2.Culture of rat skeletal muscle satellite cells and their response to stimulation by insulin-like growth factor

Objective To observe the proliferation and differentiation properties of rat skeletal muscle satellite cells that are dissected,cultured and purified in vitro and investigate their response to stimulation by insulin-like growth factor I(IGF-I).Methods The cells were isolated from Wistar rats by two-step enzyme digestion and purified by the Percoll density centrifugation to observe their prolif- eration and differentiation properties and draw growth curve and fusion rate curve.Then,the response of the satellite cells to stimulation by IGF-I at various concentrations was determined by using MTT meth- od.Results Plenty and purified satellite cells were obtained via these methods.Conclusion IGF- I at appropriate concentration(100 ng/ml)can promote the proliferation and differentiation of rat skele- tal muscle satellite cells.

4.COMPARISON OF GROWTH FACTOR mRNA EXPRESSIONS IN HUMAN SKIN FIBROBLASTS BETWEEN TWO AND THREE-DIMENSIONAL CULTURE SYSTEMS

To compare the growth factors mRNA expressions in human skin fibroblasts cultured in two (2D) or three dimensional systems (3D). Human skin fibroblasts were isolated and cultured in collagen jelly (3D) or the regular culture plates (2D). After 72 hours, the total RNA of fibroblasts were extracted. Specific oligonucleotide primers for the growth factor VEGF and bFGF were synthesized and reverse transcriptase polymerase chain reaction (RT PCR) was utilized to amplify the mRNA of those growth factors. RT PCR productions were separated by electrophoresis in 1% wt/vol agarose gels and photographed. Semi quantitative data were statistically analyzed. Although the morphology of fibroblasts in 3D culture was different from that in 2D culture, the RT PCR analysis revealed the same expression and concentration of VEGF mRNA and bFGF mRNA in human skin fibroblasts cultured in different systems.

6.Continuous irrigation with recombinant human acid fibroblast growth factor combined with negative-pressure drainage for the treatment of refractory skin ulcer

Objective To evaluate the clinical efficacy of continuous irrigation with recombinant human acid fibroblast growth factor (rh-aFGF) combined with negative-pressure drainage in the treatment of refractory skin ulcer.Methods A clinical trial was performed from January 2010 to December 2012.Sixty patients with refractory skin ulcer were randomly and equally classified into three groups:group A receiving continuous irrigation with rhaFGF combined with negative-pressure drainage,group B receiving irrigation of raw surfaces with rh-aFGF followed by bandaging after debridement,group C receiving covering of raw surfaces with Vaseline gauze and absorbent gauze followed by bandaging after debridement.Dressings were changed once daily in every group.The treatment lasted 21 days.The efficacy of these regimens was evaluated on day 7,14 and 21.Results At the above time points for observation,both the granulation and epithelium tissues of ulcers grew well in group A,with a decrease in the volume of wounds and area of exposed deep tissues.There was a significant increase in the cure rate and reduction rate of wound volume in group A compared with the other two groups (all P < 0.01).Conclusion Refractory dermal ulcer can be treated with continuous irrigation with rh-aFGF combined with negative-pressure drainage.
7.Adaptation of emphysema rabbit diaphragm muscle mechanics to chronic ultra-low frequency electrical stimulation and chronic physiological frequency electrical stimulation

Objective To investigate the effect of chronic ultra-low frequency electrical stimulation on mechanical characteristic of diaphragmatic muscle of emphysema rabbit.Methods The rabbit model of emphysema was made by inhaling papain of ultrasonic atomization once a week for 3 weeks in 24 adult Japanese rabbits,6 receiving 10 Hz chronic ultra-low frequency electrical stimulation,6 receiving 40 Hz,6 receiving 2.5 Hz and 40 Hz.Another 6 rabbits served as control.The twitch tension (Pt),titanic tension(Po),time to peak tension(TPT),half-relaxation time(1/2RT),fatigue index(FI) and fatigue recovery index(FRI) were measured in control group,emphysema group,emphysema+chronic ultra-low frequency electrical stimulation groups.Results As compared with the control rabbits, Pt and Po decreased,TPT and 1/2RT prolonged,FI and FRI increased in the only emphysema rabbits.Pt and Po were higher,TPR and 1/2Rt was shorter in the only emphysema rabbits than the rabbits in 40 Hz and(2.5+40) Hz groups(P0.05),more increase in FR and FRI(P
8.Analysis of misdiagnosis and treatment of acute superior mesenteric arterial embolism

Objective To explore the early diagnosis and treatment of acute superior mesenteric arterial embolism (ASMAE).Methods The clinical data of 25 patients with acute superior mesenteric arterial embolism were retrospectively analyed.Results All 25(100%) patients had severe abdominal pain with abdominal signs,21(84%) patients had history of valvular heart disease or atrial fibrillation,and 17(68%) patients had hematochezia.All the cases were misdiagnosed preoperatively,including 11 patients were misdiagnosed as abdominal pain of unknown causes,3 patients as acute pancreatitis,3 patients as acute gastroenteritis,2 patients as necrotizing enteritis.1 patient as acute appenditis,1 patient as acute myocardial infarction,and 1 patient as acute cholecystitis;but 3 patients were preoperatively suspected to have impairment of mesenteric blood flow.All of the 25 patients underwent necrotic bowel resection,6 patients died,and 7 patients abandoned treatment because of serious complications such as short bowel syndrome etc.Mortality rate was 51%(13/25).Conclusions Early diagnosis of patients with acute superior mesenteric arterial embolism is difficult;the main cause of misdiagnosis is lack of recognition of its clinical signs.Fully grasping the characteristics of patients with early stage disease and effective early intervention are the fundamental measures for reducing mortality.
9.Application of low-dose dual-source CT prospective electrocardiography gated scanning coronary imaging in evaluation of high body mass index patients

Objective To evaluate the necessity and feasibility of low-dose dual-source CT coronary angiography imaging in the evaluation of high body mass index (BMI) patients. Methods Sixty patients with BMI≥28 kg/m2 and heart rate 65-90 beats/min were selected, and they were divided into prospective electrocardiography gated scanning group and retrospective electrocardiography gated scanning group by random digits table method with 30 cases each. The image quality, CT dose weighing index (CTDI) and dose length product (DLP) were compared between 2 groups. Results There was no statistical difference in the score of coronary image quality between retrospective electrocardiography gated scanning group and prospective electrocardiography gated scanning group:(3.47±0.61) scores vs. (3.18±0.94) scores, P>0.05. The CTDI and DLP in prospective electrocardiography gated scanning were significantly lower than those that in retrospective electrocardiography gated scanning: (32.44 ±10.18) mGy vs. (45.86 ±15.34) mGy and (458.00±95.27) mGy/cm vs. (532.15±154.43) mGy/cm, and there were statistical differences (P<0.05). Conclusion For high BMI patients, different low-dose dual-source CT scanning, especially prospective electrocardiography gated scanning, can guarantee image quality and reduce the radiation dose and the radiation damage at the same time.
10.Changes of PKC membrane translocation of the brain in neonatal SD rats after hypoxic-ischemic insult

Objective To explore the role of protein kinase(PKC)in the nosogenesis of hypoxic-ischemic encephalopathy(HIE).Methods Twenty-four seven-day-old SD rats were divided randomly into normal group (N),model control group(C)and HIE group(HIE).The biochemistry techniques of SDS-PAGE and western bolt were used to examine the level of PKC membrane translocation of cortex and hippocampi.Results The level of PKC membrane translocation of cortex and hippocampi in N group was 0.28±0.02,0.29±0.02 respectively,and 0.29±0.02,0.30±0.02 in C group,and 0.38±0.04,0.40±0.06 in group HIE.The level of PKC membrane translocation of cortex and hippocampi was higher in HIE group as compared with that in N,C group(P<0.01),but there was no different between N group and C group(P>0.05).Conclusion PKC are activated in HIE,which indicates that PKC may involve in the pathophysiologic mechanism of HIE.
