Objective To study the clinical features of primary biliary cirrhosis (PBC) in order to improve the clinical diagnosis. Methods Clinical data of fifty patients with PBC were retrospectively studied. The clinical features, results of laboratory tests, pathological findings of all the fifty patients were analyzed. Results forty-four patients were female (88%), and the mean age was 44.94?12.33 years at the time when definite diagnosis was made. The most frequent symptoms complained were fatigue (34/50, 68%), anorexia (24/50, 48%) and pruritus (13/50, 26%).The major signs included jaundice (32/50, 64%), splenomegaly (24/50, 48%) and liver palms (14/50, 28%). Serum alkaline phosphatase (ALP) and r glutamyl transpeptidase (r-GT) levels were markedly elevated in most patients, and the levels of ALP and GGT declined during the advanced stage. The levels of ALT and AST were elevated slightly. The total level of serum bilirubin was elevated in thirty-two patients (64%). Positive anti-mitochondrial antibody (AMA)/AMA-M2 were found in forty-one patients (82.0%). Pathological examination was done in 32 patients, it was shown that 28 patients (87.5%) were in early stage and 4 patients (12.5%) in advanced stage. Conclusion PBC occurs mainly in middle- and old aged women. The main clinical manifestations are fatigue, anorexia and pruritus. The elevated level of ALP, r-GT and positive AMA/AMA-M2 may be crucial to diagnosis of PBC.

Microbiology 1992;0(06):-
N-carbamoylase is a part of hydantoinase operon which can transform N-carbamoylamino acid to corresponding ammo acids. The L-N-carbamoylase of Arthrobacter BT801, codied by the HyuC gene, is the rate-limiting and the only stereoselective enzyme. HyuC DNA fragment was amplified by PCR from the plasmid of pUC18-169. The target fragment was introduced into pPIC3. 5K plasmid to construct the pPIC3. 5K-hyuC expressing vector which was then transduced into Pichia pastoris GS115 cells after being linearized by BglⅡ digestion. Multi-copies insertion transformants were screened on G418 plates. The recombinant protein was proved to have biological activity of hydrolyzing N-carbamoylphenylalanine into phenylalanine through enzyme activity determination.

Objective To assess the radiofrequency ablation (RFA) lesions in canine prostate with real-time harmonic gray-scale contrast-enhanced ultrasonography and investigate the feasibility to predict these lesions. Methods A total of 5 male beagle dogs took RFA of the prostate.Radiofrequency (RF) was delivered under transrectal ultrasound (TRUS) guidance.A lesion was created in the right and left lateral lobes of canine prostate, respectively. The examination of conventional gray-scale ultrasonography, power Doppler ultrasonography,real-time harmonic gray-scale contrast-enhanced ultrasonography were performed immediately after RF procedure. All animals were subsequently sacrificed and the prostates were harvested for pathologic analysis.Thermal lesion volume was calculated based on expected shape of ellipsoid using equation. The volumes of the calculated thermal lesion on contrast-enhanced ultrasonography in dogs were compared with the volumes obtained from the pathological specimens. Results The RFA lesions could not be imaged accurately with real-time conventional gray-scale or power Doppler ultrasonography.However,contrast-enhanced low MI perfusion imaging was able to have real-time monitoring for the lesion (no blood flow). The volume of lesions was measured with contrast-enhanced ultrasonography without significant difference with those of pathologic results.Ultrasonic imaging(conventional gray-scale or power Doppler sonography)of the ablation areas also had its own characteristics. Conclusion The real-time harmonic gray-scale contrast-enhanced ultrasonography appears to be an accurate modality for assessment of RFA lesions.Conventional gray-scale and power Doppler sonography had their own characteristics to evaluate the RFA lesion.

Objective To explore whether acute graft versus host disease (aGVHD) could be alleviated by syngeneic bone marrow mixed with granulocyte colony stimulating factor (G CSF) mobilized H 2 haploidentical marrow grafting. Methods Female BALB/c and neonatal BALB/c mice were recipients and male (BALB/c?C 57 BL/6) F 1 (BCF 1) mice were donor mice respectively. Donor mice were injected subcutaneously with G CSF daily at 0.01 ?g/g body weight or saline for 6 days, and splenocytes were harvested on day 6. Spleen index(SI) represented GVHD in neonatal mice after the intraperitoneal injection of mixed spleen cells.Lethally irradiated ( 60 Co, 8.5 Gy) adult mice were transplanted with a mixture of syngenetic plus G CSF mobilized (control diluents) H 2 haploidentical marrow cells.Survival time and survival rate of the recipients were observed after mixed marrow transplantation (MBMT). GVHD was assessed by observing signs of weight loss, ruffled fur, diarrhea and histological change of skin, liver and small intestines. Enzyme linked immunosorbemt assay (ELISA) method was used to detect cytokines (IL 2, IL 4, IFN ?). Fluorescence activated cell sorting (FACS) analysis was used to detect T cell phenotype. Results (1) The neonatal mice subject to injection of 2∶1 and 1∶1 mixed spleen cells and H 2 haploidentical spleen cells all suffered from aGVHD. The severity of aGVHD in recipient mice receiving G CSF mobilized splenocytes was dramatically reduced. (2) The aGVHD signs and histological change were observed in most mice of 2∶1 and 1∶1 MBMT groups. However, the survival time of G CSF mobilized MBMT groups was longer than in control groups ( P
