1.Comparison of levobupivacaine and bupivacaine in spinal anesthesia during transurethral resection of prostate for elderly patients

Eighty elderly patients undergoing elective resection for the prostate were randomly divided into two groups, the levobupivacaine (observe group) and bupivacaine (control group) were used for spinal anesthesia, respectively. The level of sensory block and maintain time were T7±4 and (224 ±28) min in observed group, those in control group were T6±3 and (227 ±30) min, respectively ( both P> 0. 05). The mean arterial blood pressure of control group was (71 ±8) -(72 ±8) mm Hg (1 mm Hg = 0. 133 kPa) from 5 to 30 min after injection; meanwhile that of observed group was(75 ± 9) mm Hg (P < 0. 05). The incidences of hypotension and nausea were 10% (4/40) and 0% (0/40) respectively in observe group, and those were 30% (12/40) and 15% (6/40) respectively in control group (both P<0.05). The results indicate that levobupivacaine and bupivacaine have similar anesthetic effects, but levobupivacaine has fewer side effects and is more suitable for elderly patients.
2.POSTNATAL DEVELOPMENT OF S100B AND GFAP IN THE CENTRAL NERVOUS SYSTEM IN RATS

Objective To study the expressions of S100B and glial fibrillary acidic protein(GFAP) in the central nervous system of Sprague-Dawley rats during postnatal development. Methods Twenty-four male SD rats were divided equally into four groups according to different postnatal times:7-day group,14-day group,21-day group and adult group.Immunohistochemistry was used to investigate the expressions of S100B protein and GFAP in the brain and the spinal cord. Results The amount and density of S100B positive astrocytes decreased significantly in frontal cortex,hippocampus,striatum,substantia nigra and spinal cord during postnatal development.It seemed that the second through the third week after birth was a critical period for these changes.The amount and density of GFAP positive AST increased gradually in the brain,but it was the opposite in the spinal cord.Double-labeled immunofluorescence of S100B and GFAP in hippocampus CA1 area showed that S100B positive stains were evenly distributed in the pyramidal,polymorphic and molecular layers from the seventh till the twenty first day after birth,but apparently decreased in each layer especially in the molecular layer in adult hippocampus while the immunoreactivities of GFAP increased.The proportion of double labeled cells also increased with the aging and more of them were found in the pyramidal and polymorphic layers.Conclusion Different patterns of the expressions of S100B and GFAP exist during postnatal development.It may reflect the different roles of these proteins on the glial cell development.And it may also indicate that the expressions of S100B and GFAP are regulated by different mechanisms during the course of development,which reflects the differentiation of subpopulations of astrocytes during ontogenesis.

5.Analysis of diagnosis and therapy for seven cases of urinary tract endometriosis

Objective To investigate the method of clinical diagnosis and treatment for urinary tract endometriosis (UTE).Methods The clinical data of 7 patients with UTE were analyzed retrospectively.Mean age was 36.5-year old.Two cases were diagnosed by taking cystoscopic biopsy tissue,the others were confirmed by intraoperative and postoperative pathology.Under the adjuvant treatment of endocrine,2 patients were performed partial cystectomy and ureteral resection and end-to-end anastomosis in 4 patients,1 patient undertook ureteric reimplantation.Results Among 7 patients,2 patients had endometrial ectopic bladder,4 patients had endometrial ectopic ureter,1 patient had endometrisis of bladder and urter.After operation,no leak and recurrence occurred 2-3 years of follow-up.Conclusions UTE is rarely seen and usually escape the attention of the physicians,endocrine therapy with surgical resection is the primary method.Choice of surgical approach should be based on a variety of factors lesion location,scope,depth,bilateral renal function,etc.
7.Relationship between Change of Cerebral Hemodynamic and Brain Injury in Neonates

The cerebral blood flow of infant is effected by physiological and pathological factors.As the cerebrovascular autoregulation of neonates is poor,in pathological cases,especially when hypoxemia and hypercapnia impaired regulation of its own,lead to changes in cerebral blood flow,then resulting in severe brain injury.It has made enormous progress in the research on the changes of cerebral blood flow in newborns in recent years.In normal infants,cerebral blood flow velocities is positively correlated to gestational age and body weight,and increase gradually with day-age in the first week after birth.The cerebral blood flow of newborn with brain injury can present as insufficiency,over-perfusion or low speed high-resistance earlier and high speed low-resistance later.Different results may be related to the duration and severity of asphyxia,but all of those are signs of damage of self-regulatory function of cerebral blood flow.Cerebral hemodynamic change is the important pathogenesis mechanism of brain injury.
8.Expression of Dishevelled protein is correlated with the proliferation and invasion of glioma

Objective To investigate the correlation between Dishevelled protein expression and the proliferation and invasion of glioma cells.Methods 67 cases of brain glioma specimens were collected.The expression of Dishevelled protein was detected with immunohistochemical method.The immunoreactivity score (IRS) of Dishevelled protein,and proliferation index (PⅠ) and invasion index (Ⅱ) were measured and their correlations were analyzed.Results The positive rate of Dishevelled protein in glioma was 65.7 % (44/67).IRS,PⅠ and Ⅱ were 4.15±3.13,(30.93±17.92) %,(20.38±13.36) %,respectively.Both PⅠ and Ⅱ significantly increased with an increase in the pathological grade of brain glioma (P < 0.001).Furthermore,PⅠ and Ⅱ were significantly higher in the Dishevelled protein-positive group than those in the Dishevelled protein-negative group [(38.27±17.60) % vs (16.02±8.92) % of PⅠ and (30.03±13.81) % vs (10.63±4.41) % of Ⅱ,respectively,P < 0.001].PⅠ and Ⅱ of glioma cells were positively correlated with IRS of Dishevelled protein (r =0.940 between PⅠ and IRS,and r =0.953 between Ⅱ and IRS,respectively).Conclusion Dishevelled protein plays an important role in the proliferation and invasion of brain malignant glioma.
9.Construction of HIV-2 gag recombinant fowlpox virus and its immune reactivity in mice

Objective To provide experimental evidences for developing a safe and effective re- combinant fowlpox virus which can prevent the infection of HIV-2.Methods A fowlpox virus(FPV) transferring vector was constructed by inserting HIV-2 gag gene to the downstream of a synthetic complex promoter ATI-p7.5?20 of vector pUTA2.Transfection was then carried out,and recombi- nant FPV(rFPV)was screened by 5'-bromo-deoxyuridine(BrdU),genome PCR and western blot detection.Balb/c mice were immunized with rFPV by muscular injection.Anti-HIV-2 antibody, CD4~+ and CD8~+ T-cell count and specific target-killing activity of spleen CTL in immunized mice were analyzed by ELISA,FACS and LDH release assay,respectively.Results A transferring vector pA- gag was constructed and confirmed by amplifying a fragment of 766 bp from the rFPV genome.Mean- while,HIV-2 multi-antibody-specific protein blot(55 000)was detected from the recombinant virus and the HIV-2 specific antibody was detected from the immunized Balb/c mice.HIV-2 specific target- killing activity of spleen CTL was observed in immunized mice.Conclusion A recombinant fowlpox virus expressing HIV-2 structural protein Gag has been obtained,and it can stimulate HIV-2-specific eelluar and humoral immune reactions in mice.
10.Establishment of model of diabetes and lens posterior capsule opacification induced by alloxan in rabbit

Background The establishment of diabetic animal model is a crucial step for the study about diabetic eye diseases. At present,the main modeling method include the injection of streptozotocin and alloxan. But the shortcoming of the former is an expensive price, and that of the later is high death rate of animals. Objective This experiment was to discuss the way which decrease the death of alloxan-injected animal and explore the effects of high blood glucose on the posterior capsular opacification (PCO). Methods Forty clean healthy male New Zealand white rabbits were randomly divided into 2 groups. 90mg/kg of alloxan were injected via ear vein once in 20 rabbits to create the diabetic animal models,and the equivalent amount of normal saline solution was injected at the same way as normal blood glucose group. The successful models were selected in the animals with the blood glucose level over 12. 0 mmol/L two weeks later, and PCO of lens were graded based on the method of Odrieh under the slit lamp. Extracapsular lens extraction was then performed on the right eye of rabbits in both groups, and the posterior capsules were obtained from these eyes at the 6th, 10th and 14th days after operation. The expression of proliferating cell nuclear antigen ( PCNA ) in posterior capsular lens epithelial cell was detected by immunohistochemistry. Results The modeling successful rate was 70% after injection of alloxan. The body weight of rabbits in high blood glucose group was significantly lowed and the blood glucose was significantly elevated in comparison with normal blood glucose group ( all P<0. 05). Two weeks after surgery ,2 eyes occurred 2 grade of PCO and only one eye showed the 1 grade of PCO in the high blood glucose group. However, 1 grade of PCO was found in 3 eyes in the normal blood glucose group. Biopsy revealed that PCNA was positively expressed in the cell nuclei of LECs in high blood glucose group rather than the normal blood glucose group from the 10th day after surgery. The proliferation index of PCNA was 0. 86±0. 04 and 0. 25±0. 03 respectively in high blood glucose group and normal blood glucose group, showing a significant difference between them (t = -16. 171 ,P = 0. 000). Conclusion Stable diabetic models of rabbits can be created by intravenous injection of 90 mg/kg alloxan. High blood glucose level is one of the important factors for the development of PCO.
