1.Simultaneous Determination of 3 Ingredients in Shilintong Tablet by HPLC

OBJECTIVE:To establish a method for simultaneous determination of vicexin 2,vicexin 1 and schaftoside in Shilin-tong tablet. METHODS:HPLC was performed on the column of Phenomenex C18 with mobile phase of methanol-0.1% phosphoric acid (gradient elution)at the flow rate of 0.8 ml/min,the detection wavelength was 334 nm,the column temperature was 25 ℃, and the injection volume was 10 μl. RESULTS:The linear range was 0.31-3.10 μg for vicexin 2 (r=0.9999),0.28-2.80 μg for vicexin 1(r=0.9999)and 0.39-3.90 μg for schaftoside(r=0.9999),respectively;RSDs of precision,stability and reproducibili-ty tests were lower than 3.0%;recoveries were 95.2%-100.4%(RSD=1.6%,n=9),96.3%-100.8%(RSD=1.4%,n=9) and 95.7%-100.9%(RSD=1.7%,n=9),respectively. CONCLUSIONS:The method is simple,sensitive ,accurate,reliable and repro-ducible,and can be used for the quality control of Shilintong tablet.
2.Simultaneous Determination of 5 Active Ingredients in Begonia Grandis Dry from Different Habitats by UP-LC

Objective:To establish a method for the determination of protocatechuic acid, protocatechuic, catechin, epicatechin and rutin in Begonia grandis Dry. Methods:The 5 active ingredients in the samples were extracted by 70% methanol and analyzed by ultra performance liquid chromatography (UPLC). Chromatographic separation was performed on a Waters ACQUITY HSS T3(2. 1 mm × 100 mm, 1. 8 μm) column with gradient elution, and the mobile phase was solvent A: acetonitrile and solvent B:0. 2% glacial acetic acid (0-12 min:A:5→25 and B:95→75;12-30 min:A:25→50 and B:75→45). The effluent was monitored by a VWD detector set at 280 nm. The flow rate was set at 0. 2ml/min and the injection volume was 1μl. The column temperature was maintained at 30℃. Results:The calibration curves were all linear (r≥0. 99) over the test ranges. The average recovery was 96. 92%-99. 75%(n=6). Conclusion:The established method can rapidly obtain an accurate and reproducible result, which can be used to control the quality of Begonia grandis Dry.
3.Deepen Clinical Teaching Reformation Under the Employment Oriented Guidance

College students' employment viewpoint has transferred ultimately from traditional"government-planning assignment" to modern job-seeking mechanism of "two way choice and independent option".Recently,medical college has reformed clinical teaching in 5-years program.But some problems still exist in clinical teaching reflected from the employment situation of medical graduates and the evaluation of recruiting employers.Some work can be done to solve problems existing in medical students' employment.It is an important issue in adapting social requirement through reforming the model of clinical teaching and arranging the period of clinical training reasonably.

5.Study on GFAP changes of astrocyte and expression of endothelin-1 in brainstem injury of human being

Objective To study GFAP changes of astrocyte and expression of Endothelin-1 in molecular pathology in brainstem injury of human being and explore its forensic medical significance. Method 15 brain specimens of acute brainstem injury and 5 brain specimens of no brainstem injury in different time were selected and their microscopic morphological changes were observed with HE staining, and we study the pathological changes of GFAP of astrocyte and expression of Endothelin-1 in monoclonal antibody im-munohistochemistry SP method. Results The number of GFAP positive cells was increased, the bulk of the cell became bigger and expression of Endothelin-1 could be observed. Conclusion The above pathological changes might be a valuable base used for postmortem diagnosis of acute brainstem injury.
6.Analysis of Clinical Control of Type 2 Diabetes in 7356 Outpatients

Objective To study the current status of clinical control of outpatients with type 2 diabetes mellitus,and to discuss the factors influencing on disease control. Methods According to the clinical targets recommended by Asian-Pacific Type 2 Diabetes Policy Group,we calculated the success rates of blood glucose,lipid profile,blood pressure and body mass index(BMI) of patients who hospitalized in our department for more than one year(group A,n=7 356), and compared these results with the corresponding rates calculated based on the earlier database(group B,n=1 084).The success rates of metabolic indexes of patients who insisted on hospitalizing monthly(group C,(n=1 367))were calculated.Besides,the factors influencing disease control were discussed through questionnaire.(Results)There were 7356 patients in group A,and the success rates of HbA1c,blood pressure,blood lipid profile and BMI were 30%,25.8%,32.6% ~ 81.3% and 50.9%,respectively.Except for the parameter of high-density lipoprotein,the other rates were higher than the responding ones of group B.There were 1367 patients in group C, and the success rates of main metabolic indexes were higher than those of group A and B.The result of the questionnaire indicated that poor understanding of diatetes,lack of health education,and relative scarcity of diplomate were the main factors that influenced the disease control. Conclusion The disease control of most of the type 2 patients with diabetes are far from satisfaction.The application of scientific disease care system is effective for improving the disease control.
7.Exploring of the Diabetes Care Model

Objective To explore the new diabetes care model. Methods A new flow for diabetic outpatient to visit the doctor, a uniform system for diagnosis,treatment and follow-up,an electronic medical record system,and a long-term diabetes education which mainly delighted patients on "diabetic self-management" were established.(Results)The service quality from the medical staff were improved markedly.Database including the clinical data of 10 020 patients and 2 563 serum samples and DNA samples were built up.A set of diabetes care model that laid the foundation for clinical and research work was formed. Conclusion Long-term education for patients with diabetes mellitus constitues the basic and important part of diabetic care model.The application of computer in diabetic care is the effective means.It is necessary to establish a uniform scheme for diagnosis,treatment and follow-up for the patients,and the improvement of medical staff's professional making ensures the successful implementation of the model.The combination of clinical practice and research work propels the development of scientific management model for patients with diabetes mellitus.
8.Expression and significance of Slit2 in different stages after the spinal cord injury of rats

Objective To investigate the expression and significance of Slit2 in different time point after the spinal cord injury of rats.MethodSeventy adult wistar rats were randomly divided into three groups: spinal cord injury by fully transection on T10 level spinal cord (Group A); laminectomy and the spinal cord uninjuried (Group B); natural without operation (Group C).Then the rats were sacrificed and the spinal cord was taken out fresh quickly on different time-point(12h, 1, 3, 5, 7d after operation). The tissues perfusion by formaldehyde were taken out on 3, 5, 7, 14d after operation. The expressions of Slit2 were tested by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemical measurement. By means of above,the author investigated the expressions and location of Slit2 after the spinal cord injury.All data were statisticsed by SPSS 11.5 software.ResultThe expression of Slit2 mRNA of appeared in the spinal cord tissue 12 hours after injury, reached peak on the 3rd day, declining gradually later. The positive expression of Slit2 located in the cytoplast on oligodendrocyte and astrocyte.The positive cells were found at 3d after spinal cord injury, reached peak on 7d after injury, declining after 14d. The change of Slit2 was correlative with the rehabilitation and regeneration of the axon on the forepart period.ConclusionAs an important factor in axonal growth-guidance,the author crewed that Slit2 may be participated in the regeneration and rehabilitation of axons after the spinal cord injury.
