1.A novel amorphous calcium phosphate drug-eluting stent:its biocompatibility after implantation into mini-swine coronary artery

BACKGROUND:As previously reported, poly-L-lactic acid stents have been mostly used as coronary artery stents, but this stent has a lack of early support force and produces some acidic products that wil induce local vascular inflammatory reactions.
OBJECTIVE:To explore the biocompatibility of a novel amorphous calcium phosphate drug-eluting stent, after implantation into mini-swine coronary artery.
METHODS:Twenty mini-swine were randomly equivalently divided into observation group and control group. The right femoral arteries of animals were separated layer by layer and the new type of biodegradable stent and poly-L-lactic acid stent were implanted, respectively.
RESULTS AND CONCLUSION:The vessel lumes in the two groups were smooth and unobstructed showed by coronary artery angiography immediately after implantation. At 4 weeks after implantation, the coronary angiography showed the patency of vessel lumens with no stent thrombosis and stenosis in both two groups. The C-reactive protein levels in the two groups had no significant differences before and 4 weeks after stent implantation. At 4 weeks after implantation, the number of lymphocytes and inflammatory score in the observation group were significantly higher than those in the control group, but the neointimal area and percentage of stenosis area had no significant difference between the two groups. These results indicate that the novel amorphous calcium phosphate drug-eluting stent after implantation into mini-swine coronary artery has good biocompatibility.
2.Changes of Astrocyte in the Cultural Model of Amyotrophic Lateral Sclerosis

Objective To establish the model of amyotrophic lateral sclerosis (ALS) with selective motor neuron disorder by organotypic spinal cord cultures, and analyze the role of astrocyte in the pathagenisis of ALS.Methods Organotypic spinal cord cultures were prepared using lumbar spinal cord slices from 8-day-old SD rat pups. The threohydroxyaspartate (THA) was applied into culture medium to establish ALS organotypic spinal cord cultures model. Motor neurons survival was evaluated by monoclonal SMI-32 immunohistochemical staining and glial fibrillary acidic protein staining to show astrocyte survival.Results Compared with the control group, there was significantly astrogliosis in the anterior horn and surrounding white matter in THA 100 μmol/L group, and the level of gliosis was increased followed the elongation of THA interference time. With the increasing of the number of astrocyte, the morphology of astrocyte was changed.Conclusion There is significantly astrogliosis in the anterior horn and the time of astrogliosis is markedly earlier than the time of motor neuron loss in the ALS model intervented with THA.
3.18F-FDG PET/CT features of spinal tuberculosis

Objective To investigate the diagnostic value of 18F-FDG PET/CT features of spinal tuberculosis.Methods 18 F-FDG PET/CT imaging manifestation of 125 cases (70 males,55 females,average age 44.3 years) with osseous or mixed vertebra bone destruction from January 2006 to June 2014 were analyzed retrospectively.The patients included 32 cases with spinal tuberculosis (50 lesions,study group) and 93 cases with other diseases (150 lesions,control group).The corresponding FDG PET/CT features were analyzed by two classification logistic regression (Forward:LR),then FDG PET/CT features with statistical significance in diagnosis of spinal tuberculosis were obtained.The sensitivity and specificity of those FDG PET/CT features were calculated.Results Odds ratio (OR) of FDG PET/CT features with statistical significance in diagnosis of spinal tuberculosis from high to low were paraspinal cold abscess (20.790),cold zone(10.528),intervertebral disc involvement(5.394),continuous vertebral involvement(3.493).The diagnostic sensitivity and specificity were 22.0% (11/50),99.3% (149/150) forcold abscess,70.0% (35/50),90.0% (135/150) for cold zone,82.0% (41/50),83.0% (125/150) for intervertebral disc involvement,82.0% (41/50),78.0% (117/150) for continuous vertebral involvement,respectively.Spinal tuberculosis often had two or more signs.The continuous vertebral involvement + intervertebral disc involvement was most common,with the diagnostic sensitivity and specificity of 78.0% (39/50) and 88.7% (133/150).The diagnostic sensitivity and specificity of continuous vertebral involvement+intervertebral disc involvement+cold zone were 58.0% (29/50),96.0% (144/150),and those of four features conourrence were 14.0% (7/50),100% (150/150),respectively.Conclusion Regression analysis shows that FDG PET/CT features with statistical significance in diagnosis of spinal tuberculosis are paraspinal cold abscess,cold zone,intervertebral disc involvement,continuous vertebral involvement,which have high diagnostic sensitivity and specificity.
4.The impact of smoking on C-reactive protein level in gingival crevicular fluid before and after initial treat-ment of patients with periodontitis

Objective:To study the impact of smoking on C-reactive protein(CRP)concentration in gingival crevicular fluid(GCF) before and after initial treatment.Methods:18 smokers and 18 non-smokers with moderate or severe periodontal disease were recrui-ted into this study.The clinical indexes of periodontal examination of the patients were recorded,GCF samples of the patients were collected,CRP level in the samples was measured by radioimmunoassay balance method before and 4 weeks after initial treatment. Results:4 weeks after initial treatment,the clinical indexes and CRP concentration in GCF of the smoking group and non-smoking group were significantly lower than those before treatment(P <0.05),the changes of the smoking group were less than those of the non-smoking group(P <0.05).Conclusion:Smoking is an unfavorable factor of the initial periodontal therapy.

Hyperlipidemic pancreatitis (HLP) has an increasing incidence.Primary HLP is common in patients with familial difficiency of lipoprotion lipase or apo-C Ⅱ.While for secondary HLP,the common reasons are excessive drinking,diabetes,obesity,fat-rich diet and special medications.Lipidlowering drugs,plasma exchange (PE) or continuous veno-venous hemofiltration (CVVH),low molecular heparin as well as insulin,have been widely used for the treatment of HLP.Changing of diet and lifestyle,in addition to controlling risk factors are the keys to preventing HLP.This article summarizes the latest perspectives and research progress of HLP.

Objective To explore the value of electronic colonoscope,MSCT and MRI in the etiological diagnosis of intestinal obstruction.Methods The clinical data of 140 ileus patients were retrospectively studied.Results 140 cases of ileus included 60 cases of colon ileus,70 cases of small intestines ileus and 7 indefinite cases.67 cases were checked with colonoscopy,among them,41 cases with colon tumors,6 cases with colon polypi,6 cases with colon set folds,2 cases with sigmoid colon turned round,2 cases with ileum tumor and 10 cases with unknown reason.80 cases were checked by MSCT checks,among them,26 cases with small intestines tumor 24 cases with small intestines muck piece,23 cases with unknown reasons,2 cases with muck stone 2,4 cases with bowel chamber outside lymphoid lump,1 case with bowel fastens film blood to bolt.30 cases were kept to cure.2 cases were turn round to reset under the eolonoscopy,3 cases were set folds air to infuse bowel to reset,6 cases with inside the mirror descend colon polyp were resected with hish frequency electricity resection.99 cases were cured by surgical operation.5 cases were dead and 135 cases were recovered from illness.Conclusion Electronic colonoscopy,MSCT and MRI checking are accurate methods to diagnose why and where of ileus,and which establish importance basts for clinical treatment.

Objective To study more significant detective combination by comparing three methods of antinuclear antibody,extractable nuclear antigen and antinuclear antibodies 3.Methods Three methods were adopted as follows:detecting antinuclear antibody with HEP-2 and prime hepar as antigen media,determining extractable nuclear antigen by EUROASSAY including nRNP/Sm、Sm、SSA、SSB、Scl-70 and Jo-1,using crithidia iuciliae as antigen,to detect ds-DNA antibody with indirect immunofluorescence assay,and to study 15 autoantibodies by EUROLINE.Results To compare positive rates of three dilution(1:10,1:80,1:100)in ANA detection,there existed significant difference(P<0.01)between two dilution of 1:10 and 1:80 only,.In the detection of multiple nuclear types(cytoplasmic pattern,centromere pattern and homegeneous pattern etc),the positive rate of antinucleatr antibodies 3 was higer than that of ENA.IIF was prior to the EUROLINE in the detection of ds-DNA.Conclusion During detecting ANA with IIF,the dilution of 1:80 has advantages in raising positive rate,and ANA3 of 15 antigens provides facilities for clinical diagnosis.
9.In vitro Transdermal Absorption of Xiaochuan Ointment

Objective:To study the transdermal absorption in vitro of sinapine thiocyanate,tetrahydropalmatine and asarinin in Xiaochuan ointment so as to offer reference for its clinical application.Methods:A Franz diffusion cells method with isolated rat skins was carried out to study the percutaneous rate of sinapine thiocyanate,tetrahydropalmatine and asarinin determined by LC-MS/MS.Results:With the increase of administration dosage,the cumulative penetration of sinapine thiocyanate,tetrahydropalmatine and asarinin showed few changes.The results showed that the transdermal behavior of sinapine thiocyanate fit to a Higuchi equation,and that of tetrahydropalmatine and asarinin fit zero-order process.The penetration rate of tetrahydropalmatine and asarinin respectively was 0.362×10-1 and 0.330×10-2 μg·cm-2·h-1.Conclusion:Xiaochuan ointment exhibits transdermal penetration and absorption properties,which provides evidence for its transdermal administration.
10.Expression,Purification of Fusion Protein TGF?-PE40 and the Cytotoxicity of TGF?-PE40 on Tumor Cells

Objective: To express and purify transforming growth factor ?(TGF?)-pseudomonas exotoxin 40 and investigate its cytotoxic effect on cancer cells overexpressing epidermal growth factor (EGF) receptor. Methods: Recombi nant plasmid pV28 was constructed by inserting the gene coding TGF?-PE40 into the vector pET28a Expression of fusion protein was conducted using the host BL21. Production of the recombinant protein was induced by IPTG, following extraction and purification of inclusion bodies with His-tag purification system. Cell viability assay (by MTT) was performed to determine the cytotoxic effect of TGF?-PE40 on cancer cells ( A431 and SK-OV3). Results: Recombinant plasmid pV28, which expresses TGF?-PE40, was constructed successfully. Purity of TGF?-PE40 was about 98% after a purification procedure using His-tag column. Cytotoxic experiment showed that at a concentration of 0. 86?0. 07 UUUUg/ml, TGF?-PE40 could reduce 50% viability of A431, which has high expression of EGFR.
