1.Correlation of expression of ER, PR and c-erbB-2 with ultrasonographic features in invasive ductal cancer of breast

Objective To analyze the relationship of the expression level of estrogen receptor (ER), progesterone receptor (PR) and human epidermal growth factor receptor-2 (HER-2, c-erbB-2) of breast invasive ductal carcinoma (IDC) with ultrasonographic characteristics. Methods Totally 104 patients with IDCs confirmed pathologically were involved in this study. ER, PR and c-erbB-2 expression in the IDC specimens was determined by immunohistochemical staining technique. The correlation between the ultrasonographic features and the positive expression of ER, PR or c-erbB-2 was analyzed. Results The positive expression rate of ER and PR in the group with tumor spiculation sign and posterior acoustic attenuation was higher than that in the group without. The positive expression rate of ER differed significantly (P<0.05) while that of PR did not (P>0.05). The over-expression rate of c-erbB-2 in the group of microcalcification, sufficient blood flow and axillary lymph node metastasis was higher than that in the group of non-microcalcification, deficient blood flow or without axillary lymph node metastasis (P<0.05). The expression of ER, PR and c-erbB-2 was not related to the size of tumor (P>0.05). Conclusion The expression of ER and c-erbB-2 is closely related to the ultrasonographic characteristics of IDC, which may, to some extent, reflect the expression level of ER and c-erbB-2.
2.Effect of fluvastattn on extracelluar signal-regulated kinase activity in glomerular mesangial cells exposed to high glucose concentration

When the glomerular mesangial cells of rats were cultured in vitro at high glucose concentration,the activity of extraeellular signal-regulated kinase(ERK),the expression of transforming growth factor-?_1(TGF-?_1)mRNA and the content of typeⅣcollagen in the supematant were higher than those at normal glucose concentration.These effects were inhibited by fluvastatin.The results showed that the activation of ERK signal transduction pathways appeared to play a role in the onset and progression of diabetic nephropathy. Furthermore,fluvastatin could protect the kidney by inhibiting ERK signal transduction pathway and TGF-?_1 expression.
3.Preventive effect of itraconazole oral solution for invasive fungal infection in neutropenic patients with acute leukemia after chemotherapy

Objective To evaluate the efficacy of itraconazole oral solution for prevention of invasive fungal infection ( IFI ) in neutropenic patients with acute leukemia after chemotherapy.Methods Clinical data of 136 neutropenic patients with acute leukemia after chemotherapy at the Department of Hematology,Nanfang Hospital from January 2008 to December 2010 were retrospectively analyzed.Patients were divided into itraconazole group ( n =67 ) and control group ( n =69).There were 36 patients with acute nonlymphocytic leukemia ( ANLL),31 with acute lymphoblastic leukemia (ALL) in itraconazole group;while in control group,there were 30 patients with ANLL,38 with ALL and 1 with biphenotypic acute leukaemia (BAL).Patients in itraconazole group received intraconazole after chemotherapy until the neutrophil count was increased to 0.5 × 109/L or the body temperature returned to normal and without any imaging evidence of IFI.The incidence of IFI and clinical features were compared between the groups using SPSS 13.0 software.Pearson x2 test was used for nominal variables,for measurement data,t (normal distribution) or Mann-Whitney U (skewed distribution) test were used.Results There were 12 cases ( 17.9% ) suffering from IFI in itraconazole group and 32 cases (46.4%) in the control group (x2 =12.59,P < 0.01 ).For ANLL patients,the incidence of IFI in itraconazole group was significantly lower than that in control group ( 16.7% vs.56.7%,x2 =11.53,P <0.01 ).In itraconazole group,the incidence of IFI in female patients was significantly lower than that in male patients ( 8.6% vs.28.1%,x2 =4.35,P <0.05 ).And for the female patients,the incidence of IFI in itraconazole group was significantly lower than thatin the control group (8.6% vs.44.7%,x2 =11.98,P<0.01).Conclusion Itranconzole oral solution can effectively prevent IFI in neutropenic patients with acute leukemia after chemotherapy,especially for the female patients with ANLL.
4.Effect of percutaneous coronary intervention and medical therapy on quality of life and cognitive function in patients with coronary heart disease

Objective:To explore the effect and difference of percutaneous coronary intervention (PCI) and medical therapy on quality of life and cognitive function in patients with coronary heart disease (CHD),and to investigate the relationship between quality of life and cognitive function.Methods:A total of 320 patients with CHD,who underwent coronary angiography and PCI (PCI group,n=160),or underwent coronary angiography and medical therapy (drug therapy group,n=160),were selected.The quality of life was assessed by using the Health Survey Form SF-36 (SF-36) and the Seattle Angina Questionnaire (SAQ),and the cognitive function was assessed by using the Mini-Mental State Examination (MMSE).General data of patients were collected on the day of coronary angiography.Telephone follow-up was conducted in 1 month after treatment,and the outpatient review was carried out in 3 and 6 months after treatment.Results:A total of 309 valid questionnaires were collected.The scores of quality of life in the PCI group and the drug therapy group after treatment were both increased compared with those before treatment (both P<0.05).The SF-36 scores of four dimensions (role physical,bodily pain,vitality and mental health) in the PCI group were all significantly greater than those in the drug therapy group (all P<0.05).The SAQscores of two dimensions (angina stability and angina frequency) were both higher in the PCI group than those in the drug therapy group in 6 months of post-operation (all P<0.05).There was no significant difference in cognitive function before and after the treatment in the 2 groups (P>0.05).There was no significant difference in cognitive function between the PCI group and the drug therapy group (P>0.05).In the PCI group,physical function,role physical,bodily pain,and role emotional were positively correlated with cognitive function (r=0.207,0.182,0.184,0.176 respectively,all P<0.05).In the drug therapy group,there was no correlation between quality of life and cognitive function.Conclusion:The quality of life for the patients is improved in the PCI group and the drug therapy group,but the improvement degree in the PCI group is more obvious.Both PCI and drug therapy do not result in the decrease of cognitive function,and there is no difference between the 2 groups.There is positive correlation between quality of life and cognitive function in the PCI group,there is no correlation between quality of life and cognitive function in the drug therapy group.

6.Study on chromatographic fingerprint of extractive from Tripterygium wilfordii with supercritical carbon dioxide.

OBJECTIVETo establish the chromatographic fingerprint of supercritical carbon dioxide extract of Tripterygium wilfordii.
METHODHPLC method was applied for quality assessment of T. Wilfordii, HPLC analysis was performed on Kromasil C18 (4. 6 mm x 250 mm, 5 microm) with the mixture of acetonitrile-1% per thousand H3PO4, as mobile phase in gradient mode. The samples were detected at UV of 267 nm with column temperature of 35 degrees C, analytic time was 80 min; Flow-rate was 1.0 mL x min(-1). The chromatographic fingerprint of ten batches of samples was determined, for establishing the chromatographic fingerprint of T. Wilfordii.
RESULTIndicating 27 peaks in common, identified 21 peaks with chemical reference and HPLC-MS, and the HPLC fingerprint was established.
CONCLUSIONThe method is steady and accurate with a good repeatability and can be used as a quality control method for T. Wilfordii.

7.Preparation of liposomes containing extracts of Tripterygium wilfordii and evaluation of its stability.

OBJECTIVEThe liposomes containing extracts of Tripterygium wilfordii were prepared and the possibility of entrapment of complex chemicals by liposomes were studied.
METHODThe liposomes containing extracts of T. wilfordii were prepared by thin-film dispersion method, the effect of process parameters and composition of materials on the entrapment efficiency of the main components were studied. The stability of the liposomes dispersion was also evaluated.
RESULTThe liposomes made by thin-film dispersion method were mostly small unilamellar vesicles and their particle size was 30 nm to approximately 50 nm. The optimum entrapment efficiency of tripterine and the total alkaloids were respectively 98.10% and 88.63% but the liposomes dispersion was unstable when kept at 4 degrees C.
CONCLUSIONThe complex chemicals can be entrapped by the liposomes, but its stability need to be improved furtherly.

8.The study on frequency distribution of regulatory T cells and its functional markers in peripheral blood of chronic hepatitis B.

OBJECTIVETo explore the role of treg cells and its functional markers in pathogenesis of chronic hepatitis B, and the correlation with disease progression.
METHODS20 cases of healthy control people,53 cases of chronic hepatitis B patients and 24 cases of liver cirrhosis patients were enrolled into the groups. Detecting the frequencies of CD4+ CD25+ Fox3+ cells, CD4+ CD25+ CD127(low) cells, CD39+ treg cells and CTLA-4+ treg cells in treg cells by flow cytometry. Clinical parameters were investigated in the same time.
RESULTSThe frequencies of treg cells, CD4+ CD25+ CD127(low) cells and CD39+ treg cells were significant different among healthy control group, CHB group and LC group (P < 0.01). The frequencies of treg cells, CD4+ CD25+ CD127(low) cells and CD39+ treg cells were significantly different in moderate-severe CHB group compared with mild CHB group (P < 0.05, P < 0.05, P < 0.01). In CHB group the frequencies of CD4+ CD25+ Foxp3+ cells were positively correlated with ALT (r = 0. 289, P < 0.05) and AST (r = 0.302, P < 0.05), the frequencies of CD4+ CD25+ Foxp3+ cells had a significant positive correlation with the frequencies of CD4+ CD25+ CD127(low) cells (r = 0.478, P < 0.01).
CONCLUSIONThe frequencies of treg cells and its functional markers probably had a dynamic tendency in the process of chronic hepatitis B and were closely related with the change of liver function parameters. CD39+ treg cells may be a group of functional treg cells, which indicated that CD39 be a sensitive marker to react treg cells function. In some sense, CD4+ CD25+ CD127(low) cells frequency could represent treg cell frequency.

9.Preparation and identification of monoclonal antibody against human thrombomodulin..

To produce specific monoclonal antibody (McAb) against human thrombomodulin (hTM), the full-length hTM cDNA-expressing plasmid pThr402 was transfected into CHO cells by Lipofectamine 2000 reagent. The hTM-expressing CHO cells, which was confirmed by flow cytometry and Western blot, were obtained by G418 selection. Then the McAb against hTM was prepared with classic hybridoma technique. A cell line of CHO-TM5 with high level of hTM was used to immunize female Balb/c mice 3 times at an interval of 4 weeks. On the third day after the third immunization, mice were sacrificed and spleen cells were harvested to prepare hybridoma cells with SP2/0 cells at the ratio of 10 to 1. Hybridoma cells were then cultured at 96-well plates for screening. Cellular enzyme-linked immunoabsorbent assay (CELISA) was applied twice. The first CELISA was done with polythene ELISA plate with a monolayer of CHO-TM5 cells. The positive clones from the first screen were then selected by reacting with similar screening ELISA plate but with CHO cell monolayer instead. Only clones that were positive for the first screening and negative for the second screening were kept, and called as CHO-TM5(+)CHO(-) hybridoma cells. Balb/c mice were intraperitoneally injected with the selected hybridoma cells. Ascites were collected and monoclonal antibodies were purified using FPLC, and its Ig class, subclass, and titer were then determined respectively. The specificity of the yielded McAb was identified with CELISA, flow cytometry, ABC immunohistochemistry and immunoblotting. One line of hybridoma cells with high expression of specific McAb against hTM, NH-1, was obtained. The Ig subclass of the McAb was IgG1 and the titer of ascitic McAb was 1x10(-6). Flow cytometry, CELISA and Western blot assays demonstrated that McAb NH-1 could specifically recognize hTM expressed in CHO-TM5 cells and human umbilical vascular endothelial cells. Meanwhile, the tissue specificity of antigen recognized by McAb NH-1 was identified by immunohistochemical ABC staining. NH-1 can specifically recognize the natural hTM expressed mainly in vascular endothelial cells, which will potentially be useful for investigation of the functions and clinic values of hTM.

10.Clinical profiles of circulating dendritic cell phenotype and lymphocyte subsets in patients chronically infected with HBV during lamivudine treatment.

BACKGROUNDTo investigate the changes of circulating dendritic cell (DC) and lymphocytes subsets in chronic hepatitis B patients treated with lamivudine.
METHODSSixteen chronic hepatitis B patients treated with lamivudine were included and followed up for 48 weeks in this study. Before and during lamivudine treatment, DC collected from peripheral blood sample was cultured in vitro and surface markers of DC and lymphocytes subsets were detected by flow cytometry simultaneously.
RESULTSOf the 16 patients, 11 were consistently HBV DNA negative in serum and HBV DNA YMDD variants appeared in 5 cases. In the consistent responder group, HLA-DR level of DC decreased transiently in 12 weeks and recovered in 48 weeks (P<0.05). At 48 weeks CD80, CD40 and CD1a were improved compared with baseline level (P<0.05). In the YMDD variant group, CD83 and HLA-DR level of DC decreased at 12 weeks treatment (P<0.05) and HLA-DR was still lower compared with baseline (P<0.05). In the consistent responder group, no significant changes occurred in lymphocyte subsets number at 12 weeks treatment, but CD4 + T cell was improved and NK cell dropped at 48 weeks compared with baseline level (P<0.05). In the YMDD variant group lymphocyte subsets had no statistically significant change.
CONCLUSIONIn the consistent responder group, the expression of surface costimulatory molecules of DC, such as CD80 and CD40,was partly recovered after the virus of hepatitis B had been inhibited efficiently, HLA-DR levels of DC decreased transiently at 12 weeks and recovered in 48 weeks and CD4+ T cell improved and NK cell dropped at 48 weeks. In the YMDD variant group, HLA-DR levels of DC were lower consistently during treatment compared with baseline level.
