Objective To investigate the feasibility of laparoscopic operations in treating large ovarian cysts. Methods A total of 38 patients with large ovarian cysts underwent laparoscopic cyst cystectomy or adnexectomy on the affected side from December 2002 to October 2005.The 2nd or 3rd trocar(usually on the tumor side)was inserted into the cyst, the fluid in which was drained with a suction unit. The cyst was then removed after being elevated toward the incision, which was extended to 20 mm.Results Large ovarian cysts in the 38 cases were all removed successfully, including cystectomy in 28 cases and adnexectomy in 10 cases respectively. No complication occurred. The follow-up period was 1-24 months(mean:10 months)for 35 cases. No abnormal findings in pelvic examination and B ultrasonography were observed. Conclusions Laparoscopic operation is a safe and feasible therapeutic method for large ovarian cysts.

Objective To explore the feasibility and to improve the efficiency of testing circulating tumor cells(CTC)in patients with renal cell carcinoma by Cell-search System(CSS).Methods Eight patients with renal cell carcinoma hospitalized in the PUMC urology department for further clinical evaluation in Jan.to Jun.2012 were enrolled in this study. There were 5 males and 3 females with mean age of 64 (57-70)years.There were 2 cases in clinical stage T3N0M0 and 6 cases in T4N0M1;3 cases were treated with radical nephrectomy,5 cases were treated with targeted therapy drugs.7.5 ml peripheral blood samples from these patients were collected and saved in test tube at the temperature of 15 ℃-30 ℃.The tests had to be done within 96 hrs after the blood sample drawn. We selected fluorescent anti-CKS/18/19 antibodies as CTC makers for renal cell carcinoma. Then the number of CTC was quantitative detected by CSS.The feasibility and detection rate of testing CTC in patients with RCC by CSS was evaluated, and the improvement of method was discussed. Results Quantitative test results of CTC in 8 patients with renal cell carcinoma were negative.25 CK+CD45 + double positive cells were found in one T4N0M1 patient's peripheral blood. The detection rate of the CTC in TNM stage T3N0M0 and T4N0M1 renal cell carcinoma patients using CSS with epithelial tumor cell maker anti-CK8/18/19 was zero. Conclusions Anti-CK8/18/19 antibodies can not be used in testing circulating tumor cells in patients with renal cell carcinoma by Cell-search System.

Objective To investigate the application effects of new-type glove constraint in intensive care unit. Methods A total of 113 eligible patients were randomly divided into two groups by random digits table method, of which 59 patients used the traditional restraint for body constraint in the control group while 54 patients applied the new-type glove constraint in the treatment group. The skin conditions of restraint position, tube coming off and the satisfaction of patients and relatives were compared between two groups. Results The incidence of skin abnormality at restraint position was 0 in the treatment group and 13.6%(8/59) in the control group. The difference had statistical significance (χ2=7.880, P<0.01). The satisfaction of patients and relatives was 33.3%(18/54) in the treatment group and 5.1%(3/59) in the control group, and the difference had statistical significance (χ2=16.619, P<0.01). Conclusions The use of new-type glove constraint may give a maximum activity space to patients;reduce patient′s psychological adverse impacts;and decrease the skin and body damages. It is worthy of clinical promotion and application.

Journal of Medical Research 2006;0(04):-
Objective To investigate the effects of variable sample preparation about sialy LewisA/X on the surface of colon carcinoma cells. Methods Human colon carcinoma cell suspension was prepared by using EDTA,trypsin, cell scraper,trypsin-EDTA and trypsin-cell scraper.Cell counts and sialy-LewisA/X on the surface of colon carcinoma cells were detected by flow cytometry. Results Trypsin made fluorescence intensity of sialy-LewisA/X on the surface of colon carcinoma cells become weaken.Fluorescence intensity of sialy-LewisA/X had no significant deviation among EDTA,cell scraper and trypsin-cell scraper.Cell counts were multitude in EDTA group or trypsin-cell scraper group. Conclusion Trypsin-cell scraper is a available approach on preparing cell suspension of adherent cell.

OBJECTIVE:To observe the clinical efficacy and safety of different doses of atorvastatin in the treatment of chron-ic subdural hematoma(CSDH). METHODS:One hundred and tweaty-eighie CSDH patients selected from our hospital during Jun. 2013-May 2015 were divided into observation group(n=62)and control group(n=66)in accordance with random number table. Both groups were given conventional treatment of brain cell nutrition. Control group received Atorvastatin tablet 20 mg,po,qd;observation group received Atorvastatin tablet 40 mg,po,qd. Both groups were treated for 6 months. Clinical efficacy,CSS and ADL score, hematoma volume, the levels of serum inflammatory factors (hs-CRP,MMP-9,IL-6,TNF-α) were observed in 2 groups. ADR was recorded during treatment and recurrence rate was also recorded. RESULTS:2 patients withdrew from observa-tion group and 6 from control group. Finally,120 patients met the criteria were included, with 60 cases in each group. Total re-sponse rate of observation group(88.3%)was significantly better than that of control group(73.3%),with statistical significance (P<0.05). After 1,3,6 months of treatment,CSS score, hematoma volume, the serum levels of hs-CRP,MMP-9,IL-6 and TNF-α in 2 groups were significantly decreased, while ADL score was increased significantly;the improvement of above indexes in observation group was significantly better than in control group, with statistical significance(P<0.05). There was no statistical significance in the incidence of ADR between 2 groups(P>0.05). The recurrence rate of control group(13.3%)was significantly higher than that of observation group(3.3%),with statistical significance(P<0.05). CONCLUSIONS:Daily dose of shows ator-vastatin 40 mg better therapeutic efficacy and lower recurrence rate in the treatment of CSDH with good safety.

Objective To investigate the cytokine spectrum of cultured human amniotic-derived mesenchymal stem cells (A-MSC) for understanding its basis of molecular biology in hematopoietic in vitro. Methods Their hematopoietic cytokines expression was analyzed using RT-PCR in the mRNA level. Results It showed that in vitro subcultured human amniotic-derived mesenchymal stem cells were capable to express many important hematopoietic cytokines such as LIF, SCF, M-CSF, G-CSF, GM-CSF, IL-3, IL-6, IL-11 and so on. Conclusion Production of abundant of hematopoietic cytokines by human amniotic-derived mesenchymal stem cells may be effective for hematopoietic support and HSC transplantation.

Journal of Regional Anatomy and Operative Surgery 2015;(5):479-483
Objective To investigate the relationship between hypoxia and stemness of cancer stem cells ( CSCs ) . Methods U87 cells,U251 cells and primary glioma cells were experienced hypoxia. Detected the ultrastructure of these cancer cells with transmission elec-tron microscopy;detect the cell growth with MTT assay;cell cycle and CD133 expression were detected by flow cytometry;and the cell mi-gration ablity were detected through transwell chamber assay;the colony-forming efficiency were deteced by colony-forming assay; and real-time quantitative PCR and Western blot were carried out to detect the mRNA and protein expression of markers of stem cells and their differ-entiation respectively. Results Hypoxia maintained the undifferentiated state of primary glioma cells, slowed down the growth of glioma cells which were in a relatively quiescent stage, increased the colony forming efficiency and migration of glioma cells, and increased the expression of markers of stem cells, but the expression of markers for stem cell differentiation was reduced after hypoxia treatment. Conclusion Hypoxi-a may induce the “dedifferentiation” of differentiated glioma cells which then acquire the stemness.
