1.Effect of ketamine on the cGMP level in different brain areas in a rat model of CFA-induced arthritis

Objective To investigate the effect of ketamine on the cGMP content in cerebral cortex, hypothalamus and brain stem in a rat model of CFA-induced arthritis. Methods Twenty-four SD rats weighing 200-220 g were randomly divided into three groups with 8 animals in each group, group Ⅰ: normal rat + intraperitoneal normal saline (ip NS);group Ⅱ : arthritic rat+ ip NS;group Ⅲ: arthritic rat + ip ketamine 100mg?kg-1 . Arthritis was produced by injecting CFA 0.05 ml into left ankle joint which developed inflammatory response within 24h and became hyperalgesic. The hyperalgesia persisted for more than 6 weeks. Three days after the arthritis model was established, paw withdrawal latency (PWL) to radiant heat was measured. 15 min after ip ketamine the animals were decapitated and cerebral cortex, hypothalamus and brain stem were separately isolated for determination of cGMP content using radioimmunoassay. 2.Different inhibition of hepatocarcinoma cell growth by As_2O_3 in SMMC-7721 and BEL-7402 cell lines

AIM: To explore the different inhibitory effect of arsenic trioxide (As_2O_3) on hepatocarcinoma cell growth in SMMC-7721 and BEL-7402 cell lines and its mechanism. METHODS: The cell culture and trypan blue staining were used to study the inhibitory effect of arsenic trioxide on cell growth, and the glutathione (GSH) contents in hepatocarcinoma cells treated with arsenic trioxide were detected. RESULTS: Arsenic trioxide inhibited the growth of BEL-7402 cells in a time and dose-dependent manner. The inhibitory effect was significant at a lower dose of 0.50 ?mol/L for 24 h, however, to SMMC-7721 cells, a higher dose of 2.00 ?mol/L for 96 h was needed. The inhibitory rate of arsenic trioxide (0.25-2.00 ?mol/L) on BEL-7402 cell growth was higher than that on SMMC-7721 cells. The content of GSH in SMMC-7721 cells was much higher than that in BEL-7402 cells [(50.8?5.2) (?mol/g) protein and (18.7?1.4) ?mol/g protein, respectively]. CONCLUSION: There was a significant difference in inhibition of hepatocarcinoma cell growth by arsenic trioxide between BEL-7402 and SMMC-7721 cell lines, the cause of which may be due to the difference in GSH content in BEL-7402 and SMMC-7721 cells. 3.Effect of propofol on hypoxic pulmonary vasoconstriction in rats

Objective To investigate the effect of propofol on hypoxic pulmonary vasoconstriction(HPV)and assess the underlying mechanism in rats.Methods Twenty male SD rats weighing 300-400 g were.anesthetizedwith intraperitoneal phenobarbital.Heart and lungs were removed after thoracotomy.Pulmonary.arterial rings 4 mmin length and 1.0-1.4 mm in diameter were prepared and suspended in Earl solution maintained at 37℃ with a pHof 7.40 and aerated with 20% O_2-5% CO_2-25% N_2.The rings were stimulated with phenylephrine(PE)10~(-6)mol?L~(-1) with different preloads(300,500,700,900,1 100 mg).The isometric tension of the arterial rings wasmeasured.The optimal preload was determined to be 900 mg which allowed best contractility.Hypoxia was inducedby aerating the solution with 95% N_2-5% CO_2 and the flow rate was adjusted to maintain PO_2 of the solution at40-20 mm Hg and pH at 7.40.HPV of the rings were recorded.Then propofol was added to achieve a finalconcentration of 1,3,10,30,100 ?mol?L~(-1) and HPV was again induced and the changes in HPV wererecorded.In addition the effects of propofol(10,30 ?mol?L~(-1))on vasoconstriction produced by KCI and PE werealso measured.Results The lower doses of propofol(10 ?mol?L~(-1))significantly inhibited HPV(P

5.Estrogen receptor ? (ER?) gene polymorphism of perimenopausal syndrome patients with liver qi stagnation syndrome (LQSS)

Objective:To investigate the association between polymorphisms of estrogen receptor ? (ER?) gene (rs3020444) and perimenopausal syndrome (PS) in patients with liver qi stagnation syndrome (LQSS). Metheds:The ER?T/C genotype in 100 patients of PS of LQSS type (PS-LQSS group),86 patients of PS of non-LSDS type (PS-non-LQSS group) and 100 healthy subjects (control) was determined by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) assay. Results:Partitions of ?2 method showed that the higher prevalence of ER?-TT genotypes was noted in PS-LQSS group (?2=8.307,P=0.004),and the higher prevalence of T alleles was noted in PS-LQSS group (?2=7.129,P=0.008). and Multinomial logistic-regression indicated that the odds ratios (OR) of the ER?-TT genotypes vs TC/CC for PS-LQSS was 2.222 (95%CI:1.172-4.744,P=0.015) after adjusting for common miscellaneous factors. Conclusions:It was suggested that ER?-TT genotypes was significantly associated with PS-LQSS,and ESR?-TT may be one of the genes that contribute to PS-LQSS.

7.Application of sharing mode of nursing observation on typical cases in clinical work

Objective To apply the sharing mode of nursing observation on typical cases to promote nurses' capability and observation skills of clinical nursing and deepen the connotation of high-quality nursing service.Methods The sharing mode of n ursing observation on typical cases was established in 2010 and run through the process of record-report-evaluate-share-reward.The registration form of nursing observation on typical cases was designed by the nursing department,on which the observers recorded typical cases and reported it via network to the nursing department.And the latter assigned nursing experts at regular intervals to evaluate and shared cases through a variety of forms (publishing briefings,organizing exchange meetings,etc.),in which the excellent examples were rewarded.Results Since 2010,298 cases had been submitted,with more than 100 cases shared and 57 rewarded.Conclusions The sharing mode of nursing observation on typical cases has elevated nurses' observation capability and comprehensive quality,deepened the connotation of high-quality nursing service and ensured the safety of the patients.

Arbuscular mycorrhizal fungi can infect the roots of Cercidiphyllaceae and form arbuscular mycorrhizae.Infection rate is from 12% to 30%,but infection strength is weak.4 species of Acaulospora and 11 species of Glomus were isolated and identified,including Acaulospora tuberculata,A.spinosa,A.foveata,A.denticulate,Glomus geosporum,G.clarum,G.constrictum,G.monosporum,G.laroideum,G.versifome,G.microcarpum,G.mosseae,G.hoi,G.halonatum and G.reticulatum.Among them,G.cercidiphyllorum is advantage species.So AMF may be a potent resource of biology which can stimulate the growth of Panax notoginseng.
9.Primary study of real time elastosonography in estimating the characteristic of nodes in resected hepatocirrhosis specimens

Objective To evaluate the feasibility of real time elastosonography in estimating the characteristic of nodules in resected hepatocirrhosis specimens.Methods Thirty-eight reseeted hepatocirrhosis specimens underwent elastosonography.The nodules that have drawn attention were performed elastosonography through rhythmic pressing and releasing the probe by manual form on the liver.The hepatic strain on the region of interest was shown by chromatic scale.To compare nodules rigidity with surrounding hepatic tissues, hepatocirrhosis nodules were classified into hard nodules, medium rigidity nodules, mixture of hard and soft nodules, and soft nodules.All nodules were confirmed by pathology.Results Forty-four nodules of 38 hepatocirrhosis exemplar received real time elastosonography.Of 44 nodules, hard nodules were 18,of which 12 (66.7%) were hepatoeellular carcinomas, 2 (11.1%) were dysplasia nodules, 4 (22.2%) were regenerative nodules.Medium rigidity nodules were 7,all were regenerative nodules.Mixture of hard and soft nodules were 11, of which 8 (72.7%) were hepatocellular carcinomas, including 4 accompanied necrotic tissue, 1 (9.1%) was dysplasia nodules accompanied necrotic tissue, the other 2 (18.2%) were regenerative nodules accompanied necrotic tissue.And soft nodules were 8, of which 4(50.0%) were necrotic nodules, 1 (12.5%) was dysplasia nodules accompanied canceration, 1 (12.5%) was hepatocellular carcinoma,2(25.0 %) were regenerative nodules.Conclusions Real time elastosonography can effectively evaluate the comparative rigidity on hepatoeirrhosis nodules,and thus may have potential usefulness on estimating the characteristic of hepatocirrhosis nodules.
10.Relationship between proportions of peripheral natural kill cells and dendritic cells and liver injury in patients with chronic hepatitis B virus infection

Objective · To investigate the correlation between proportion of natural killer (NK) cells and dendritic cells (DCs) and liver injury in patients with chronic hepatitis B virus (HBV) infection. Methods · Peripheral blood samples were collected from 65 patients with chronic HBV infection and 10 healthy volunteers (HCs group). Flow cytometry was used to detect the proportion of peripheral NK cells and DCs. The patient were divided into asymptomatic HBV carrier (AsCs) group, chronic hepatitis B in low or moderate grade (CHB-LM) group, and chronic severe hepatitis B (CSHB)group.The relationship of the proportion of NK cells, the proportion of DCs, and the ratio of NK/DC with the degree of liver injury was analyzed. Results · The proportion of peripheral NK cells was significantly lower in CHB-LM group (9.29%±1.20%) and CSHB group (3.60%±0.79%) than that in HCs group (14.77%±2.90%) and AsCs group (14.05%±2.32%) (P=0.000). And the proportion of peripheral DCs was significantly higher in CHB-LM group (4.10%±1.18%) than that in HCs (1.67%±0.31%), AsCs (2.05%±0.46%) and CSHB groups (2.08%±0.41%) (P=0.000). The ratio of NK/DC was decreased in CSHB and CHB-LM groups compared to that in HCs and AsCs groups (P=0.000). The correlation analysis indicated the proportion of NK cells was negatively correlated with the levels of ALT, AST, TB and DB (r=-0.50, r=-0.49, r=-0.53, r=-0.51, P=0.000 for all) in patients with chronic HBV infection. Conclusion · The proportion of peripheral NK cells and the ratio of NK/DC in patients with chronic hepatitis B were decreased. The decrease of the frequency of NK cells and the ratio of NK/DC were associated with liver injury in patients with chronic HBV infection.
