Objective: To investigate the effect of miR-144-3p modulating proliferation, migration and apoptosis of liver cancer Huh-7 cells through blocking frizzled class receptor 4 (FZD4)/Wnt/β-catenin pathway and the possible mechanism. Methods: A total of 18 pairs of cancer tissues and corresponding para-cancerous tissues from liver cancer patients, who underwent surgery in Workers' Hospital of Liuzhou City from March 2012 to July 2017, were collected for this study; in addition, hepatic cancer cell lines (Huh-7, SMMC7721 and MHCC97) and human normal liver epithelial cell line THLE-3 were also collected. The expression of miR-144-3p in liver cancer tissues and cell lines was detected by qPCR. MiR-144-3p mimics/inhibitor and FZD4 siRNA were transfected into liver cancer Huh-7 cells; the proliferation, migration and apoptosis of Huh-7 cells were evaluated by CCK-8 assay, Transwell assay, wound healing assay and Annexin V-FITC/PI double staining flow cytometry assay, respectively. The interaction between miR-144-3p and FZD4 was verified by dual-luciferase reporter gene assay. Results: The expression of miR-144-3p was down-regulated in liver cancer tissues and cell lines (P<0.05 or P<0.01). Over-expression of miR-144-3p significantly inhibited cell proliferation viability, migration but induced apoptosis of Huh-7 cells (all P<0.01). Moreover, dual-luciferase reporter gene assay showed that miR-144-3p directly interacted with FZD4 and suppressed its expression. Furthermore, in vitro experiments verified that miR-144-3p targeted FZD4 to suppress the proliferation, migration and promote apoptosis of Huh-7 cells via blocking Wnt/β-catenin pathway (all P<0.01). Conclusion: miR-144-3p inhibits malignant biological behaviors of liver cancer Huh-7 cells via blocking Wnt/FZD4/β-catenin signaling pathway, which may provide potential molecular targets for early diagnosis or treatment of liver cancer.

Chinese Journal of Internal Medicine 2013;(7):574-577
Objective To investigate whether unfolded protein response (UPR) plays a role in the pathogenesis of spondyloarthritis (SpA),and to assess UPR-related gene expression in SpA and other arthritis patients. Methods Eighteen patients with SpA,12 with rheumatoid arthritis (RA) and 6 with osteoarthritis (OA) were recruited.Macrophages were isolated from synovial fluid samples by immunomagnetic separation. The expression of UPR-regulated genes, including binding immunoglobulin protein (BiP),glucose-regulated protein 94 (GRP94),C/EBP homologous protein (CHOP),growth arrested and DNA damage-inducible 34 (GADD34),X-box binding protein 1 (XBP-1) and endoplasmic reticulum DnaJ homolog 4 (ERdj4),was tested by real time polymerase chain reaction (PCR).Results Compared with macrophages in OA patients, the expression of BiP and GRP94 mRNA [(6.06 ± 2.08) × 10-2 vs (1.11 ±0.72) × 10-2 for BiP mRNA,11.80(7.30-38.40) × 10-3 vs 1.27(1.02-4.18) × 10-3 for GRP94 mRNA,both P values <0.01] was significantly increased in macrophages in SpA patients.XBP1 mRNA was up-regulated [(12.70 ± 5.20) × 10-3 vs (4.14 ± 2.56) × 10-3,p < 0.01] in SpA group as well.UPR-regulated gene expression in SpA patients with HLA-B27 positive or HLA-B27 negative was similar. However, none of UPR-regulated genes showed different expression between the SpA group and RA group except for GADD34 mRNA [7.30 (5.56-15.40) × 10-3 vs 21.30 (12.20-27.60) × 10-3,P =0.009].Conclusions Our data suggest that UPR possibly participates in the pathogenesis of SpA,although the relationship between HLA-B27 and UPR still needs further investigation.

Journal of Endocrine Surgery 2012;06(5):301-306
ObjectiveThe number of positve axillary lymph nodes(ALNs) is one of the most important factors affecting prognosis of patients with breast cancer. The resected lymph nodes number( LNN)can affect the number of positive ALNs.In this study,we aim to evaluate the influence of LNN on the progonosis of breast cancer patients with positive ALNs.Methods449 breast cancer patients with positive ALNs who underwent total mastectomy and axillary dissection were divided into 2 groups:group A (LNN < 10)and group B (LNN ≥ 10).The clinicopathological features of the 2 groups were compared.Kaplan-Meier method and Cox proportional hazard method were respectively used to make univariate and multivariate survival analysis for disease-free survival (DFS).ResultsThe median follow-up was 43.59 months for group A and 41.34 months for group B.Group A had a significantly higher proportion of patients in pN1,with lymph node metastasis and adjuvant radiotherapy than group B.Univariate analysis showed pN stage,LNN,lymph nodes metastasis rate,HER-2,adjuvant chemotherapy regime and adjuvant radiotherapy were factors influencing DFS. Multivariate analysis showed that pN stage,LNN,lymph nodes metastasis rate, adjuvant chemotherapy regime and adjuvant radiotherapy were factors influencing DFS.We performed analysis stratified by adjuvant chemotherapy regime containing taxan and adjuvant radiotherapy.Stratified analysis showed patients with adjuvant chemotherapy regime containing taxus and adjuvant radiotherapy showed significantly higher DFS rate than those without taxus in adjuvant chemotherapy regime or without adjuvant radiotherapy in group A.However, the 2 different treatment regime had no difference in DFS rate for patients in group B.ConclusionsThe decrease of LNN may affect the diagnosis and therapeutic selection of breast cancer patients with positive ALNs.LNN is an inexpensive and easily available factor for predicting DFS of breast cancer patientswithpositiveALNs.Adjuvant chemotherapy regime containing taxus and adjuvant radiotherapy can improve DFS rate of patients with LNN < 10.
