1.Determination of oxymatrine and matrine in Compound Shiwei Tablets by HPLC

AIM:To establish an assay of oxymatrine and matrine in Compound Shiwei Tablets(Folium Pyrrosiae, Radix Sophorae Flavescentis, Radix Astragali, Herba Polygoni Avicularis, etc.) and Radix Sophorae Flavescentis by HPLC. METHODS: Diamonsil C 18 (10 ?m,250 mm?4.6 mm) analytical column was used.The mobile phase consisted of methanol-0.2% phosphoric acid solution (45∶55). The detection wavelength was 220nm. RESULTS : The average recoveries of oxymatrine and matrine in Compound Shiwei Tablets were 100.3% and 100.6% , respectively and in Radix Sophorae Flavescentis were 101.0% and 101.2%, respectlively. CONCLUSION: This method could be used for the quality evaluation of Compound Shiwei Tablets and Radix Sophorae Flavescentis.
2.An optimal method for cryopreservation of microamount round spermatids of the mouse.

OBJECTIVETo search for an optimal protocol and freezing conditions for the cryopreservation of microamount round spermatids of the mouse.
METHODSWe compared the survival rates of frozen-thawed microamount round spermatids of the mouse achieved by vitrification or standard slow freezing with different concentrations of glycerol (5, 7, or 9%) and different lengths of equilibrium time (0, 15, 30, 45, or 60 min).
RESULTSUnder the conditions of 7% glycerol and 30 min equilibrium, both vitrification and standard slow freezing achieved high survival rates of spermatids, and the former obtained an even higher rate than the latter ([72.9 ± 15.4]% vs [58.2 ± 17.7]%, P < 0.05).
CONCLUSIONA high rate of frozen-thawed microamount round spermatids of the mouse can be achieved by vitrification under the conditions of 7% glycerol and 30 min equilibrium.

3.CT Diagnostic Value and Clinical Analysis of the Liquid in the Renal Subcapsule(Analysis of 25 Cases)

Objective To evaluate the value of CT in the diagnoses and treatment of the liquid in the renal sub capsule.Methods The spiral CT images and clinic material of the liquid in the renal sub capsule were concluded in 25 cases.Results The total accuracy of CT showed 100% for the liquid in the renal sub capsule and reliable for the injury of liver and spleen,fracture of lumbar vertebra and abdominal accumulation of water.Conclusion CT diagnoses is the evidance to choose treatment for the liquid in the renal sub capsule with conservation,intervention,or operation.
4.Regimens for inoperable locally advanced non-small cell lung cancer

Concurrent chemoradiotherapy is the optimal regimen for patients with inoperable locally advanced non-small cell lung cancer (LA-NSCLC).The application of new technologies such as positron emission tomography (PET)/CT and four dimensional CT (4D-CT) enhances the accuracy of radiotherapy and decreases adverse reaction.Induction chemotherapy and consolidation chemotherapy do not show benefit to survival.The targeted therapy and immunotherapy have the potential of improving the outcomes of inoperable LA-NSCLC.
5.Influence of nebulized Pulmicort respules inhalation after endoscopic sinus surgery on expression of ENaC protein

Objective To investigate the influence of nebulized Pulmieort respules inhalation after endoscopic sinus surgery (ESS)on the expression of epithelial Na~+ channel(ENaC)protein in nasal mucosa. Methods Forty-four patients with nasal polyps undergoing ESS were randomly divided into Pulmieort respules treatment group(n=21,nebulized Pulmieort respules inhalation for 10 d after ESS)and Rhinocort control group(n=23,Rhinoeort aqueous nasal spray for 10 d after ESS).All the patients were performed biopsy of membrane on the residual middle turbinate 14 d after ESS,eosinophils (Eos)and neutrophils(Neu)per hundred inflammation cells were counted under microscope during ESS and after ESS,and the expression of ENaC protein was detected by immunofluorescence assay. Results The percentages of Eos and Neu decreased in two groups after treatment,and the percentage of Neu in Pulmieort respules treatment group was significantly lower than that in Rhinoeort control group(P<0.05).The expression of ENaC protein after treatment in Pulmieort respules treatment group was significantly lower than that in Rhinoeort control group(P<0.01). Conclusion Application of pulmieort respules after ESS can decrease Neu infiltration and inhibit expression of ENaC protein,which can relieve acute inflammation and edema of nasal mucosa.

7.Occurrence factor and preventive strategy for deep venous thrombosis following artificial hip replacement

OBJECTIVE: A series of studies has been reported concerning deep venous thrombosis following artificial hip replacement. This paper is aimed to summarize the occurrence factor and preventive measures for deep venous thrombosis following artificial hip replacement.METHODS: A computer-based online search of VIP database was undertaken by the first author to identify the articles about the deep venous thrombosis following artificial hip replacement published in between January 1994 and October 2009 with the key words of"artificial hip replacement and deep venous thrombosis". Inclusive criteria: ①Occurrence factor of deep venous thrombosis following artificial hip replacement. ②Diagnostic criteria and diagnostic methods of deep venous thrombosis following artificial hip replacement. ③Preventive strategy for deep venous thrombosis following artificial hip replacement. Inclusive criteria: repetitive research or obsolete documents. Totally 25 literatures were included in this paper.RESULTS: The agreement has been basically achieved for pathogenesis and risk factor of deep venous thrombosis following artificial hip replacement. The pathogenesis included blood hypercoagulable state, slow blood flow (or stasis), and vessel wall damage. Meanwhile, elderly, cerebrovascular disease, varicose vein or intravenous surgery, were the risk factors of deep venous thrombosis. Mechanical therapy or medication could be selected for preventing patients against deep venous thrombosis following artificial hip replacement with different features. It was an acceptable method for most patients using mechanical therapy, which could not induce drug adverse reaction, but the clinical efficacy remained uncertain for high-risk patient population, thus, medication should be combined. Low molecular heparin was considered first-choice drugs for preventing deep venous thrombosis following artificial hip replacement, which was characterized by common uses and reliable effects. CONCLUSION: Studies on influencing factor and preventive treatment of deep venous thrombosis following artificial hip replacement has arisen more attention in medical circles. The understanding of pathogenesis, correlation factors and preventive measures plays an important role in decreasing incidence deep venous thrombosis.
8.Dynamic measurement of CRP, IL-6 and Fib levels of unstable angina and its clinical significance

Objective To study the clinical significance of dynamic determination of serum CRP,IL-6 and fibrinogen(Fib) levels in unstable angina pectoris (UAP) patients.Methods 162 UAP patients were selected as UAP group(including 52 cases of low risk group,53 cases in medium risk group and 57 cases in high risk group),58 healthy people were selected as control group.The CRP,IL-6 and Fib contents were observed in UAP patients after cardiac events.Results At discharge,CRP,IL-6,Fib in UAP low risk group were (3.47 ±0.55)mg/L,(10.38 ± 1.57) ng/L,(1.64 ± 0.65) g/L,which were significantly lower than the admission contents [(6.50 ±0.87) mg/L,(14.35 ± 1.22) ng/L,(3.01 ± 0.28) g/L] (t =2.761,2.945,2.118,P =0.028,0.003,0.031).CRP,IL-6,Fib levels in the medium risk group were (6.87±0.75) mg/L,(14.35 ± 1.02) ng/L,(2.26 ± 0.33) g/L,which were significantly lower than the admission contents [(9.21±1.03) mg/L,(19.24±1.58) ng/L,(4.15 ±0.47) g/L] (t =2.985,3.157,3.548,P =0.003,0.001,0.000).CRP,IL-6,Fib levels in the high risk group were (8.71 ±0.84) mg/L,(17.37 ± 1.26) ng/L,(4.67 ±0.85) g/L,which were significantly lower than the admission contents [(12.48 ± 1.24) mg/L,(25.13 ± 1.84) ng/L,(7.28 ± 0.64) g/L] (t =3.472,4.018,5.097,P =0.000,0.000,0.000).Six months after hospital discharge,in CRP,IL-6,Fib elevated groups,the incidence rates of cardiac event were 40.20%,40.35% and 47.50%,which were significantly higher than those in normal group (5.0%,4.17%,14.29%,x2 =23.655,2.211,14.442,P=0.000,0.000,0.000).Conclusion CRP,IL-6 and Fib levels in UAP patients were significantly higher than those in healthy people,and with remission,contents would be lower,which was the index to reflect the severity of UAP,but its specificity is bad,need to use caution.
9.Promotion effects of nerve regeneration of chemically extracted acellular nerve allograft in vitro

[Objective]Electrophoretic analysis of rat chemically extracted acellular nerve allograft(CEAN) protein was performed to investigate the protein of CEAN.Dorsal root ganglia of chicken were cultured onto the surface of CEAN slice to observe the effect of CEAN structure on the fiber regeneration.[Method]The CEAN were perpared according to Sondell method and electrophoresed to observed whether the electrophoresis strip of 28-30kDa(Myelin Protein) exited or not.Dorsal root ganglia of chicken were cultured onto thc surface of CEAN slice and dyed with nerve fiber fluorescence to observe the orientation of nerve fiber growth on the surface of CEAN slice.[Result]Electrophoretic analysis of rat CEAN protein showed that the electrophoresis strip of 28-30 kDa totally disappeared.A large number of nerve fibers grew from DRG along the basement membrane of CEAN.[Conclusion]No myelin protein remained in the CEAN,while the basement membrane structure of CEAN can induce the nerve fiber to grow.
10.Biological characteristics of human umbilical cord Wharton’s jelly-derived mesenchymal stem cells and their differentiation into Schwann-like cells

[Objective] To investigate the biological characteristics of Wharton’[KG-38x]s jelly-derived mesenchymal stem cells (WJMSCs) and their differentiation into Schwann cell-like cells, and to provide a new cell seed source for nerve tissue engineering or cell therapy. [Methods]Umbilical arteries, veins and umbilical cord tunica externa were removed , and the remaining tissue (Wharton’[KG-38x]s jelly) was harvested.The umbilical cord was cut into small fragments and cultured with DMEM in orderto gather. Cultured WJMSCs were confirmed by the detection of MSC-specific cell-surface markers. WJMSCs were differentiated along a glial cell lineage using an established cocktail of growth factors. Immunocytochemical staining and Western blot were used to evaluate the characteristics of differentiated WJMSCs.[Results]WJMSCs were immunonegative for the haemopoietic cell-surface markers (CD34 and CD45) and neural stem cell marker (nestin), and immunopositive for MSC surface markers CD44, CD105, Stro-1 and vimentin. WJMSCs treated with a mixture of glial growth factors adopted a spindle-like morphology similar to Schwann cells. Immunocytochemical staining and Western blot analysis revealed that the treated cells expressed the glial markers p75 and glial fibrillary acidic protein indicative of differentiation.[Conclusion]WJMSCs can be differentiated into the cells that are Schwann-like in terms of morphologic features and phenotype and may be suitable as Schwann-cell substitutes for nerve repair in clinical application.
