1. Youchasaponin induces apoptosis of human leukemia Jurkat cells in vitro and its possible mechanism

Objective: To investigate the apoptosis of human leukemia Jurkat cells induced by Youchasaponin in vitro and to explore its possible mechanism. Methods: The effects of Youchasaponin with different concentrations on the proliferation of Jurkat cells were detected by cell count assay. The cell cycle distribution and the apoptosis rate of Jurkat cells were determined by flow cytometry (FCM). The expression levels of caspase-3, poly (ADP-ribose) polymerase (PARP), p-Bcl-2, Bcl-2, Bax and caspase-9 proteins were analyzed by Western blotting. Results: The proliferation of Jurkat cells was significantly inhibited after treatment with Youchasaponin (1-16 μg/mL) in a dose-dependent manner. The percentage of Jurkat cells at G0/G1 phase and G2/M phase was decreased after treatment with Youchasaponin (1-4 μg/mL) for 24 h, while the percentage of Jurkat cells at S phase was increased. The apoptosis rate of Jurkat cells induced by Youchasaponin was increased in a dose-dependent manner. The expression levels of p-Bcl-2 and Bcl-2 proteins were down-regulated, and the expression levels of caspase-3, PARP and caspase-9 proteins were up-regulated, whereas the expression level of Bax protein had no change. Conclusion: Youchasaponin can inhibit the cell proliferation and induce the apoptosis of human leukemia cells. This effect may be related to the mitochondrial pathway of apoptosis. 2.Cytotoxicity of Human Tumor Draining Lymph Node Cells Stimulated by Cancer-Associated Apomucin in vitro

Human tumor antigen elicit a cell-mediate immune response in the host have been identified. In this study, a pancreatic cancer cell line SW1990 associated apomucin was examined for the induction of tumor draining lymph node cells(TDLN) anti-tumor immune responses.The TDLN of patients with breast cancer(4 cases), gastric cancer(3 cases), colon cancer(3 cases) were isolated, cocultured with apomucin 50 ?g/ml and low dose rhIL-2 50U/ml in vitro, the cytolytic effects were measured by MTT assay. The results showed that: the proliferated lymphocytes can lysis LS174T colon cancer cells, KATO-Ⅲ gastric cancer cells, SW1990 pancreatic cancer cells and K562 leukemia cells. The most cytolytic effects were higher than LAK. This results might provide a rational basis for future attempts to elicit and manipulate apomucin vaccine for active specific immunotherapy or adoptive immunotherapy for adenocarcinoma.

4.Correction of severe kyphosis and kyphoscoliosis by total spinal osteotomy

[Objective]To explore the experience of the correction of severe kyphosis and kyphoscoliosis by total spinal osteotomy.[Method]To correct kyphosis and kyphoscoliosis by total spinal osteotomy and screws-rods internal fixation apparatus,then graft the bone.[Result]From 1984 to 2005,50 patients were cured with the way above.There were no neural injury observed.The clinical symptom was improved in some extent.All patients were follow up from 2 months to 15 years,averaged 2.4 years,and the postoperative X ray and CT scans verifies all screws were in a proper position.No cases of implant failure were observed,and all the patients had a solid bone fusion on radiographs taken 3 months after surgery.[Conclusion]The operation to correct severe kyphosis and kyphoscoliosis by total spinal osteotomy with efficient screws-rods internal fixation apparatus,not only have a larger operate area which made opreation easier and more safe,but combine osteotomy with internal fixation and finished at the same time,it is an efficient way to correct kyphosis and kyphoscoliosis.
5.Pretreatment and Determination of Dissolvable Zinc Salt in Deodorizing Cosmetics

Objective To explore a method for determination of dissolvable zinc salt in deodorizing cosmetics. Methods The contents of dissolvable zinc salt in deodorants extracted by pure water were detected by flame atomic absorption spectrophotometry. Results The correlation coefficient of the standard curve was more than 0.999. The detection limit, precision and recovery rates of the method were 8.2?10-3 ?g, 1.26% and 97.0%~98.5% respectively. The lowest detection limit of zinc salt was 2.7?10-2 ?g. No other interfering factors were found during detection process. Conclusion This method could meet the requirements of hygienic physico_chemical detection for cosmetics.

10.Application of micro transesophageal echocardiography (micro-TEE) in neonat and infant cardiac surgery

Objective To evaluate the value of phased array probe of transesophageal echocardiography miniature (micro-TEE) in the neonatal and infant cardiac surgery intraoperation.Methods Micro-TEE probe was used in children with weight ≤5 kg undergoing cardiac surgery to detect the atrial and ventricular cavity size,the left and right ventricular outflow tract diameter were detected by transthoracic echocardiography(TTE) 3-7 d after operation,and compared with the measured value immediately after the TEE.Results Before and after intubation,airway index not statistically significant (P > 0.05).Preoperative TEE and TTE inspection results were consistent with intraoperative diagnosis.Postoperative TEE measuring intracardiac structural data (each room,chamber size and left and right ventricular outflow tract diameter) and postoperative TTE measurement value representing the difference was not statistically significant (P > 0.05).Conclusions Micro TEE intraoperative guardianship of low birth weight infants is safe and feasible,and can supplement and improve the preoperative diagnosis,and can accurately and timely evaluate postoperative curative effect.
