2.Quality Control and Administration of Disinfectant in Manufacturing Laboratory

OBJECTIVE To prevent infection during the processes of dispensation and distribution of disinfectant. METHODS All the key processes were put under strictly control including purchase,preservation of disinfectant,quality of dispensing processes and administration of the disinfectant distribution. RESULTS Both effective ingredients and microorganism were coincided with the definite criterion. CONCLUSIONS Quality control and administration of disinfectant in manufacturing laboratory is important to guarantee the quality of disinfectant dispensation and to prevent from nosocomial infection as well.
3.Investigation of catheter-related thrombosis around the port in patients with breast cancer after chemotherapy

Objective·To investigate the incidence, risk factors and treatment of the catheter-related thrombosis (CRT) in breast cancer patients after implantation of totally implantable venous access port (TIVAP) in chemotherapy. Methods·A total of 190 cases after implantation of TIVAP were investigated. Color Doppler ultrasound was used to monitor the neck blood vessels to find whether there was CRT before chemotherapy and before taking out the port. The incidence of CRT, occurrence time, risk factors and treatment efficacy were observed. Results·There were 112 (58.9%) cases with CRT and 108 (56.8%) patients with asymptomatic thrombosis, and only 4 cases had symptomatic thrombosis, the incidence of which was 2.1%. Most thrombosis developed on the 21th day after catheterization, and the patients over the age of 60, with clinical stage Ⅲ - Ⅳ and chemotherapy regimens TEC (docetaxel combined pirubicin and cyclophosphamide) were the risk factors for thrombosis. All the patients with asymptomatic thrombosis accepted anticoagulant treatment with low molecular heparin, earthworms enzyme or aspirin, respectively, but there was no significant difference in efficacy in the three groups (P=0.743). Conclusion·Port catheter related symptomatic thrombosis incidence is low but the incidence of symptomatic thrombosis is high in the breast cancer patients after chemotherapy. Age, tumor stage and TEC chemotherapy regimens are the risk factors for catheter-related thrombosis.

6.Application of in vitro bionic digestion and biomembrane extraction for metal speciation analysis, bioavailability and risk assessment in lianhua qingwen capsule.

One of the causes of the high cost of pharmaceuticals and the major obstacles to rapidly assessing the bioavailability and risk of a chemical is the lack of experimental model systems. A new pre-treatment technology, in vitro bionic digestion was designed for metal analysis in Lianhua Qingwen capsule. The capsule was digested on 37 degrees C under the acidity of the stomach or intestine, and with the inorganic and organic compounds (including digestive enzymes) found in the stomach or intestine, and then the chyme was obtained. Being similar to the biomembrane between the gastrointestinal tract and blood vessels, monolayer liposome was used as biomembrane model Affinity-monolayer liposome metals (AMLMs) and water-soluble metals were used for metal speciation analysis in the capsule. Based on the concentration of AMLMs, the main absorption site of trace metals was proposed. The metal total contents or the concentration of AMLMs in the capsule were compared to the nutritional requirements, daily permissible dose and heavy metal total contents from the "import and export of medicinal plants and preparation of green industry state standards". The metal concentrations in the capsule were within the safety baseline levels for human consumption. After in vitro bionic digestion, most of trace metals were absorbed mainly in intestine. The concentration of As, Cd, Pb was 0.38, 0.07, 1.60 mg x kg(-1), respectively, far less than the permissible dose from the "import and export of medicinal plants and preparation of green industry state standards".

7.Risk Factors of Laryngeal Carcinoma:A Case-control Analysis

Objective To analyze the risk factors of laryngeal carcinoma.Methods The data of laryngeal carcinoma inpatients of our hospital from October 2005 to November 2007 were reviewed retrospectively by a case control analysis.Results The males who have smoking addiction,alcohol consumption,family history of malignant tumor and have more risks to suffer from laryngeal carcinoma.For The family history,OR is 2.01 in the single factor analysis,and in the multivariate analysis the OR is 2.27.Conclusion Gender,smoking,alcohol and family history of malignant tumor have great effect on the risk of laryngeal carcinoma.Family history of malignant tumor is an independent risk factor.
8.Expression and Clinical Significance of c-Src in the Epithelial Ovarian Carcinoma

Objective:we aim to determine the relationship between Cell sarcoma (c-Src) expression in patients with EOC and the disease phenotype.Methods:c-Src expression was evaluated using Western blotting analysis in 21 ovarian carcinomas and 4 normal ovarian tissues.Immunohistochemistry was used to evaluate c-Src expression in 134 ovarian carcinomas and 26 normal ovarian tissues.The association between c-Src expression and clinically pathologic characteristics were also assessed in these patients.Results:Our results indicated elevated c-Src protein in EOCs compared with that in normal tissues.The overexpression of c-Src was significantly associated with aggressive features,such as advanced disease stage,poor histological grade,lymph node metastasis,and tumor recurrence (P<0.05).In addition,the overexpression ofc-Src is significantly associated with EOCs' prognosis.Conclusion:c-Src overexpression was significantly associated with the malignant biological behavior of tumor,suggesting c-Src as a potential preventive target in these patients.
9.Application of Flow Cytometry in Viability Detection of Pichia pastoris Cells

Cell viability of Pichia pastoris was detected by flow cytometry (FCM) with two reagents fluorescein diacetate (FDA) and propidium iodide (PI). Compared with FDA/PI double-stained dot plots and PI single-stained dot plots,the latter could divide dead and living cells into two separate zones,and get the correct proportion. Then PI single-stained method was used to detect the change of cell viability in Pichia patoris fermentation. At glycerol batch and fed-batch phase,little dead cells were detected. At methanol fed-batch phase,cell viability decreased when cell weight increased,and was only 73.8% at 88 h.
10.Clinical observation of palpebral spring placement for lagophthalmos secondary to facial nerve palsy

Background The dysfunction of the blink reflex the eyelid-closure ability appears in the patients with facial paralysis,and its management is the implantation of mechanical-assisted eye-closure device in the upper eyelid.A novel device is palpebral spring implant.However,there is no similar study in China.Objective This study was to evaluate the clinical efficacy of palpebral spring placement for lagophthalmos caused by facial nerve palsy.Methods This clinical research complied with Helsinki declaration and the protocol was approved by Ethic Committee of Henan Eye Institute & Henan Eye Hospital.Written informed consent was obtained from each patient prior to the surgery.A retrospective serial case-observational study was performed.The medical records of 11 patients who underwent palpebral spring placement for hypophasis due to facial nerve palsy were reviewed at Henan Eye Hospital from August 2010 to November 2012.Palpebral spring placement was performed by the same surgeon to ensure a more even outcomes.Palpebral spring was made by nickel wire,with the diameter of 0.3 mm and implanted on tarsal plate in 11 eyes of 11 patients with symptomatic facial nerve palsy.The lower tip of Levine spring was encased into a small terylene bag and sutured to the anterior tarsal surface during the surgery.Preoperative and postoperative symptoms,upper eyelid margin to mid pupil distance (ULMD),degree of lagophthalmos and eyelid moving scope were examined and compared between before and after operation.The operating complication was followed-up for 8-38 months.Results The discomforted symptoms disappeared in all the operated eyes.The ULMD was (3.51±0.73) mm in preoperation and (3.20±0.86) mm in posteration,without significant difference between them (t=1.36,P=0.10).The degree of lagophthalmos was (5.94±1.57) mm and (1.06±0.98) mm in preoperation and postoperation respectively,showing a significant difference between them (t =9.42,P =0.00).The eyelid moving scope was (5.89±0.70) mm in postoperation,which was significantly increased in comparison with (0.11 ±0.33) mm of preoperation (t =22.97,P =0.00).The palpebral spring implant was regulated in 1 patient during the follow-up duration due to the trauma.No complication in other 10 patients appeared during the follow-up duration,such as implant exposure,metal fatigue or infection.Conclusions Palpebral spring placement is safe and effective for lagophthalmos in patients with facial nerve palsy.
