difference and heterogeneity of the association across different provinces,and provide evidence for prevention and control of overweight and obesity.Methods:Using the data of 40 692 children aged 9 -1 1 years from Chinese National Survey on Students’Constitution and Health in 201 0,the height and weight were measured and the time spent in physical activities,screen behaviors and homework were collected.We analyzed the associations among physical activities,sedentary behaviors and overweight/obesity in boys and girls separately,and assessed the heterogeneity of associations across different provinces with Meta-analysis.Results:The prevalence of overweight/obesity of the boys (27.1 %)was higher than that of the girls (1 2.9%),compared with the rural students (1 5.6%),the situation of overweight/obesity of the urban students (23.8%)was more serious,the prevalences of overweight/obesity were separately 22.6%,1 9.2% and 1 7.5% among developed,middle-developed and undeveloped social economic status subgroups.The boys with physical activities no more than 1 hour per day were more likely to be overweight/obese,with the OR of 1 .09 (95%CI:1 .02,1 .1 7).The girls with screen time more than 1 hour per day or homework time more than 2 hours per day were more likely to be overweight/obese,with the ORs of 1 .1 3 (95%CI:1 .02, 1 .26)and 1 .1 8 (95%CI:1 .03,1 .35)respectively.The high level of sedentary behaviors (more than 1 35min per day)was associated with overweight/obese in the girls,and the OR was 1 .1 9 (95%CI:1 .08,1 .33).There was no significant heterogeneity of effect values between physical activities and over-weight/obesity in the boys and between sedentary behaviors and overweight/obesity in the girls across the different provinces (P >0.05).Conclusion:The associations among physical activities,sedentary be-haviors and overweight/obesity existed in Chinese children aged 9 -1 1 years,which were different in boys and girls.There was no statistically significant heterogeneity across different provinces.The strate-gies to prevent and control overweight/obesity in primary school students should focus on gender-specific needs.
8.Clinical efficacy of combined procedures for the surgical correction of congenital ptosis accompanied by epicanthus

Objective To observe the effects of combined surgeries for the correction of congenital ptosis accompanied by epicanthus.Methods 80 cases (105 eyes) of congenital ptosis were analyzed in this study.64 cases (80 eyes),with levator muscle strength more than 4 mm,were treated with suspension of frontal aponeurosis,and 16 cases (25 eyes),with levator muscle strength less than 4 mm,were treated with shorten operation of levator palpebrae superioris.Furthermore,all these patients were corrected the epicanthus at the same time.Results Except the under correction in 5 cases (8 eyes),the remaining patients were corrected satisfactorily.Postoperative palperbal fissure length was averagely increased by 2.0± 1.0 mm,inner canthic diameter was averagely decreased by 4.0± 2.0 mm,the eyelid height was increased by average 9.0±1.0 mm,the inclination of palpebral fissure was 12.0°±2.0°;64 cases (80 eyes) were treated with suspension of frontal aponeurosis,postoperative recurrence were 7.5% (6/80),no postoperative recurrence were 92.5% (74/80),complication were 5.0% (4/80);16 cases (25 eyes) were treated with shorten operation of levator palpebrae superioris,postoperative recurrence were 8.0% (2/25),and no postoperative recurrence were 8.0% (2/25);complication rates were 4.0% (1/25).Conclusions This procedure of combined surgeries can reduce the recurrence rate effectively.Moreover,the satisfactory,functional,and aesthetic results can be obtained simultaneously with this method.
9.Formulation Optimization of Transferrin Modified Tetrandrine and Vincristine Liposomes by Central Com-posite Design-response Surface Method

OBJECTIVE:To prepare transferrin(TF)modified tetrandrine(TET)and vincristine(VCR)active targeting lipo-somes,and to optimize its formulation. METHODS:TF modified TET and VCR liposomes were prepared by ammonium sulfate gradient method. Using comprehensive score of encapsulation efficiency of TET and VCR as index,central composite design-re-sponse surface method was used to optimize and validate mole ratio of EPC/Chol,mole ratio of EPC/PEG2000-DSPE and TF mass fraction. RESULTS:The optimal formulation was that the mole ratios of EPC/Chol and EPC/PEG2000-DSPE were 1.5:1 and 20:1, TF mass fraction was 0.10%. The encapsulation efficiency of TET and VCR were 97.80% and 93.00%,respectively. The compre-hensive score was 94.44(n=3)which was close to the predicted value of 93.81. CONCLUSIONS:The optimal formulation is sta-ble and can be used for the preparation of TF modified TET and VCR liposomes.
10.Study on Anti-tumor Effects of Vinblastine Hydrophilic Group Modified Cationic Liposomes in Tumor-bearing Mice

OBJECTIVE:To study the anti-tumor effects of Vinblastine (VLB) hydrophilic group modified cationic liposomes in tumor-bearing mice. METHODS:Tumor-bearing model were induced by inoculating yellow ascites of S180 ascites tumor mice. Tumor-bearing mice were randomly divided into model group,VLB sulfate injection group,VLB liposomes group,VLB hydrophil-ic group modified liposomes group,VLB cationic liposomes group and VLB hydrophilic group modified cationic liposomes group, i.e. group A,B,C,D,E and F,with 18 mice in each group. Group A was given normal saline intravenously via mice tail,other groups were given VLB 1.5 mg/kg every 2 days for consecutive 5 times. The anti-tumor effects of different VLB preparations were compared,using living conditions,survival time,tumor volume and weight,and tissue pathological section as indexes. RE-SULTS:Compared with group A,B,C,D and E,the mice of group F were more active,and had longer survival time,smaller tumor volume and lighter tumor weight,with statistical significance(P<0.05). The tissue pathological section of mice in group F indicated that coagulation necrosis,disintegration,and dissolution of tumor cell nucleus. CONCLUSIONS:VLB hydrophilic group modified cationic liposomes have obvious anti-tumor effect,which are better than other VLB preparations.
