1.Clinical Research on Elderly Hypertension of Kidney Deficiency and Blood Stasis Syndrome Intervened with Bushen Huoxue Decoction

Objective To observe the clinical efficacy of Bushen Huoxue Decoction on elderly hypertension of kidney deficiency and blood stasis syndrome. Methods Totally 106 patients with elderly hypertension of kidney deficiency and blood stasis syndrome were divided into control group (53 cases) and treatment group (53 cases), according to random number table method. The control group was treated with routine western medicine, while the treatment group was given Bushen Huoxue Decoction along with western medicine treatment for 8 weeks. The changes of TCM syndrome, blood pressure, TG, TC, LDL-C, HDL-C, high sensitivity C reactive protein (hs-CRP), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α) before and after treatment of the two groups were observed. Results The total effective rate of anti-hypertension was 94.33%(50/53) in the treatment group and was 77.35% (41/53) in the control group. The total effective rate of clinical symptoms was 96.22%(51/53) in the treatment group and was 79.24%(42/53) in the control group. After treatment, TG, TC, LDL-C, HDL-C, hs-CRP, IL-6, and TNF-α were improved significantly in the treatment group, with statistical significance compared with the control group (P<0.05). Conclusion Bushen Huoxue Decoction combined with western medicine achieves the good efficacy on elderly hypertension of kidney deficiency and blood stasis syndrome.
3.Impact of sleep deprivation on coronary heart disease and progress in prevention and treatment with traditional Chinese medicines.

Sleep deprivation (SD) has been taken as an independent predictor for cardiovascular risks, which was closely related to the increased morbidity and mortality in coronary heart disease (CHD). In this article, after reviewing the impact of modern medical method sleep deprivation on CHD and studies on principle method recipe medicines for preventing and treating CHD, the authors observed the autonomic nerve dysfunction, hormonal metabolism dysfunction, endothelial dysfunction and inflammatory responses after sleep deprivation, which can cause or aggravate CHD. On the basis of the traditional Chinese medicine theories of "heart dominating the blood and vessels and the mind", the authors considered that traditional Chinese medicines can tonify heart and soothe the nerves, reducing all of the risk factors through multi-target and multi-pathway, and improve sleep and decrease the risk factors caused by sleep deprivation, which provides a new idea for the prevention and treatment of CHD.

6.Treatment of 1 500 cases of peripheral facial paralysis with constant magnetic aerobic atraumatic method along meridian

BACKGROUND: Peripheral facial paralysis is treated at present only with traditional traumatic methods, such as acupuncture, moxibustion,needling with physical therapy, point-injection, thread burial therapy,scraping therapy, etc. Those methods do not act directly on the lesion of facial nerve and they are long in duration of treatment and low in curative rate. The constant magnetic aerobic atruamatic method along meridian just makes up the shortcoming of the traditional therapies.OBJECTIVE: To observe the therapeutic effects of constant magnetic aerobic atruamatic method along meridian on treatment of peripheral facial paralysis.DESIGN: Case analysis.SETTING: Department of Facial Paralysis, College of Stomatology, Fourth Military Medical University of Chinese PLA.PARTICIPANTS: Totally 1 500 cases of first diagnosed peripheral facial paralysis were employrd in 18 provinces and autonomic regions in China from June 1998 to June 2004, of which, 935 cases were male and 565 cases female. Of 103 cases were aged varied from 1.5 to 19 years, 1 105 cases from 20 to 50 years and 292 cases from 51 to 74 years. Of 672 cases were left palsy and 828 cases were right palsy. The duration of sickness was varied from 1 day to 40 years.METHODS: Constant magnetic aerobic therapy for 20 minutes was applied on facial-palsy effective points for all of cases [No.1 point: in the depression inferior to mastoid process behind ear lobe on the affected side,corresponding to the mastoid foramen, from which, the facial nerve emerges. No.2 point: on the arch top of mastoid process behind ear lobe on the affected side. No.3 point: about 3.33 cm (1 cun) above No.1 point. No.4: in the conjunction between the point about 0.5 cm posterior to the external auricular tract and the one 0.5 cm inferior to helix cms. No.5 point:about 1 cun anterior to ear apex. No.6 point: on the top of the upper border of helix on the affected side] and supplementary points [Qianzheng (Extra),Dicong (ST 4), bilateral Hegu (LI 4)], once per day, 10 days makes one course. There were a couple of days at interval between two courses, totally two courses were required. Point and oxygen therapies were applied simultaneously. Oxygen therapy: 20 minute/time, once per day, 10 days made one course, at the volume of oxygen 2-4 L/minute. Evaluation of therapeutic effects: It was to compare the changes between the affected side and healthy side in forehand wrinkles, frowning, closing eyes, whistling, blowing and smiling and the changes between two sides in nasal labial groove and philtrum during showing the teeth and at static state. Comparison of bilateral muscularis function of healthy people and patients were obtained with the six-point and four-line measurement.MAIN OUTCOME MEASURES: Results of evaluation on clinical therapeutic effects.RESULTS: Totally 1 500 cases of peripheral facial paralysis entered result analysis, no dropped out case. After 20 days treatment, 1 385 cases (92.33%) were recovered, 92 cases (6.13%) appeared significant effect, 22cases (1.74%) were improved and 1 case (0.07%) was no effect. The total effective rate was 99.93%.CONCLUSION: Constant magnetic field in combination with oxygen therapy is characterized as no pain, no trauma, no cross infection and no second injury of facial nerve, safe and convenient operation, short duration of treatment and high curative rate. It improves remarkably the symptoms of facial paralysis and recovers facial function.
7.Effect of lipopolysaccharide on astrocyte proliferation and release of its calcium and nitric oxide and reactive oxygen species

BACKGROUND: Astrocyte distributes the most widely in the central nerve system,but the role of astrocyte in the pathogenisis of neurodegenerative diseases is still unclear.OBJECTIVE :To investigate the effects of lipopolysaccharide (LPS) on the level of the free intracelluar calcium, the production of reactive oxygen species (ROS) and nitric oxide (NO), and the cell viability of astrocytes.DESIGN: Randomized grouping controlled experiment.SETTING: The Brain Circulation Research Laboratory, Second Affiliated Hospital of Suzhou University.MATERIALS: The experiment was carried out in the Brain Circulation Research Laboratory, Second Affiliated Hospital of Suzhou University from May 2005 to August 2005. The astrocytes were derived from the cerebral cortex of 8 Newborn SD rats aged 1-3 days provided by the Animal Experimental Center of Suzhou University. LPS was purchased from Sigma, MTT assay kit and NO assay kit were obtained from Beyotime Biotechnology.METHODS: The astrocytes were isolated and cultured using the method described by Kevin St. P. McNaught. The cells were purified by proliferation repeatedly. Astrocytes were divided into control group, LPS5, 10, 20,40 mg/L group according to different dosage of LPS added. The cell viability at 30 min and 60 min respectively was measured using MTT method.The NO accumulation in cultured cell supernatant at 30 min was assayed with Griess reagent after the treatment of LPS. Calcium and ROS accumulation in cultured astrocyte of rats stimulated by different dosage of LPS was measured at 30 min and 60 min respectively by confocal laser scan microscope (CLSM).MAIN OUTCOME MEASURES: Cell viability of astrocyte, the change of level of NO, calcium and reactive oxygen species of astrocyte.RESULTS: ① The cell viability 60 min after administration of LPS 5 mg/L,LPS 10 mg/L and LPS 20 mg/L was higher than that of control group(P < 0.05). The cell viability of LPS 10 mg/L group was higher than that of LPS 5, 20 mg/L group.② NO production 30 min after LPS treatment of LPS 40 mg/L group was higher than control and the other 3 experimental groups (P < 0.05 ). ③ The calcium of the LPS 5, 10 and 20 mg/L group was 200-400 times higher than that of the base state. The calcium of the LPS 40 mg/L group was 1 000 times than its baseline state; Absolute fluorescence intensity of ROS was superior than the detect range of CLSM, we could speculate that the relative fluorescence intensity of the group LPS 40 mg/L was at least 300-400 times higher than that of its base state.CONCLUSION: LPS could increase the level of the free intracelluar calcium and induce the production of ROS and NO in astrocytes.
8.Renal transplantation in highly sensitized patients

Objective To explore the protocol of tissue matching and anti-rejection therapy in highly sensitized patients (HSP). Method The panel reactive antibody (PRA), human leukocyte antigen (HLA) matching and renal transplantation outcomes of 45 HSPs were retrospectively analyzed. Results Hyperacute rejection occurred in 2 patients. Acute rejection occurred in 9 patients and reversed by anti-rejection therapy. One year patient/graft survival rate was 95.6% / 91.1% respectively. Conclusions To avoid specific antibody through HLA matching is the key point for successful renal transplantation of HSP. Antithymocyte globulin (ATG) induction therapy combined with tacrolimus, mycophenolate mofetil therapy can decrease the rate of acute rejection and prolong graft survival.
10.Construction of a DNA vaccine encoding herpes simplex virus type 1 glycoprotein D

Objective To construct a recombinant plasmid DNA containing herpes simplex virus type 1(HSV-1) glycoprotein D (gD) gene.Methods The HSV-1 gD gene was obtained by polymerase chain reaction (PCR) and inserted into TA cloning vector pGEM-T, then cloned into the eukaryotic expression vector pcDNA3.1 to generate pLy-D. The recombinant plasmid pLy-D, which was confirmed by partial sequencing and restriction endonuclease analysis, was transfected into Cos-7 cells and used to inoculate ICR mice via muscular injection. Immunohistochemistry and enzyme-linked immunoabsorbent assay (ELISA) were employed to test the gD expression in transfected cells and the specific anti-HSV-1 antibody in the serum of immunized mice, respectively.Results The gD eukaryotic expression plasmid pLy-D was constructed. Using the immunohistochemistry technique, the gD expression in pLy-D-transfected cells was detected. The ELISA demonstrated that specific anti-HSV-1 antibody could be induced in immunized mice after three times injection.Conclusions We constructed HSV-1 gD eukaryotic expression plasmid pLy-D which could express gD protein in transfected cells and could induce humoral immune response in mice. This observation will be helpful in designing HSV prophylactic vaccine.
