Chinese Journal of Ultrasonography 2003;0(07):-
Objective To investigate the relation between serum homocysteine(HCY) and the development of atherosclerosis(AS).Methods Diet contained 1% methionine was fed to make models of hyperhomocysteinemia and atherosclerosis in 18 male New Zealand rabbits. Hyperlipemia group was as control group. Ultrasonographic image was performed in 0,4,8 weeks respectively with HP 8500 and recorded the stenosis and plaque of abdominal aorta compared with pathology. Results Stenosis and plaque of abdominal aorta were observed in 4 weeks in high fat diet group. Stenosis and plaque of abdominal aorta were observed in 8 weeks in high methionine diet group. This results were confirmed by pathology anatomy.Satistically, significant correlation was found between AS and hypercholesterolemia or hyperhomocysteinemia respectively. But there was no correlation between hypercholesterolemia and hyperhomocysteinemia. Conclusions Hyperhomocysteinemia is an independent important risk factor of atherosclerosis.

In order to assess whether gene transfection could be mediated by ultrasound in association with P85 and find the appropriate parameters of ultrasound irradiation, the effects of ultrasound with or without P85 on gene transfection of HepG2 cells were examined. The HepG2 cells were irradiated by ultrasound at 1 MHz, 0.4-2.0 W/cm(2) and 50% duty cycle with plasmid encoding enhanced green fluorescent protein (EGFP) as a report gene. Forty-eight h later, the expression of EGFP was detected under the fluorescence microscopy. Transfection efficacy was quantitatively assessed by flow cytometry, and cell viability was evaluated by trypan blue exclusion. The results showed that the transfection efficacy was increased with the increases in ultrasound output power and the ideal transfection efficacy was achieved in HepG2 cells irradiated by ultrasound at 0.8 W/cm(2) for 30 s. The transfection efficacy in ulstrasound+P85 group was three times higher than in single ultrasound group [(17.63+/-1.07)% vs (5.57+/-0.56)%, P<0.05]. The cell viability was about 81% and 62% in ultrasound group and ultrasound+P85 group respectively. It was concluded that ultrasound in combination with P85 could mediate the gene transfection of HepG2 cells, ideal transfection efficacy was achieved by ultrasound irradiation at 0.8 W/cm(2) for 30 s, and P85 could somewhat increase the damage to cells caused by ultrasound.

Chinese Journal of Geriatrics 2011;30(5):390-392
Objective To study the metabolic characteristics of temporal lobe injury during the radiotherapy in the elderly with nasopharyngeal carcinoma through 1H- magnetic resonance spectroscopy (MRS). Methods The 15 aged and 23 young cases among 80 cases with nasopharyngeal carcinoma confirmed clinically and receiving radiotherapy for the first time were involved in the same period of July 2006 to May 2009. Conventional MRI scanning and bilateral temporal MRS were performed before and during radiotherapy at radiation dosages of 20 Gy, 40 Gy, and 60 Gy respectively. The spectroscopy morphology and the ratios of the three main metabolites were compared and analyzed. Results The values of N-acetylasparate (NAA)/creatine (Cr) was lower and choline-containing compound (Cho)/Cr ratio was higher in aged group than in young group before radiotherapy (P>0.05). The spectroscopy morphology stepped down, NAA, Cho, NAA/Cr, Cho/Cr and NAA/Cho decreased to some extent along with the adding of radiation dosages in the course of radiotherapy. At the end of radiotherapy, the decreased amplitude of NAA/Cr value was larger in aged group than in control group (P<0.01), and the amplitude of Cho/Cr value was lower than other group. Conclusions The neurons of aged patients with nasopharyngeal carcinoma have a weak tolerance to the radiation, brain injury occurs earlier and heavier in aged patients than in young patients. It is necessary to prevent neuron injury early.

Objective To study the relationship between PDD and the polymorphism of apolipoprotein E(apoE) gene,to inquire the etiological basis of PDD.Methods 11 Patients with PDD 40 patients with PD without dementtia and 52 unrelated controls were collected to detect the genotype of apoE gene by PCR- RFLP. Results There was no difference in the apoE allele frequencies and genotype distribution in PDD group,PD without dementia group and control group(P >0.05). Conclusion There is no relationship between the polymorphism of apoEgene and the onset of PDD.The pathogenesis of PDD differs from that of AD.

AIM:To investigate the inhibitory effect of ground dragon on the expression of ?-SMA and FN in the lung tissue with asthma.METHODS:The BALB/c mice were divided into four groups:control group(group A,n=20),asthmatic model group(group B,n=20),large-dose ground dragon treatment group(group C,n=20)and low-dose ground dragon treatment group(group D,n=20).To establish a mouse model of chronic asthma,we sensitized the mouse with 0.02% ovalbumin(OVA)by intraperitoneal injection, and stimulated the mice with 1% OVA by atomization. The treatment groups were given ground dragon before stimulation every time. After the last time of stimulation, the mice were subjected to laboratory tests.Inflammatory cells in bronchoalveolar lavage fluid were counted.Total level of IgE in serum was detected by ELISA.FN mRNA and ?-SMA mRNA in the lung tissue were measured by RT-PCR and AlphaImager 2200 semi-quantitation analysis system.Expressions of FN and ?-SMA were measured by the method of two-step immunohistochemistry and leica QWIN V3 analysis system.RESULTS:(1)Compared with those in group A,the expressions of ?-SMA and FN in group B were significantly increased(P0.05).(2)Compared with those in group A,the expression levels of ?-SMA mRNA and FN mRNA in group B had a great increase(P0.05).CONCLUSION:The ground dragon inhibits ?-SMA and FN expression in the lung tissue of mice with chronic asthma, indicating that ground dragon may inhibit airway remodeling in asthma through the inhibition of ?-SMA and FN expressions.
