Objective To evaluate the effects of propofol and thiopontal on calcium and potassium channels in rat ventricular myocytes and to elucidate the underlying mechanisms of their inhibitory effect on myocardium. Methods Freshly isolated ventricular myocytes were prepared from hearts of rats by trypsin. The effects of propofol and thiopental on L-type calcium current(Ica) and delayed rectifier potassium current(IK)were compared using whole-cell patch clump technique. Results Propofol and thiopontal produced a concentration-dependent inhibition of Ica. Peak concentration of propofol(50 ?mol?L~(-1)) and thiopental(100 ?mol?L~(-1)) during induction of anesthesia decreased Ica by 28% and 46% and shifted the steady-state inactivation curve to more negative voltage, but had no effect on the steady-state activation curve. Propofol and thiopental also decreased IK in a concentration-dependent manner, but the effects of both anesthetics on IK were smaller compared with their effects on Ica. Conclusion The findings of this study suggest that the negative inotropic of propofol and thiopental are, at least in part, related to decrease in Ca~(2+) trans-sarcolemmal current by accelerating L-type calcium channel inactivation. Both anesthetics decrease delayed rectifier potassium current, thus partially antagonizing the effect of decreased calcium current.

[Method]Learning Zhu Danxi related books and Kampo medicine literature, analyse the impact of Danxi ’s medical science on each genre of Japan Kampo medicine,explain the role of Danxi ’s medical science in the formation and development in Japan Kampo medicine system,and ummarize its development path in Japan.[Result]The development of Danxi ’s medical science in Japan experienced ups and downs, combined with local realities,and derived more Japanese-oriented medical theories. [Conclusion]Danxi ’s medical science not only for the formation of Japanese Kampo medicine system plays an important role, but also has a profound impact on Japan's three Kampo medical genre.Its development track is worth thinking and summarizing.

Objective:To study the cytotoxic effects on tumor cell lines Hela,BGC823,MCF-7,L342,H9101,D6 induced by Fas ligand and anti-human DR5 monoclonal antibodies(Anti-DR5 mAb) and the underlying mechanism.Methods:Fas/DR5 mRNA were detected by RT-PCR. Cytotoxicity exerted by FasL/Anti-DR5 mAb on tumor cell lines was measured by MTT colorimetry and the induced apoptosis was determined by agarose gel electrophoresis.Results:The expression of DR5 on BGC823 and Hela cells were higher and DR5 didn’t express in D6. The expression of Fas on H9101 and L342 were higher and Fas didn’t express in D6. Cell line H9101 and L342 were sensitive to Anti-DR5 mAb and FasL in a dose dependent manner; Cell line MCF-7 and BGC823 were sensitive to FasL but were partially sensitive to Anti-DR5 mAb; Cell line Hela was partially sensitive to FasL but was sensitive to Anti-DR5 mAb; Cell line D6 was insensitive to two apoptosis inductions.Conclusion:Apoptosis induced by Fas ligand and Anti-DR5 mAb vary among tumor cell lines. The underlying mechanism may be relevant to Fas/DR5 mRNA expression,the release of Caspase-8 and Bcl-2.

Purpose To investigate the effects of different concentration of ketamine on Ca2 transsarcolemmalinflux induced by KCl in isolated rat ventricular myocytes. Methods Freshly isolated rat ventricular nyoeyteswere loaded with Fluo-3AM, a Ca2 + indicator. The effects of different concentration ketamine( 1 × 10- s, 1 × 10- 4,1 × 10-3 mmol/L) on the change of intracellular Ca2+ concentration induced by KCl were investigated. ResultsLow concentration ketamine(1 × 10-5 mrnol/L) did not change Ca2+ transsarcolemmal influx. Although mediumeoncentration ketamine( 1 × 10-4 rmol/L) made the influx slower, the eventual peak concentration of intracellularCa2+ had no difference from that of the control group. The high concentration ketamine (1 × 10-3 mmol/L) inhibited Ca2-1 influx,intracellular Ca2+ fluorescent intensity decreased about 13.2% (P＜0.05). ConclusionsKetamine inhibits Ca2 + trranssarcolemmal influx in isolated rat ventricular myocytes dosedependently, which may inpart explain its negative inotropic effect.

【Objective】 To explore the complication and engra ftment of human cord blood hematopoietic stem cells in utero transplantation thr o ugh abdominal cavity of fetal rats , and to establish an animal model for clini cal application. 【Methods】　Human cord blood (MNC) cells were transplanted into th e abdominal cavity of fetal rats, the complications and the outcome of pregn ancy were observed. The condition of engraftment was detected by flow cytometr y and immunohistochemistry methods after the fetus were born. 【Results】 Huma n CD3 cells were detected in rats and the engraftment rate was 64%. At 1 and 2 months of age, the mean value of human CD3 cells were 0.28%±0.05% and 0.41 %± 0.05% respectively （P＜0.05）.Human CD3 、CD20及 CD+34 ce lls were also detected in liver、spleen and thymus of rats at 2 months of age. The i ncidence of complication was significantly different between transplanted grou p and non-transplanted group. 【Conclusion】　Human cord blood cells transfused into the abdominal cavity of fetal rats were engrafted . There were some complication s occurred during operations which affected the outcome of pregnancy.

Objective:To evaluate the clinical effects of ball attachment and Locator attachment for implant-supported overdenture.Methods:67 cases with edentulous jaw were treated with 150 Straumann and Bego implants and implant-supported overdentures using ball attachments Locator attachments,respectively.All cases were followed up regularly,biological and mechanical complications were observed,the patient satisfaction after restoration was compared between groups.Statistical analysis was performed using SPSS 17.0 software.Results:Follow-up was completed in 58 cases,25 with ball attachment,26 with Locator attachment and 7 with ball changed by Locator.Biological complications comparison showed that the incidence of gingival hyperplasia in the ball group was higher than that in the Locator group(P ＜ 0.05).There was no statistical difference of mechanical complications between the 2 groups (P ＞ 0.05),but the incidence of all kinds of mechanical complications in the Locator group was less than those in the ball group,the average repair frequency of the ball and Locator group was 1.9 times and 0.9 times respectively.Patients'satisfaction on chewing and retention was higher in the Locator attachment group than that in the ball attachment group(P ＜ 0.05).No statistical difference was found in patients' satisfaction between genders (P＞0.05),and no correlation of satisfaction was found with patients' age and follow-up time.Conclusion:The clinical effects of implant-supported overdenture using the Locator attachment is superior to that using the ball attachment.

OBJECTIVE: The purpose of this study is to assess the effects of aging on air-conducted sound elicited cervical vestibular evoked myogenic potential (cVEMP) and ocular vestibular evoked myogenic potential(oVEMP) in normal people. METHOD: Sixty normal subjects were recruited for the vestibular evoked myogenic potential examination. Among them, 20-40 year-old group was setted as the first group, 41-60 year-old group was setted as the second group, > 60-year-old group was setted as the third group. Each group included 20 normal subjects. SPSS 16.0 software was used to analyze the data. RESULT: As the age growing, P1 latency prolonged, N1-P1 amplitude decreased, and amplitude asymmetric ratio increased in cVEMP; N1 latency prolonged, P1-N1 amplitude decreased, and amplitude asymmetric ratio increased in oVEMP. CONCLUSION With the age growing, the time of otolithic organ input pathways prolonged, the function of otolithic organ decreased and the decrease level may be asymeetrical on both sides.

AIM To elucidate the direct effects of propofol and thiopental on cardiac contraction by measuring intracellular Ca 2+ concentration change in cultured rat ventricular myocytes. METHOD Ventricular myocytes of neonatal rats were obtained by enzymatic digestion with typsin and were cultured for 4～5 days, then were loaded with Fluo 3AM, a new Ca 2+ indicator. The effects of propofol (50, 250 ?mol?L -1 ) or thiopental (100, 500 ?mol?L -1 ) on changes of intracellular Ca 2+ fluorescent intensity induced by KCl, isoprenaline or caffeine were investigated. RESULTS Both intravenous anesthetics caused dose dependent inhibit Ca 2+ transsarcolemmal influx induced by KCl and isoprenaline, peak Ca 2+ fluorescent intensity decreased. The inhibitory effect of thiopental was more than propofol at equiopotency. Neither anesthetics altered the amount of Ca 2+ release from intracellular stores in response to caffeine. CONCLUSION The negative inotropic effects of propofol and thiopental, at least in part, may be mediated by a decrease Ca 2+ transsarcolemmal influx though voltage gated and receptor gated calcium channel, but it is not relate to sarcoplasmic reticulum Ca 2+ release.

OBJECTIVETo observe the impacts of acupoint catgut embedding therapy and acupuncture-moxibustion therapy on the long-term efficacy and patient's life quality in the treatment of allergic rhinitis.

METHODSSixty-nine patients were randomized into the combined acupuncture-moxibustion and acupoint catgut embedding therapy group (combined therapy group, 36 cases) and an acupuncture-moxibustion group (33 cases). In the acupuncture-moxibustion group, acupuncture was applied at Yingxiang (LI 20), Shangyingxiang (EX-HN 8), Yintang (GV 29), Shangxing (GV 23), Tongtian (BL 7) and Zusanli (ST 36). Moxibustion was applied at Zusanli (ST 36), Feishu (BL 13), Dazhui (GV 14) and Fengmen (BL 12). In the combined therapy group, on the basis of the treatment as acupuncture-moxibustion group, the catgut embedding therapy was applied at Feishu (BL 13), Fengmen (BL 12), Pishu (BL 20), Shenshu (BL 23), Zhongwan (CV 12) and Qihai (CV 6). The treatment duration was 4 weeks in the two groups. The clinical efficacy of allergic rhinitis and rhinoconjunctivitis quality of life questionnaire (RQLQ) score were observed before and after treatment as well as in the 4-weeks follow-up after the end of treatment respectively.

RESULTSThe markedly effective rate was 72.7% (24/33) in the combined therapy group and 48.4% (15/31) in the acupuncture-moxibustion group after treatment. The efficacy was similar between the two groups (P > 0.05). It was 57.6% (19/33) in the combined therapy group and was 22. 6% (7/31) in the 4-week follow-up after treatment, indicating the long-term efficacy in the combined therapy group was superior to that in the acupuncture-moxibustion group (P<0. 05). Scores of RQLO after treatment and in 4-week follow-up after treatment in both groups were improved as compared with those before treatment (all P < 0.05). In 4-week follow-up, the improvements in sleep and affection in the combined therapy group were superior to the acupuncture-moxibustion group (3.27 +/- 3.23 vs 4.61 +/- 3.56, 3.48 +/- 3.67 vs 5.81 +/- 4.15, both P < 0.05).

CONCLUSIONThe acupoint catgut embedding therapy combined with acupuncture-moxibustion therapy are safe and effective in the treatment of allergic rhinitis and display the more roles in the long-term efficacy.
