1.Diagnosis and treatment of gastrointestinal stromal tumor: an analysis of 23 cases

Objective To evaluate the clinical diagnosis and treatment of gastrointestinal stromal tumors (GISTs). Methods The clinical data of patients with GISTs admitted into surgical department from July 2000 to Oct 2005 were retrospectively analyzed. Rusults Twenty-three patients received operation. Twenty-one patients underwent complete tumor resection, two did laparotomy only. Among the patients 7 cases were benign ,6 potential malignant, and 10 malignant . All patients were followed-up from 10 to 72 months. Three of the patients had local recurrence and metastasis, and 5 patients died. Conclusion It was difficult the diagnosis of GISTs because of the lack of clinical feature. Endoscopy and imaging examination are main methods to detect GISTs. Final diagnosis should be based on pathology. Surgical rection is the main treatment.

3.The effect of comfort care to alleviate postoperative pain of tibial eminence fracture by ;fixation

Objective:To study the effect of comfort care to alleviate postoperative pain of tibial eminence fracture by fixation. Methods:55 patients with tibial eminence fracture between Apr 2013 and Mar 2015 in our hospital were selected and divided into observation group with 26 patients and control group with 29 cases according to the nursing methods. The control group was given the orthopaedic routine nursing method, and the observation group was given the comfortable nursing method to compare two groups of patients with postoperative pain and psychological status before and after the intervention (HAMA anxiety score and HAMD depression scale). Results:There were significant differences between two groups of patients with pain score after the intervention (t=9.545, P<0.05), and HAMA and HAMD scores decline. HAMA and HAMD scores in the observation group were significantly lower than that in the control group (t=11.593, t=10.628;P<0.05). Conclusion:Comfort care can significantly relieve postoperative pain of the tibial condyle spine fractures and improve postoperative anxiety and depression. It has good clinical application value.
4.The application of Micro-CT in the microstructure of vertebrae

With the aging of population, the incidence of osteoporosis and intervertebral disc degeneration increased remarkably.As a consisting part of spine, vertebrae play important roles in spinal diseases.The microstructure of vertebrae is closely associated with not only its biomechanical properties, but also its adjacent intervertebral discs.A detailed understanding of vertebrae's microstructure can help us understand the initial mechanism, progression and prognosis of vertebrae and its adjacent discs' diseases.On the other hand, traditional examinations are less effective in detecting vertebrae's microstructure.Being a noninvasive, high-resolution and bony-sensitive neo-technology, Micro-CT has obvious advantages in detecting vertebrae's microstructure.Many researches have reported the usage of Micro-CT in reveling microstructures while they also raised problems.Thus, herein we searched the related information of vertebrae's microstructure scanning with Micro-CT, explained its basic mechanism, reviewed its progress in technology and arithmetic, and concluded the latest developments.Depending on the microstructure revealed by Micro-CT, achievements in biomechanical properties of vertebrae and intervertebral disc degeneration have been made,and related problems have been summarized.Also, new applications of Micro-CT in spinal diseases are discussed for acquiring reasonable clinical data in the future.

6.Evaluation on the Scientific and Research Influence of Institution Based on PageRank

The paper puts forward the method for evaluation on the scientific influence of institution based on PageRank algorithm,configures the initial weight of each institution based on the problems of dividing equally the webpage weight number in the classical PageRank algorithm and cited frequency of the application chapter,constructs the network matrix cited by institution through the citation situation of medical SCI papers published by partial colleges and universities in 2015,respectively calculates the PageRank values of institution when self-citation is excluded or included,and conducts comparative analysis on the sequence of cited frequency indicators of the institution as well as H index.This method can be used to evaluate scientific influence of institution from two dimensions of quality and quantity.
7.Rapid detection of E. coli O157 : H7 and other Shiga toxin-producing E. coli using ramification amplification method

Objective To develop a sensitive, specific, rapid and easy method for detecting E. coli O157 : H7 and other Shiga toxin-producing E. coli in food and clinical specimens. Methods A circular probe and capture probe specific for Shiga toxin-2 (stx2) gene had been synthesized and was used for determining the sensitivity of ramification amplification method (RAM). Different serotypes which contained stx2 gene, including an E. coli O157 : H7, an E. coli O46 : H38, an E. coli O111 : NM, an E. coli O22 : H8 and E. coli ATCC23716 (stx2 gene negative) were used for determining the specificity. All strains were detected by RAM to determine whether they contained stx2 gene. Real-time RAM was further studied to detect stx2 gene. Results The lowest number targets detected by RAM assay was 10 copies of stx2, indicating that RAM assay was as sensitive as conventional PCR. The result of specificity showed that different serotypes of strains were all positive for stx2 gene, while nonpathogenic E. coli ATCC23716 was negative. Among 32 isolates, 28 STEC isolates containing stx2 gene were positive by RAM assay, while others were negative. The RAM results were 100% in concordance with that of PCR. The real-time RAM results also showed that as many as 10 bacteria can be detected and the time of appearance of detectable signal was depended on the target concentration. Conclusion RAM assay can offer an alternative method for PCR to detect E. coli O157 : H7 and STEC in all types of specimens because of its simplicity and isothermal amplification.
8.Down-regulation of the bcl-2 expression level in a human cholangiocarcinoma cell line using ribozyme

Objective To study the role of down regulation of bcl 2 expression of human cholangiocarcinoma cells with bcl 2 mRNA cleaving ribozyme (RZ). Methods Synthetic gene of bcl 2 mRNA cleaving RZ was recombined with the expression vector of eukaryotic cells. The recombined vector was used to transfect human cholangiocarcinoma cells. The expression of bcl 2 in human cholangiocarcinoma cells was observed by immunocytochemistry and flow cytometry. Results The expression of bcl 2 in human cholangiocarcinoma cells transfected with RZ gene was lower than that of control group. Spontaneous apoptosis was observed in a few of the cells. Conclusion bcl 2 mRNA cleaving RZ can down regulate the expression of bcl 2 of cholangiocarcinoma cells notably and can also induce spontaneous apoptosis.
9.Purification of Polysaccharide from Angelica sinensis by Ultrafiltration Membrane Technology

Objective To isolate and purify polysaccharide from Angelica sinensis by ultrafiltration technology. Methods Extract of polysaccharide of Angelica sinensis was ultrafiltrated with different aperture membrane, the separation parameters on membrane separation efficiency of polysaccharide of Angelica sinensis were optimized by and orthogonal test. Results Polysaccharide of Angelica sinensis was separated with 200, 100, 50, 20 kD membrane. The contents of polysaccharide was 30.47% before ultrafiltration. The main component of Angelica sinensis polysaccharide was the fraction with 200 kD (65.42%). The optimum condition of ultrafiltration was obtained as follow:one times of extract volume, 35 ℃, 0.3 MPa. Conclusion The technology was simple and available, and the content of Angelica sinensis polysaccharide was higher.
10.Analysis of serum neopterin level in adults patients with Kashin-Beck disease in Qinghai Province

Objective:To investigate the changes of serum neopterin (NPT) level in adults patients with Kashin-Beck disease (KBD) in Qinghai Province.Methods:In July 2018, according to the "Diagnosis of Kashin-Beck Disease" (WS/T 207-2010), in KBD endemic region of Xinghai and Guide counties in Hainan Tibetan Autonomous Prefecture of Qinghai Province, adult KBD patients and healthy people over 20 years old were selected as KBD group and internal control group, respectively. At the same time, healthy people over 20 years old were selected as the external control group in non-KBD endemic region of Xunhua County with similar production scale and lifestyle. Fasting elbow vein blood samples of the 3 groups of people were collected, serum NPT levels were determined by enzyme linked immunosorbent assay (ELISA).Results:Totally 272 people were enrolled in the study, 104 cases (50 males and 54 females) in KBD group, aged (47.61 ± 12.72) years old; 95 cases (35 males and 60 females) in internal control group, aged (48.28 ± 14.87) years old; and 73 cases (35 males and 38 females) in external control group, aged (51.88 ± 13.93) years old. There were no significant differences in gender composition and age among the 3 groups (χ 2 = 3.135, F = 2.236, P > 0.05). The serum NPT levels of KBD, internal control, and external control groups were (504.35 ± 413.92), (417.34 ± 109.90) and (397.49 ± 118.07) ng/L, respectively, and the difference among the 3 groups was statistically significant ( F = 4.129, P < 0.05). Among them, the serum NPT level of KBD group was significantly higher than that of internal control group and external control group ( P < 0.05); there was no significant difference in the serum NPT level between internal control group and external control group ( P > 0.05). Conclusion:The serum NPT level of adult KBD patients in Qinghai Province is elevated.
