Objective To verify the performance of the ADVIA Centaur XP automatic chemiluminescence analyzer produced by the Siemens company in quantitative detection of serum insulin (IRI) and C‐peptide(CpS) .Methods The blood samples from 40 in‐dividuals undergoing the physical examination were detected for calculating the imprecision and bias of IRI and CpS .The linear range of IRI was verified .The reference range of IRI and CpS was verified .Results The intra‐batch variable of coefficient of IRI and CpS was less than 6 .25% ,the inter‐batch imprecision were less than 8 .33% ;the bias of IRI and CpS detection was less than 12 .5% ;the linear range of IRI was 0 .50 - 307 .76 mIU /L ;the detection results of 40 serum samples showed that all IRS were with‐in the reference range provided by manufacturer ,the results in 2 cases of CpS was out of the reference range provided by manufac‐turer ,indicating that it conformed to the relevant requirements .Conclusion The main performance of the ADVIA Centaur XP auto ‐matic chemiluminescence analyzer for detecting IRS and CpS meets the requirements ,which could be used for clinical detection .

ObjectiveTo sum up the diagnosis and differential diagnosis of concealed penis,analyze the indications and advantages of different operative methods for concealed penis.Methods62 children with concealed penis were treated by different surgical operations according to the different types,and the effect of surgical treatment was arelyzed.ResultsOpertaions were successed in all 62 cases,children were discharged after 5 ～ 10 operation days.After follow-up for 3 months to 2 years,62 children had received good recovered with an effective exposure of penis and no severe edema and other complications.ConclusionChildren with concealed penis should be given personalized scheme,choose the best method to extend the length of the penis and improve the penis appearance.

[Objective]To investigate the effects of alendronate(ALN)on subchondral bone and articular cartilage in the rat osteoarthritis model induced by anterior cruciate ligament transaction(ACLT).[Method]Twenty-four three-month-old female Sprague-Dawley rats were divided randomly into three groups:Sham group(n=8),ACLT+NS group(n=8) and ACLT+ALN group(n=8)(20 ?g?kg-1?2 d-1 subcutaneous injection for 12 weeks).Bone mineral density(BMD) measurement and bone histomorphometric analysis were made for the subchondral bone of right distal femur and proximal tibia in all rats at 12 weeks post surgery.Degree of cartilage degeneration was scored by Mankin scoring system.Immunostaining for matrix metalloproteinase 13(MMP-13) was performed to investigate the effect of ALN on cartilage matrix loss.[Result]BMD of the subchondral bone in the ACLT+NS group were significantly lower than those in the Sham group and the ACLT+ALN group(P

OBJECTIVE: To establish miniSTR fluorescent detection system with all detected fragments below 150 bp and to enhance the efficiency of detecting the degraded DNA samples. METHODS: All candidate primers were designed by Primer Premier 5 and screened by FastPCR 6.0. The miniSTR multiplex system was established by these selected loci labeling by four fluorescent dye. The parameters of PCR and primer concentrations were subsequently optimized. The electrophoresis was fulfilled under POP4 on 3100-Avant and the typing data was validated by standard DNA 9947A and 007. Fresh blood samples and difficult degraded DNA samples were tested to evaluate the usefulness of the system. RESULTS: All amplicons in the established miniSTR fluorescent detection system (D12ATA63, D2S1776, D1GATA113, D4S2408, D17S974, D20S482, D3S3053, Amelogenin, D6S474, D9S1122) were less than 150bp. The profile showed a balanced peak height without extra stutter by optimal protocol. Allele frequencies showed no deviations from Hardy-Weinberg equilibrium. The system showed accumulated probability of discrimination 0.999 999 983 and accumulated triplet excluding probability of paternity 0.996 8. It could detect corrupt muscle tissue, low copy number DNA samples and human tissues fixed by 40% formaldehyde solution for 12 days. CONCLUSION The miniSTR fluorescent detection system could be solely used for personal identification of degraded DNA samples or complementally used for paternity tests. And the system could enhance the ability of detecting the trace and degraded DNA.

OBJECTIVE: To evaluate the relative factors influencing olfactory dysfunction in patients with chronic rhinosinusitis (CRS). METHOD: Visual analogue scale (VAS) was applied to measure the severity of olfactory dysfunction of 270 patients with CRS. Patients were divided into two groups, one was that the quality of life (QOL) of patients was affected by olfactory dysfunction (VAS > 5), the other was that without QOL affected by olfactory dysfunction (VAS ≤ 5). The association between age, gender, nasal polyps, allergic rhinitis, smoking history, early nasal surgery history and other clinical factors, and serum total IgE level, peripheral blood eosinophil count, peripheral blood mononuclear cell count and olfactory dysfunction was analyzed. RESULT: The number of patients with nasal polyps, allergic rhinitis, previous nasal surgeries, the level of serum total IgE, and the severity of edema were significantly increased in patients with impaired QOL associated with olfactory dysfunction (P < 0.05). Sex distribution, age, smoking history, deviation of nasal septum, eosinophil and mononuclear cell count did no statistically differ between the groups with and without impaired QOL associated with olfactory dysfunctions (P > 0.05). Serum total IgE increased (OR = 1.003, P < 0.01) and severe edema (OR = 2.483, P < 0.01) were the risk factors for the impairment of olfactory function, more notably for edema; whereas previous nasal surgeries was a protective factor (OR = 0.408, P < 0.01). CONCLUSION Sever edema and increased serum total IgE are risk factors, whereas previous nasal surgeries history is a protective factor for the olfactory dysfunction.

[Objective]To assess the protective effects of Xianlinggubao on articular cartilage and subchondral bone in experimental osteoarthritis induced by anterior cruciate ligament transaction (ACLT)in rabbits. [Methods]Thirty healthy Japanese white rabbits were divided randomly into three groups:sham group,ACLT+XLGB group and ACLT+ NS group. Rabbits in ACLT+XLGB group and ACLT+NS group received anterior cruciate ligament transection(ACLT)on the right knee. Rabbits in sham group received untreatment as control. ACLT+XLGB group received a daily administration of XLGB at a dose of 125 mg/kg of body weight for 6 weeks.In contrast,ACLT+NS group received saline at the same dose. All rabbits were sacrificed at 6 weeks after operation. The right femora were removed,measured for bone mineral density (BMD),and then were observed for the macro-pathologic changes with HE staining and graded by Mankin's scale. Expression levels of BMP-2 and MMP-13 were detected by immunostaining. Objective To evaluate the effect of ulinastatin on hepatic ischemia-reperfusion injury in patients with hepatic alveolar echinococcosis in plateau region.Methods Sixty patients with hepatic alveolar echinococcosis in plateau region,weighing 45-75 kg,aged 30-55 yr,of American Society of Anesthesiologists physical status Ⅰ or Ⅱ,scheduled for elective partial hepatectomy,were divided into control group (C group) and ulinastatin group (U group) using a random number table,with 30 patients in each group.Ulinastatin 12 000 U/kg (in 100 ml of normal saline) was infused intravenously at 20 min before occlusion of hepatic portal in group U,and normal saline 100 ml was given instead in group C.Before operation,at 10 min after occlusion of hepatic portal,at 20 min and 1 h after unclamping hepatic portal and at 12 h after operation,blood samples were collected from the right internal jugular vein for determination of the levels of serum alanine aminotransferase,aspartate aminotransferase and malondialdehyde.Results Compared with group C,the levels of serum alanine aminotransferase,aspartate aminotransferase and malondialdehyde were significantly decreased at 20 min and 1 h after unclamping hepatic portal and 12 h after operation in group U (P＜0.05).Conclusion Ulinastatin can attenuate hepatic ischemia-reperfusion injury in patients with hepatic alveolar echinococcosis in plateau region.

AIM　To investigate the therapeutic effect of boanmycin (BAM, bleomycin A6) on colon carcinoma 26 and its hepatic metastasis in mice. METHODS　A series of models including subcutaneous transplantation, orthotopic transplantation in cecum subserosa, intra-hepatic transplantation of tumor, and intra-splenic transplantation of tumor accompanied with hepatic metastases were employed. In addition, LEICA Q 500IW image analysis system was used to determine the area of metastatic lesions in the liver in histopathological sections. RESULTS　BAM at 5 mg*kg-1 and 2.5 mg*kg-1 inhibited the growth of subcutaneous tumors by 78.7% and 61.9%, and the orthotopic tumors by 99.4% and 90.0%; and the intra-hepatic tumors by 86.9% and 75.7%, respectively. Determined by the numbers of metastatic nodules, BAM at 10 mg*kg-1 and 5 mg*kg-1 inhibited hepatic metastases from intra-splenic transplantation by 97.6% and 56.8%; and evaluated by image analysis of metastatic lesions, by 100% and 63.3%, respectively. CONCLUSION　Boanmycin is highly effective in a panel of models including the subcutaneous, orthotopic, intra-hepatic transplanted tumors, and hepatic metastases of murine colon carcinoma 26.

Objective To observe the characteristic, mechanisms and the change of pathophysiology about thoracic explosive injury from an explosive device. Methods Seventy-three millimetres prefabricated steel-ball bomb was used to be the center of explosion. According to the distance from each group to the center of explosion, 30 sheep were randomly divided into 5 groups. The injuries of thoracic wall, lung and surrounding organs were observed from fragments and blast wave. Results The mortality of thoracic explosive injury on the ground testing was 56.7%.Most of fragment wounds were blind wound. The track of wounds was often complicated and curved. The major associated injuries were myocardial contusion, splenic-hepatic rupture and gastric-enteric rupture. There were much severe pulmonary hemorrhage and lung edema. The parameters of trauma were well correlated with the severity of thoracic injury.　Conclusion The parameters of trauma were well correlated with the severity of thoracic injury. The characteristic of thoracic injury from explosive device was more complicated and severe.
