1.Effects of prolactin on in-vitro interactions between thyrocytes from patients with Graves′ disease and autologous peripheral blood mononuclear cells

Objective To study the effects of prolactin on interactions between Graves′ disease (GD) thyrocytes and autologous peripheral blood mononuclear cells (PBMC) in coculture. Methods Primary thyrocytes and autologous PBMC were cocultured at various levels of ovine prolactin (oPRL) in vitro. The activation and proliferation of PBMC as well as HLA-DR and CD40 expressions on thyrocytes were determined by immunofluorescent staining and flow cytometry. Results With PBMC in GD cocultures, oPRL caused significant increase in the percentage of CD4+CD25+ cells at 200 μg/L [(13.08±2.54)%, P<0.01] and in the proliferative index at 200 μg/L [(17.82±3.02)%, P<0.01] and 1 000 μg/L [(16.57±2.56)%, P<0.05], but not at 12.5 μg/L [(10.43±2.38)% and (14.08±2.67)%, respectively] or 50 μg/L [(9.94±2.54)% and (14.56±2.80)%, respectively)]. The values at 200 or 1 000 μg/L as above were also significantly different from those at 12.5 or 50 μg/L. The percentage of CD40+ cells [(48.25± 6.63)%,(52.28±6.94)%] and delta mean fluorescence intensity (dMF; 42.94±10.24,49.51±12.34) were significantly lower in thyrocytes in GD cocultures treated with 200 or 1 000 μg/L oPRL than those untreated [(58.38±6.62)% and 67.30±20.20] or treated with two other concentrations of oPRL. The percentage of HLA-DR+ cells [(46.79±7.51)%, P<0.01] and dMF (21.02±5.43, P<0.01) were significantly higher in the thyrocytes treated with 50 μg/L oPRL than those untreated [(33.51±8.58)% and 13.91±3.88] or treated with three other concentrations. There were no significant differences between the dose groups of oPRL but those mentioned above. Conclusions Prolactin could affect interactions between GD PBMC and autologous thyrocytes and might eventually influence the shift of helper T cells. High prolactin levels might aggravate GD.
2.Preliminary study on relationship of disease-syndrome-symptom of ovulatory disorder infertility based on factor analysis.

OBJECTIVEIn this study, the clinical data of 240 cases of data acquisition on ovulation in patients with anovulatory infertility.
METHODUsing the method of factor analysis, summed up the distribution situation of various syndromes and the relationship of disease-syndrome-symptom.
RESULTFrom the study we found that polycystic ocary syndrome > premature ovarian failure > corpus luteum insufficiency > luteinized unruptured follicle syndrome > hyperprolactinemia > anovulatory dysfunctional uterine bleeding. Using the factor analysis method, the syndromes of anovulatory infertility were divided into six types and the distribution proportion of them is: kidney Yang deficiency and blood stasis > kidney Yang deficiency > kidney Yin deficiency and blood stasis > kidney Yin deficiency and liver Qi stagnation syndrome > kidney Yang deficiency and phlegm dampness stagnation > kidney Yin deficiency. By means of statistics, the traditional Chinese medicine (TCM) symptoms as abdomen pain before menstrual onset, obfuscation colour, cold lower abdomen, insomnia, fatigue, eyes dark, face dark or ring lip dark, before and after menstruation breast swelling, chest tightness, irritable insomnia, depression and lower abdomen pain during menstruation etc contribute high degree in the process.
CONCLUSIONPolycystic ovary syndrome and premature are the main causes of ovulatory disorder infertility and the disease incidence rate assumed the tendency which rose year by year. The treatment of the disease of kidney yang deficiency syndrome scales biggest the disease degree of contribution. Blood stasis is the important factor in the process of pathogenesis evolution. In the study of disease distribution, we found that symptom distribution trend roughly the same and the same symptom severity in different diseases with different, which provides experience for the theory-different diseases with the same therapeutic method. These conclusions provide guidance and reference evidences for clinic practice.

3.Clinical Observation on Treatment of 35 Cases of Knee Joint-Effusion with Moxibustion

Objective To observe clinical therapeutic effect of treatment of knee joint-effusion with moxibustion plus auricular point sticking.Methods 35 cases(51 knees)of knee joint effusion were treated by moxibustion plus auricular points sticking for once daily.Results The total effective rate was 96.1%.Conclusion The treatment of moxibustion plus auricular point sticking has a definite therapeutic effect on knee joint-effusion with the function of reducing swelling and alleviating pain,being worthy of spread.
4.The effect of comfort care to alleviate postoperative pain of tibial eminence fracture by ;fixation

Objective:To study the effect of comfort care to alleviate postoperative pain of tibial eminence fracture by fixation. Methods:55 patients with tibial eminence fracture between Apr 2013 and Mar 2015 in our hospital were selected and divided into observation group with 26 patients and control group with 29 cases according to the nursing methods. The control group was given the orthopaedic routine nursing method, and the observation group was given the comfortable nursing method to compare two groups of patients with postoperative pain and psychological status before and after the intervention (HAMA anxiety score and HAMD depression scale). Results:There were significant differences between two groups of patients with pain score after the intervention (t=9.545, P<0.05), and HAMA and HAMD scores decline. HAMA and HAMD scores in the observation group were significantly lower than that in the control group (t=11.593, t=10.628;P<0.05). Conclusion:Comfort care can significantly relieve postoperative pain of the tibial condyle spine fractures and improve postoperative anxiety and depression. It has good clinical application value.
5.Inductive effect of Cortex Periplocae extract on apoptosis of human gastric cancer cells BGC-823

Objective To study the inductive effect of Cortex Periplocae extract(CPE) on apoptosis of human gastric cancer cells BGC-823 and its mechanism.Methods The cell morphology and super-microstructural changes of apoptosis were analysed by Giemsa staining and electric microscope,respectively.The BGC-823 apoptosis ratio,cell cycles,and changes of apoptosis in DNA level were studied by Flow Cytometry and agarose gel electrophoresis.The genes mRNA and protein expression of apoptosis-related genes bcl-2,bax,and survivin were studied by RT-PCR and immunology cell chemistry method.Results After treatment with CPE,BGC-823 cells showed some typical morphologic features and super-microstructural changes of apoptosis.DNA agarose gel electrophoresis showed characteristic "DNA ladder" pattern.Most BGC-823 cells were arrested at G_2/M phase.Some typical subdiploid peaks before G_0/G_1 phase were observed.The apoptotic rate of BGC-823 was 18.9% after 250 ?g/mL CPE treated for 48 h.The gene mRNA and protein expression of bcl-2 and survivin were inhibited by CPE,whereas that of bax was up-regulated.CPE could enhance the life span of S_(180) bearing mice in a dose-dependent manner.Conclusion(CPE can) inhibit the tumor growth by arresting the BGC-823 cell cycle at G_2/M phase and inducing BGC-823 apoptosis.Its mechanism is related to the inhibition on gene mRNA and protein expression of bcl-2 and survivin,and enhancement of those of bax.

Objective To review the physiological characters and potential function of chondroitin sulphate proteoglycan,CSPG(NG2) glial cells in the central nervous system. Methods In conclusion of the NG2 glial properties and the global, novel researches on NG2 cells. Results Cells that express the NG2 proteoglycan comprise a unique population of glial cells in the central nervous system.Study on their role in the CNS becomes more and more popular.Previous researches mainly focus on the differentiation and distinction.Now the relationship between NG2 glial cells and neurons,and the synaptic plasticity is becoming a hot topic.Conclusion This review referred the research development of NG2 glial cells,including the differentiation,migration,their interaction with neurons,and the potential role in glial cell regeneration in disease systematically.

8.Preparation and Identification of Recombinant Vasoactive Intestinal Peptide

In order to prepare the recombinant vasoactive intestinal peptide (VIP) using intein mediated rapid purification system,the cDNA encoding the recombinant VIP was designed and synthesized according to the preference of E.coli,and then was cloned into the expression vector PTWIN. The recombinant plasmid PTWIN-VIP was transformed into expression host E.coli strain ER2566.The fusion protein consisting of the recombinant VIP,intein and chitin binding domain was expressed and purified by chitin affinity chromatography. The target peptide was released from the fusion protein by changing the temperature and the pH of the cleavage buffer. The molecular weight of the recombinant VIP was determined by the mass spectrometry and the results was conformity with the theoretical value. The preliminary bioactivity assay indicated that the recombinant VIP decreased the serum resistin levels significantly in LPS-induced acute inflammation. The preparation and the characterization of anti-inflammatory effects of the recombinant VIP layed the foundation for its further application.

Objective To evaluate the development of immature oocytes after freezing-thawing by conventional cryopreservation method for mature oocytes.Methods Immature oocytes were collected from stimulated ovaries of intracytoplasmic sperm injection(ICSI)cycles.Immature oocytes were in vitro matured directly or after slow freezing-fast thawing and immunostained for tubulin and chromatin and at last visualized by confocal microscopy.Results No statistical difference was found in maturity rate between freezing groups and the controls.There was a statistically significant increase in abnormalities of chromosome(23.7% vs. 50%)and spindle(28.9% vs.53.9%)in the GV freezing group compared with the GV control(P
