1.Cytological study on vitreous extracts in Eales disease

To study the pathcytological mechanism of Eales disease and to provide a histological testimony for an effictive therapy.Methods45 cases(48 eyes) of Eales disease that were diagnosed according to vitreous hemorrhage and proliferative vitreoretinopathy or secondary retinal detachment.Cytomorphological examination was taken and vitreous extracts were examined by immunohistochemical staining.ResultLymphocytes and fibroblastes were most of those cells in the vitreous extracts.Immunohistology and electron microscopy revealed that lymphocytes were 78.8% or 61.5% in all specimens,respectively.ConclusionProliferative vitreoretinopathy induced by Eales disease closely relates with lymphocytes.The results suggested that autoimmunity might play an important role in the pathogenic mechanism of the disease
2.Effects of Sinomenine on Proliferation of Human Fibroblast-like Synoviocyte and Expression of MMP-3 mRNA from Patients with Rheumatoid Arthritis

Objective To observe the effect of sinomenine on the proliferation of fibroblast-like synoviocytes (FLS) and expression level of MMP-3 secreted by FLS in rheumatoid arthritis in vitro. Methods FLS were obtained by digesting synovial tissues with collagens and were divided into four groups:sinomenine high, middle, low concentration and control group. The proliferation of FLS was assessed by methyl- thiazolyl-tetrazolium (MTT) assay and the mRNA expression of MMP-3 was measured by semi-quantitative RT-PCR respectively after treated by sinomenine. 3.Effects of icariin on the expressions of osteoprotegerin and RANKL in rat osteoblasts

Objective To investigate the effects of icariin on the expressions of osteoprotegerin(OPG)and receptor activator of nuclear factor-kB ligand(RANKL)mRNA in rat osteoblasts cultured in vitro.Methods Calvarial osteoblasts were obtained from newborn SD rats aged<24 h by trypsin-collagenase digestion method. After treatment with different concentrations of icariin(10~(-10)-10~(-4) mol/L)for 48 h,total RNA was isolated from osteoblasts.Expressions of OPG and RANKL mRNA were detected by semiquantitative RT-PCR.Results Icariin remarkably increased OPG mRNA levels and decreased RANKL mRNA levels in calvarial osteoblasts in a dose- dependent fasion with a maximum effect at a concentration of 10~(-7)mol/L(P<0.05).Osteoblasts pre-treated with icariin prevented the inhibitory effect of dexamethasone on OPG mRNA(0.570?0.093 vs 1.083?0.081)and the stimulative effect on RANKL mRNA(1.079?0.050 vs 0.718?0.082)(both P<0.05).Conclusion Icariin upregulates OPG expression and downregulates RANKL expression in osteoblasts,which may explain the beneficial role of icariin in preventing and treating osteoporosis.
4.Effect of regulation in human umbilical vein endothelia cells treated by microRNA-132

AIM:To evaluate the regulatory effect of microRNA-132 ( miR - 132 ) in human umbilical vein endothelial cell ( HUVEC) .
●METHODS: ln vitro cultured human umbilical vein endothelia cells in hypoxic environment for 6h, then maintained under normal oxygen condition for 3h, 6h, 12h, 24h. miR-132 and peroxisome-proliferator-activated receptor-γ coactivator-1α ( PGC-1α) expression was detected by quantitative Real - time polymerase chain reaction and Western blot analysis. Human umbilical vein endothelial cells transfected miR-132 mimic and miR-132 inhibitor( anti-miR-132 ) were measured by quantitative Real-time polymerase chain reaction and Western blot.
●RESULTS: miR - 132 and PGC - 1α expression was significantly ( P < 0. 01 ) upregulated in the hypoxic environment of cells at 3h compared with the normal oxygen condition. After cells transfection, the hypoxic environment the miR-132 and PGC-1α expression were markedly increased compared with the normal oxygen condition. The cells transfected miR- 132- mimic, the expression of the miR-132 and PGC-1α were higher than that of transfected anti-miR-132 and contrast group ( P<0. 01).
●CONCLUSION:miR-132 level is highly expressed in the HUVEC under hypoxia and may be an effect of regulation for PGC-1α.

7.Pharmacokinetics and in vitro-in vivo correlation evaluation of self-emulsifying drug delivery system and conventional tablets of aniracetam

AIM: To evaluate the correlation between in vitro release and in vivo absorption of aniracetam in conventional tablets and self-emulsifying drug delivery system (SEDDS), to investigate pharmacokinetics of aniracetam self-emulsifying drug delivery system and conventional tablets of aniracetam after oral administration to rats. METHODS: Dissolution behavior of these formulations was evaluated in vitro to assess the properties of dosage forms. And a new RP-HPLC method was developed for the in vivo quantitative determination of 4-p-anisamidobutyric acid (PABA), the active metabolite of aniracetam. To approach the in vitro-in vivo correlation, fraction absorbed in vivo (f) was calculated by Wagner-Nelson method, and then compared with in vitro released drug percentages (Q%). RESULTS: Aniracetam was released rapidly from SEDDS with 80%±4% of accumulation dissolution rate compared to that from conventional tablets at 15 min. The recovery of active metabolite of aniracetam was about 90%, and the intra-days and inter-day precision were within 4% and 6%, respectively. The AUC0-∞ value of aniracetam SEDDS was (11 168±2 395) ng·mL-1·h, which was about 3 folds greater than conventional tablets. The parameter MRT0-∞ of aniracetam SEDDS and conventional tablets were (2.7±0.6) h and (1.7±0.6) h, respectively, and the difference was statistically significant(P<0.05). The linear equation of in vitro-in vivo correlation for conventional tablets was obtained by regression as well. Whereas nonlinear correlation was obtained for aniracetam SEDDS, which fitted the quadric model very well and the correlation coefficient was 0.972. CONCLUSION: Aniracetam can be released faster from SEDDS than that from conventional tablets, and SEDDS improved the bioavailability of aniracetam significantly. The SEDDS composed by oil and compound surfactants which could enhance the absorption showed the expressing rate of dissolution, and those formed the o/w microemulsion with gastrointestinal liquid could absorb through lymphatic transport route.
9.A review of drug metabolism under hypoxia environment at high altitude.

The special environmental features of high altitude, such as hypobaric hypoxia, low temperature, arid, high solar radiation, variable climate and geochemical anomaly, cause great effects on human physiology and health. It will provide valuable references and new ideas to study drug's metabolism in special environment of high altitude hypoxia, and give the guidance to clinical reasonable medication, avoiding adverse reactions and personalized medicine in plateau areas. This article reviewed the effect of high altitude hypoxia on drug metabolism, elaborated metabolic characteristics of some drugs and the activity and expression of drug metabolism enzymes under hypoxia environment at high altitude, and discussed related mechanism.

10.Papaverine treats vasospasm caused by PICC intubation with B-mode ultrasonography

Objective To study the effect of papaverine on vasospasm caused by PICC intubation with B-mode ultrasonography. Method Three mg papaverine were injected into the median cubital vein for at least 2 minutes in 15 patients with vasospasm. Results The vasospasm in the 15 patients was relieved 36~270 s seconds after injection. The followed intubation was all successful. There was no abnormality in their liver function and heart rate, or abnormal bleeding, or other serious complications. Conclusion Papaverine can relieve vasospasm caused by PICC intubation, so it can allow another intubation. It also can avoid delayed intubation reduce patients′pain and cost and reduce psychological pressure of the nursing practitioner.
