Objective: To investigate the therapeutic effects of ets-1 antisense oligoxydeonucleotide(AsODN) on gastric carcinoma. Methods: Cultured SGC-7901 cells were devided into control group, sense oligoxydeonucleotide(sODN) group and AsODN group. After transfection for 24 h, expression of ets-1mRNA was detected by RT-PCR, growth inhibition was detected by colone formation assay, in vitro invasive ability assay was carried out in Transwell chamber,the animal model of xenotransplanted gastric carcinoma in nude mice was established to detect invasive ability in vivo. 2.Construction and primary identification of a naive Fab fragment phage library

Objective:To construct a human Fab fragment phage display library and provide a platform for human antibody preparation.Methods:Peripheral blood lymphocytes were collected from healthy donor.The heavy chain Fd fragment and light chain of human immunoglobulin's genes were amplified by RT-PCR,and then cloned into phagemid pComb3XSS to generate human phage antibody library.Cutting with endonucleases such as SacⅠ,XbaⅠ,XhoⅠand SpeⅠto identify the insertion of the light chain or heavy chain Fd genes.IL-2 and digoxin as the antigen was used to scan the phage antibody library.Results:A phage antibody library of Fab had 8.4?107 members and it's recombinant rate was 70%.Through DNA sequencing of one positive clone,it was showed that its heavy chain belonged to IgG subvariety and its light chain to ? family.Conclusion:The success of constructing a nave human phage antibody library proves the useful of phage display system in human antibody preparation,and it can be used to select,purify and express of amylin Fab antibody.
3.Applied research of selective bronchial arterial embolization for severe hemoptysis

Objective To observe the clinical curative effect of selective bronchial arterial embolization for severe hemoptysis.Methods Thirty-eight hospitalized patients with severe hemoptysis was enrolled in this study.Of which 31 patients were recurrent hemoptysis,27 patients were with bronchiectasis,4 patients were with bronchial lung cysts,4 patients had unknown causes,3 patients were with tuberculosis.All patients were diagnosed by chest imaging examination,fiber bronchoscopy,bronchial arteriography,and they were all treated by selective bronchial arterial embolization.Results The most times of embolization was 3,and bronchial arterial imaging were performed for vascular malformation.After having bronchial arterial embolization,35 patients were immediately released of bleeding.Postoperative 24 h,8 patients had hemoptysis again,of which 1 case was performed with conservative treatment and subsequently had pneumonectomy,5 patients had effective conservative treatment,2 cases were performed embolization again after failed conservative treatments.Hemoptysis completely disappeared within 1 week.The total effective rate was 97.4%(37/38).Patients were followed up for 1 year,of which 13 patients relapsed again,11 patients'annual and each time's quantity of hemoptysis reduced 50%.Two patients had pneumonectomy after a number of embolization.The total effective rate was 94.7% (36/38).After treatment,3 patients had mild chest pain in short time,2 patients had shoulder pain,3 patients had chest tightness,and 3 patients had subcutaneous passive congestion.After being treated for short-term,these symptoms disappeared eventually.Conclusion Selective bronchial arterial embolization for the treatment of acute or recurrent severe hemoptysis is very effective,and can avoid the risk of surgery.It is effective for recurrent cases and worthy of clinical application.
4.Endovascular therapy for complicated aortic dissection

Objective To explore the method of endovascular therapy for complicated aortic dissections.(Methods) The clinical data of 25 patients with complicated aortic dissections were analysed retrospectively.Results The patients′ ages ranged from 31-72 years with a mean of 50.2 years.Among the 25 cases,6 cases had severe ischemia of mesenteric artery,5 cases had renal artery ischemia,4 cases had severe(ischemia) of both legs,3 cases had renal arteries ischemia combined with superior mesenteric artery ischemia,2 had complicated aortic dissection combined with AAA,and in 5 cases the true aortic lumen was totally(compressed) by the false aneurysmal lumen.All patients underwent endovascular therapy,and the instant(technique) was successfully performed in all patients.Endoleak occurred in 3 cases after the stent-graft(deployment),it stopped spontaneously in 2 of them 7 days later,and 1 case with endoleak waiting for(treatment).In the other 22 patients,angiography after the operation showed that all the diseased area were sealed completely,and the viscera arterial blood supply was restored mainly via the true lumen.All the(patients) were cured and discharged.Conclusions In the management of complicated aortic dissections,(endoluminal) technique is simple,less traumatic,safe and has less complications as compared to the traditional operation.Improvement of the endoluminal technique is essential for successful treatment of these complicated cases.
5.Application of wavelet transform in quatum dot barcodes identification

BACKGROUND:To obtain more quantum dot (QD) barcodes,the overlay peaks of fluorescence occur,leading to difficulties in identifying QD barcodes.OBJECTIVE:To identify QD barcodes of two adjacent wave length using wavelet transform technique.METHODS:Through the microscopy,the spectrum of fluorescence induced by 375 nm light was captured by spectroscopy.The spectral signal was split into multi-scale components by wavelet transform.After transformed by spline function,every component constructed a new spectrum with peaks expanded by inverse wavelet transform.RESULTS AND CONCLUSION:Interpolation operation was performed on original data to control the data length to 2n.Following wavelet transform,peak location remained unchanged,so the eigenvalue of spectrum of coding fluorescence was extracted.The spectrum of fluorescence mixed with microspheres was split,and two QD barcodes were identified.The improved barcodes identification of adjacent spectrum increase color of QD barcodes,thereby enhancing code information volume.Results show that following spectrum was processed by wavelet transform,overlay peaks of fluorescence has be expanded,and enhanced the efficiency of recognition,which lays a foundation for detecting tumor markers.
6.Analysis of four cases of folliculotropic mycosis fungoides

Here four cases of folliculotropic mycosis fungoides are reported.Of these patients,one was a female and three were males with the age varying from 32 to 52 vears.Three patients presented with multiple,densely distributed,irregularly shaped,dark red infiltrated plaques,nodules,tumors,follicular papules and acneiform lesions preferentially distributed on the head and neck,as well as patches and mildly infiltrated plaques.foilicular papules and aeneifotin lesions on the trunk and extremities.One patient presented with follicular papules all over the bodv with the exception of head and face.Characteristic findings of histopathology included massive lymphoid cell infiltration.heteromorphism of some cells and migration of infiltrated cells into follicular epithelium.No typical epidermotropism was noticed.Two cases showed mnciprotein deposition in follicular epithelium,which was positive for alcian blue staining.The infiltrates were predominated bv CD4+ T lymphocytes.Folliculotropic mycosis fungoides is a refractory disense with poor response to conventional therapy of classical mycosis fungoides.Relapse is common in patients with partial remission.
7.Study on the relationship between cyclin D1 G870A polymorphism and gastric cancer

Objective To assess whether the G870A polymorphism of cyclin D1 (CCND1)was associated with individual susceptibility to gastric cancer in China. Methods Case-control study of 214 individuals was conducted, including 108 controls (non ulcerative dyspepsia) and 106 cases of gastric cancer. The polymorphism analysis of CCND1 A/G was performed in blood samples by PCR-RFLP method. Results The frequency of GG genotype was 19.8% and 7.4% in patients with gastric cancer and controls, respectively. The risk for gastric cancer in patients homozygous (GG) for CCND1 genotypes was 3.5 times higher in comparison with patients carrying AA genotypes (P<0.05, OR =0.281). Furthermore, the stratification analyses showed that the male, elderly (≥60 years) or H. pylori infected gastric cancer patients who carrying GG genotype were more susceptible to gastric cancer compared with those carrying AA or GA genotype. Conclusion GG genotype is associated with increased susceptibility to gastric cancer in high risk area of Xi'an.
8.Urinary metabolomics study of the effects of Scutellaria baicalensis Georgi ethanol extract on D-galactose-induced rats.

The purpose of this study is to evaluate the anti-aging effects and reveal the underlying mechanism of Scutellaria baicalensis Georgi ethanol extract (SBG) in D-galactose-induced rats. Fifty rats were randomly divided into five groups: vehicle control group, D-galactose group, and D-galactose combined with 50, 100, 200 mg x kg(-1) SBG. A rat aging model was induced by injecting subcutaneously D-galactose (100 mg x kg(-1)) for ten weeks. At the tenth week, the locomotor activity (in open-field test) and the learning and memory abilities (in Morris water maze test) were examined respectively. The urine was collected using metabolic cages and analyzed by high-resolution 1H NMR spectroscopy combined with multivariate statistical analyses. The SBG at doses of 50, 100 and 200 mg x kg(-1) treatments groups could significantly ameliorate aging process in rats' cognitive performance. The 50, 100, 200 mg x kg(-1) SBG regulated citrate, pyruvate, lactate, trimethylamine (TMA), pantothenate, β-hydroxybutyrate in urine favorably toward the control group. These biochemical changes are related to the disturbance in energy metabolism, glycometabolism and microbiome metabolism, which is helpful to further understanding the D-galactose induced aging rats and the therapeutic mechanism of SBG.
9.Dynamic Changes of Phenotypes and Secretory Functions during the Differentiation of Pre-DCs to Mature DCs

The dynamic expression of cytokines and phenotypes during the differentiation process of dendritic cell precursors (pre-DCs) to mature dendritic cells (DCs) was investigated,and the effects of inflammatory stimulation with lipopolysaccharide (LPS) on DCs differentiation were understood.The differentiation of bone marrow cells isolated from Balb/c mice was induced to DCs in an 8-day cell culture system with RPMI-1640 complete culture medium containing 10% FBS,20 ng/mL recombinant mouse granulocyte-macrophage colony-stimulating factor (rmGM-CSF) and 10 ng/mL recombinant mouse interleukin-4 (rmIL-4).On the day 3,6 and 7 after culture,DCs were divided into non-LPS group and LPS group,given 500 ng/mL LPS for 24 h stimulation and no stimulation respectively.The expression levels of CD 1 l c+,MHC-Ⅱ +,CD40+,CD80+ and CD86+ were detected by flow cytometry,and those of IL-2,IL-4,IL-10,IL-12 p70 and IFN-γ in the supernatant by ELISA.On the day 3 and 6 after culture,the expression of IL-2,IL-4,IL-10 and IFN-γ in DCs showed no significant differences between non-LPS group and LPS group,whereas the differences were significant at day 7.The expression levels of cytokines (for IL-2,IL-4,IL-10,IFN-γ and IL-12 p70:152.86±6.91,778.33±8.35,44.55±2.54,5.8.26±1.09 and 2423.00±57.21 pg/mL respectively) in LPS group were higher than those in non-LPS group,especially IL-12 p70 increased obviously at day 7.It was concluded that during the differentiation process of pre-DCs to mature DCs,LPS stimulates DCs producing large amounts of IL-12 p70 and Thl-type cytokines as compared with Th2-type cytokines,and day 7 may be a key time-point for DCs polarization.
10.Parabiosis theory and effective stimulating amount, reinforcing-reducing, therapeutic time and course of acupuncture.

Parabiosis theory clearly expounds the change course of stimulatiom, namely, parabiotic equalization phase, parabiotic deregulation phase and parabiotic inhibition phase, and it can clearly explain that the effective stimulating amount is directly related to the reinforcing or reducing, and establishment of the therapeutic course. That is, the large effective stimulating amount is reducing method and the small effective stimulating amount is reinforcing method. In clinical application, for deficiency syndrome and chronic diseases, the effective intensity of acuti puncture should be smaller and the needle should be retained for 20-30 min, and for excess syndrome and acute disd eases, the effective stimulating intensity of acupuncture should be larger and the needle should be retained for longer time. In acupuncture treatment, acupoints should be grouped and used alternately, and the interval between therapeutic courses should be established.
