1.Determination of Total Polysaccharides in Weile Granules by Phenol-sulfuric Acid Method

OBJECTIVE:To establish phenol-sulfuric acid method for the determination of the content of total polysaccharides in Weile granules.METHODS:The samples were deproteinized by Sevag method,colored by phenol-sulfuric acid method(water bath at 60 ℃ for 30 min),and the content of total polysaccharides in Weile granules was determined with Glucose as control.RESULTS:The linear range of Glucose was 20.47~102.36 ?g?mL-1(r=0.999 4),with an average recovery of 99.09%(RSD=0.92%,n=6).The content of total polysaccharides in Weile granules was 41.63%.CONCLUSION:This method was simple,accurate and reproducible,and suitable for quality control of Weile granules.
2.Effect of open storage and peeling on the fluorine content of dried corn in Zhaotong city, Yunnan province, a fluorosis area

Objective To explore the effect of open storage and peeling on the fluorine content of dried com in order to provide a scientific basis for prevention and treatment of coal-combustion fluorosis. Methods In October 2006, 34 samples of freshly dried and 3-month old dried com were sampled in Zhenxiong county, Zhaotong city, Yunnan province. Fluorine content in corn was determined by combustion-hydrolysis-ion selective electrode method. Results The average fluorine content of freshly dried corn by coal-combustion was (4.29 ±1.87)mg/kg,and increased to (6.64±2.64)mg/kg after they had been openly stored for three months and the average content increased by 54.74%. But if corn was peeled off, their fluorine content decreased to (5.03±2.04)mg/kg at a rate of 24.25%. The amount of fluorine absorbed by the skin of corn was more than other parts of com during corn desiccation. Conclusions The inhabitants in coal-combustion fluorosis areas ingest less fluorine through dry com if they eat the peeled corn and take measures to keep it airtight and moisture-proof during storage thus reducing the harm of coal-combustion fluorosis.
3.Chronic toxicity of methylamine on cardiovascular endothelium of rabbits

AIM To study whether chronic administration of methylamine may induce elevation of semicarbazide-sensitive amine oxidase (SSAO) activity and initiate the injury of cardiovascular endothelium. METHODS New Zealand rabbits were treated with methylamine hydrochloride (100 mg·kg-1) by ig, once a day for 6 months. The rabbits were weighed every other week and the dosage was adjusted according to the body weight. The number of circulating endothelial cells (CEC) in the arterial blood, nitric oxide (NO) concentration in the serum and ultrastructure of endothelial cells of aorta were assessed. The plasma SSAO activity and formaldehyde concentration were assessed by liquid chromatography. RESULTS The number of CEC, NO concentration, levels of SSAO activity and formaldehyde concentration in the methylamine group were increased significantly, compared with the control group. Ultrastructure of endothelial cells in the methylamine group showed inordinate morphological changes (multiple intranuclear inclusions, karyopyknosis and karyorrhexis). CONCLUSIONChronic administration of methylamine can induce the elevation of SSAO activity and initiate the injury of cardiovascular endothelium.
4.ffect of basic fibroblast growth factor on rabbit lens epithelial cell proliferation

Objective To study the effect of basic fibroblast growth factor (bFGF) on proliferating of rabbit lens epithelial cells(RLECs).Methods The second and three generations of RLECs were exposed to different concentrations of bFGF, and the proliferation characteristics of the cells were measured with methyl thiazolyl tetrazolium (MTT) assay. The ultrastructure of cell were observed with transmission electron microscope (TEM).The changs of cells cycles were observed with flow cytometer (FCM).Results After the treatment of bFGF, RLECs showed the marked proliferation , especially in 10μg*L-1 of bFGF . TEM result showed that the cells were more active with bFGF, FCM result showed that the S phase cell obviously increased.Conclusion bFGF is an important factor that can promote proliferation of RLECs.
5.Exploration of online learning mode for refresher doctors prehospital training and evaluation of learning effect in Peking Union Medical College Hospital

Objective Carry out the reform of the online learning mode of prehospital training for refresher doctors , and evaluate the effect of reform in Peking Union Medical College Hospital .Methods An exploration was carried out on the refresher doctors prehospital training mode from traditional classroom teaching to online learning , Estab-lished the online learning system .Evaluated the effect of online learning by the way of questionnaire investigation andreexaminationamong289refresherdoctors.Results 289refresherdoctorshavecompletedthestructuredon-line courses of prehospital training , and the average pass rate of the first test was 42.2%.The questionnaire and retest was conducted , A total of 262 doctors completed the work , The recovery rate was 90.7%, and the average pass rate of retest was 74.9%, showing improvement by 32.7%.Conclusions Online learning mode is an effec-tive tool for knowledge dissemination , and has a better learning effect .
6.Effect of deamination reaction mediated by semicarbazide-sensitive amine oxidase on 3 T3-L1 adipocytes

[ ABSTRACT] AIM:To observe the effect of the metabolites generated from oxidative deamination of methylamine ( MA) or benzylamine ( BZA ) catalyzed by semicarbazide-sensitive amine oxidase ( SSAO ) on 3T3-L1 adipocytes. METHODS:3T3-L1 preadipocytes were induced to differentiation.SSAO activity was determined by high performance liquid chromatography ( HPLC) at different differentiation time points.MTT assay was applied to detect cell vitality after exposure to different concentrations of MA or BZA.Fluorescence probe DCFH-DA was used to determine the production of reactive oxygen species after incubation of 3T3-L1 adipocytes with MA or BZA.After exposure to 0.5 mmol/L MA or BZA for 4 h, malondialdehyde ( MDA) , total superoxide dismutase ( T-SOD) and glutathione ( GSH) in the adipocytes or prea-dipocytes were measured.RESULTS:SSAO activity increased with the increase in the differentiation days, and reached a maximum at the 8th day.Incubation of the cells with different concentrations of MA or BZA for 4 h did not significantly de-creased the cell vitality (P>0.05).After exposure to 0.5 mmol/L MA or BZA, the reactive oxygen species in adipocytes significantly increased, and were about 3 to 4 times as compared with control group (P<0.05).After treatment with 0.5 mmol/L MA or BZA for 4 h, MDA content significantly increased, while the activity of SOD and the expression of GSH de-creased in mature adipocytes compared with control group (P<0.05).However, MDA, T-SOD and GSH did not change significantly after treatment with equal molar of MA or BZA in the preadipocytes ( P>0.05 ) .CONCLUSION: MA or BZA induces oxidative stress in the mature adipocytes, which might result from the deamination products catalyzed by SSAO.
7.Methodological issues and suggestions for improvement in randomized controlled trials of Chinese herbal medicine for recurrent miscarriage.

Recurrent miscarriage is a common disease in clinical obstetrics and gynecology. There is no curative treatment for recurrent miscarriage in conventional medicine. Traditional Chinese medicine (TCM) has been widely used in the treatment of recurrent miscarriage in China for thousands of years. However, randomized controlled trials (RCTs) with high quality evaluating the effectiveness and safety of Chinese herbal medicine (CHM) still lack. This study was conducted in order to identify methodological problems in published or unpublished RCTs of CHM in the treatment of recurrent miscarriage, and provide suggestions for TCM researchers to conduct high-quality clinical research.
8.Intratumoral relative perfusion rate and the relation between MVD and expression of VEGF assessed with power Doppler imaging: in bladder carcinoma

Objective To evaluate the relationship between relative perfusion rate assessed with power Doppler imaging (PDI) and microvessel density (MVD), as well as expression of vascular endothelial growth factor (VEGF) in bladder carcinoma. Methods The transrectal ultrasound (TRUS) blood flow signal of bladder carcinoma was preoperatively detected with color Doppler imaging in 45 patients, and the relative perfusion rates were obtained for analysis. MVD and the expression of VEGF of excised tumor were assessed immunohistochemically. Results There were correlation between relative perfusion rate and MVD, the expression of VEGF. MVD and the expression of VEGF were related to pathologic grade and the invasiveness of tumor tissues. The expression of VEGF in bladder carcinoma was positively correlated to the tumor interstitial vascular density. Conclusion Combination of PDI and immunohistochemical parameters is useful for evaluating the angiogensis of bladder carcinoma from a different point of view.
9.Comparison of the clinical effect of phacoemulsification and middle incision extracapsular cataract extraction

To compare the clinical effect of phacoemulsification and middle incision extracapsular cataract extraction ( Ml-ECCE) . METHODS: One hundred and eighty - five eyes of phacoemulsification (137 cases) and 185 eyes of 139 cases for Ml-ECCE from January 2011 to May 2013 were involved in this study. And the ratio posterior capsular rupture during surgery, visual acuity, corneal edema, corneal astigmatism and intraocular pressure post operation were followed up.RESULTS: On 1d after surgery, uncorrected visual acuity in the group of Ml-ECCE was better than that of phacoemulsification group, while from 3d; 1 and 3mo after surgery, no significant difference was found from the above two groups. On 1d postoperation, corneal edema ratio in phacoemulsification group ( 45 eyes ) was higher than that in Ml-ECCE group(20 eyes) ( X2=11. 665, P=0. 0006 ) . No significant difference was found for the ratio of posterior capsule rupture during surgery in these two surgical technique groups(X2=0. 094,P=0. 759). On 1wk;1 and 3mo after surgery, significant difference was found for the average of surgical induced corneal astigmatism between two groups (u=6. 661, 6. 880, 4. 187, P = 0. 00, respectively ). During following up, no significant difference was found for the intraocular pressure between two groups ( u=1. 858, 0. 963, 0. 471, 1. 349, 1. 388; P= 0. 063, 0. 335, 0. 638, 0. 177, 0. 165). lntraocular pressure on 1d postoperation in phacoemulsification and Ml-ECCE groups was higher than before operation ( u = 19. 86, 19. 39, P = 0. 00, respectively). And on 1wk; 1 and 3mo postoperation, intraocular pressure in the operated eyes in both groups was lower than before operation for 2~3mmHg.CONCLUSlON: Although phacoemulsification and Ml-ECCE could both get good visual rehabilitation, with similar visual outcome, no significant effect for intraocular pressure, and no severe complications, the latter one owns the advantage that easier maneuver, quicker recovery, and cheaper instruments needed, which is suitable for the hard nuclei cataract in local hospitals. Ml-ECCE is a safe, effective and easy manipulation for local hospital for large batch of cataract surgeries.
