2. Expression of Bcl-2 and Bax in renal tissues of patients with hepatitis B virus-associated glomerulonephritis

Objective: To investigate the expression of Bcl-2 and Bax in renal tissues of patients with hepatitis B virus-associated glomerulonephritis (HBV-GN). Methods: Twenty HBV-GN specimens with complete nephrology data and 10 normal renal specimens were randomly chosen for the present study. Cell apoptosis was detected by means of terminal deoxynucleotidyl transferase mediated d-UTP nick end labeling (TUNEL) and the apoptotic index was calculated; immunohistochemistry was used to detect the protein expression of Bax and Bcl-2. Results: The apoptotic index in HBV-GN group was obviously higher than that of the control group; the apoptotic cells were mainly distributed in the proximal and distal renal tubules and the collecting duct epithelial cells, seldom seen in the glomerular cells. The expression of Bcl-2 in HBV-GN patients was predominately present in the renal tubular epithelia cells (positive in the plasma, membrane and nuclear); the expression of Bax was found in both glomerular cells and renal tubular cells, mainly in tubular epithelial cells, seldom seen in Bowman's capsule or glomerular mesangial region. Conclusion: Apoptosis in the kidney of HBV-GN patients mainly occurs in the renal tubular epithelial cells; expression of Bax and Bcl-2 is mainly in the renal tubular epithelial cells, suggesting that the injury of tubular interstitial damage may be one of the important factors for the development of HBV-GN.
3.Progress in relationship of the long non-coding RNA and sepsis

The mechanism of long chain non-coding RNA (lncRNA) is diverse. LncRNA can regulate the expression of genes at multiple levels and is closely related to some pathological process, such as tumor formation, viral replication, inflammatory damage, etc. The abnormal expression of lncRNA is involved in the patho-physiological changes of the disease, and plays a key role in many links, such as the stability of mRNA, the regulation of translation level, protein transport, RNA processing and modification, and so on. The development of sepsis inflammatory injury is a complex process involving multiple genes and pathways, in which the abnormal structure and expression of many encoding genes or non-coding genes exist.This paper review the research progress on lncRNA in the regulation of chondriokinesis, pro-inflammatory cytokines, calcium ion transport, nuclear factor-κB (NF-κB) in signal pathway and other inflammatory lesions, which may provide a new method for the diagnosis and treatment of sepsis.

Objective To explore the effects of cryopreservation on biological characteristics of cultured limbal stem cells.Design Experimental study.Participants Twenty New Zealand white rabbits.Methods Twenty New Zealand white rabbits(40 eyes) were preparated of peripheral cornea with 2 mm and 2 mm circular shallow conjunctival limbal graft.Rabbit limbal stem cells(Rb-LSCs) used for cell-suspention culture were isolated form New Zealand white rabbit limbal tissues.The limbal stem cells,having formated monolayer cultured limbal stem cells,were cryopreservated one month,then thawing.By immunofluorescence staining,flow cytometry,inverted microscope,MTT assay,we compared the subculture of limbal stem cells cryopreservation before and after the structure and properties.Main Outcome Measurements Limbal stem cell activity and biological characteristics.Results Compared with freshing cells,the cells after cryopreservation in morphology were poor adherence late,small nuclear cytoplasm ratio,irregular shape.Im munofluorescence staining to detect the low-temperature frozen cells ABCG2 monoclonal antibody 81.7% positive rate decreased to 46.9%,the difference was statistically significant(P

7.Effects of TNF-α and TGF-β1 on Gli2 expression in primary leukemia cells

Objective To investigate whether TNF-α can promote, but TGF-β can reduce Gli2 expression in primary leukemia cells. Methods (1)Western blot was used to detect Gli2 protein expression in leukemic cells. (2)Primary leukemia cells were treated with TGF-β1 or TNF-α,and expressions of Gli2 and TGF-βR were detected. (3)Primary leukemia cells were treated with TGF-β1,TNF-α or/and SIS3, and Gli2 protein expression was detected. Results (1)The primary leukemia cells could express Gli2 protein. (2)Significant reduction of Gli2 mRNA was observed in 1 ~ 10 ng/mL TGF-β1-treated primary leukemia cells for 12 ~ 24 h, No significant differences of Gli2 mRNA was found between the control group and TNF-α group. (3)Protein expresson of TGF-βRⅠand TGF-βRⅡ in 5 ng/mL TNF-α-treated primary leukemia cells for 24 h was higher than that of the control group. (4)Gli2 portein expression was lower in TGF-β1 with or without TNF-α-treated primary leukemia cells for 24 h than that in the control group, and Gli2 portein expression in the TGF-β1 plus TNF-α group was lower than that in TGF-β1 group. Conclusion Combinations of TGF-β and TNF-α can efficiently inhibit Gli2 expression than TGF-β alone in primary leukemia cells.
8.Pathological observation of degenerative aortic valve disease

Objective To investigate the normal aortic valve histological manifestation and histopathological changes of degenerative aortic valve.Methods In Tianjin Chest Hospital,there were patients due to aortic valve disease undergoing aortic valve replacement surgery in the Department of Cardiovascular Surgery,and in 22 cases,their pathological aorta tricuspid valves were successfully excised from May 2009 to May 2010.The aortic valve tissues were collected,and according to hematoxylin and eosin (HE) staining of the degenerative calcified aortic valves with stenosis,they were divided into pathological valvular calcification locus group and non calcification locus group.Six morphologically normal aortic valve tissues were taken as a control group.The situation of collagen fibers,elastic fibers and collagen content in normal and disorder valves were analyzed by using of IDA-2000 high definition color medical image analysis system.Results With the naked eye,the thickness of the valve leaves with stenosis was increased,and at the valve leaf close to the side of arota,calcified foci were produced.Under light microscope,inflammatory changes were found in the aortic valve endothelium,in the tubular wall,there were aggregations of newly formed capillary with relatively thin wall,cholesterol crystals,cells similar to foam cells and diffuse and nodular calcification commonly seen,Compared with the normal valve,the ratio between collagenous and elastic fibers no matter at calcified or non-calcified region was greater in pathological valve [(3.19-± 1.01)%,(2.62-± 0.34)% vs.(1.89-± 0.29)%,both P < 0.05],collagen content (ratio =collagen/valve area) was also higher [(65.17 ± 11.48)%,(39.25-± 11.15)% vs.(22.99 ± 4.78)%,P < 0.05] in pathological valve,and the changes in calcified region were more significant than those in non-calcified region (both P < 0.05).Conclusions Proliferation of collagen and degradation of elastic fibers are obvious in degenerative aortic valve,and the changes are more prominent at calcified region.

10.Association of blood lipid with distant metastases and clinical response in patients with hormone receptor negative breast cancer

Objective To investigate the relationship between dyslipidemia and distant metastases in pa-tients with hormone receptor( HR) negative breast cancer,and to explore the impact of blood lipid change during chemotherapy on clinical response.Methods We collected the complete clinicopathological data and fasting blood lipid in 154 patients with HR negative breast cancer.The secondary blood lipid level were measured on day14 after two cycles of chemotherapy in patients with distant metastases.Single factor chi -square test was choosen to analyze the relationship between clinicopathological characteristics,including baseline lipid level,and distant metastases,then independent risk factor for distant metastases were evaluated by logistic regression model. Data about altered blood lipid and clinical response analysis were performed using paired t-test pre-or post-chemotherapy.Results Our results from single chi-square test showed that distant metastasis from breast canc-er was significantly associated with tumor size,local lymph node metastases,grade,higher total cholesterol( TC) , higher triglycerides(TG)and higher low-density lipoprotein(LDL)(P<0.05).For distant metastases,inde-pendent risk factors were tumor size(OR=1.563),local lymph node metastases(OR=1.983),higher TC(OR=1.502),higher TG(OR=1.877).Decreased trend of TC,TG and LDL and increased trend of HDL were showed in disease control( PR+SD) group,and TG had significantly tend in objective response( PR) group.Conclusion Hyperlipidmia is significantly associated with distant metastases in patients with HR negative breast cancer,andblood lipid levels decrease with effective anti -tumor chemotherapy.Dynamic monitoring of blood lipid levelscould be a candidate surrogate biomarker for outcome prediction and distant metastases in patients with HR negativebreast cancer.
