2.Effect of Physalin B on Chemiluminescence and H_2O_2 Production of Activated Polymorphonuclear Leukocyte

Effect of opsonized zpoosan on polymorphonuclear leukocyte (PMN)were studied by chemilundnescence (CL)and hydrogen peroxide (H2O2)assays. Results showed that different concentrations of Physalin B(PhB) presented a dose-dependent inhibitive effect on CL and H2O2 production of activated PMN. It suggested that the anti-inflammatory action of PhB may be related to the inhibition of production and release of oxygen free radical by PMN.
3.Clinical significance of serum prealbumin and bilirubin in patients with coronary heart disease

Objective To investigate the changes and clinical significance of serum prealbumin (PAB) and bilirubin in patients with coronary heart disease (CHD). Methods The levels of serum PAB and bilirubin were measured respectively in 234 patients with CHD (CHD group) and 77 patients with paroxysmal supraventricular tachycardia (PSVT, control group), and the former group was divided into stable angina pectoris (SAP) group with 80 patients,unstable angina pectoris (UAP) group with 84 patients and acute myocardial infarction (AMI) group with 70 patients,according to the chnical manifestation. Results The levels of total bilirubin (TBIL),direct bilirubin (DBIL) and indirect bilirubin (IBIL) in CHD group [ (11.8 ± 6.1 ), (4.8 ± 1.0) and (7.7 ± 2.7) μ mol/L] were significantly lower than those in control group [(14.6 ±5.6), (6.2 ±1.3) and (9.5 ±6.1)μmol/L] (P <0.05). The levels of TBIL, DBIL and IBIL decreased gradually from SAP group, UAP group to AMI group, but there was no significant difference among three groups (P > 0.05 ). The levels of serum PAB in CHD group were also significantly lower than those in control group [(205.55 ±5.68) mg/L vs.(229.78 ± 9.62) mg/L] (P < 0.05),and decreased gradually from SAP group,UAP group to AMI group [(215.73 ± 7.98), (214.12 ± 3.79) and (195.75 ± 7.07) mg/L],but there was no significant difference among three groups (P > 0.05 ). Conclusion The levels of serum PAB and bilirubin are related to the severity of CHD, and can be used as important biochemical indicators in monitoring the attack and severity in patients with CHD.
4.Effects of topiramate on quantitative pharmacoelectroencephalography

Objective To investigate the influence of topiramate(TPM) on quantitative pharmacoelectroencephalography (QPEEG) in normal subjects and epilepsy patients. Methods EEG were recorded in normal and epileptic subjects prior to and at 0.5,1,2,4,6,8,12,24 hours after the administration of single dose of TPM.The EEG power spectral activity including the absolute power,percent of power,total power in both occipital and total power of the whole scalp areas were calculated through 30 s epochs without artifacts after each recording.The statistical difference between baseline assessment and each post-drug time point was analyzed by the Wilcoxon matched-paired rank test. Results The power of sloe wave(peak value 66.76 ?V 2),?1-band (peak value 57.33 ?V 2)and total power (peak value 385.12 ?V 2)was increased after the administration of TPM and the total power of the whole scalp areas was also increased(peak value 2 500.75?V 2).The percent of power of ?-band(peak value 12.4%) and ?1-band(healthy control,peak value 17.5%) or ?-band(patient,peak value 13.94%)was increased,while ?3-band(healthy control,lowest value 10.4%) decreased. Conclusion TPM might change the background activity of EEG that was different from other antiepilepsy drugs.
5.Relationship between Expression of podocin mRNA and Oxidative Stress in Rats with Adriamycin-Induced Nephrotic Syndrome

Objective To explore the possible association of oxidative stress reaction with the mRNA expression of podocin in rats with adriarnycin(ADR)-induced nephrotic syndrome.Methods mRNA expression of podocin in renal cortex were investigated by in situ hybridization staining and semi-quantitative RT-PCR in ADR-induced nephrotic rats,the level of malondialdehyde(MDA), superoxide dismutase(SOD)and total antioxidative capacity(T-AOC) in renal cortex were measured, then the relationship between them was evaluated.Results In ADR rats, MDA increased at d7 and reached a significant higher level at d14 and d21;SOD decreased at d14 and persisted to d21; T-AOC decreased at d21.The podocin mRNA mostly expressed in cytoplasm of glomerular cells. Both the number and the intensity of positive cells increased notably as time progressed in ADR rats.The level of podocin mRNA expression showed no obvious changes at d7, while prominently increased at d14 and pesisted to d21 in ADR rats.There was significant correlativity between the mRNA expression of podocin and the level of MDA,SOD,T-AOC.Conclusion The mRNA expression of podocin is correlated with oxidative stress reaction in rats with ADR-induced nephrotic syndrome.
6.Effect of chaperone-mediated autophagy in MPP(+) -induced SH-SY5Y cells and interventional effect of puerarin.

OBJECTIVETo study the protective effect of puerarin on MPP(+) -induced SH-SY5Y cells by chaperone-mediated autophagy (CMA).
METHODThe Parkinson's disease cell model was established by injuring SH-SY5Y cells with 1 mmol x L(-1) MPP+. The CCK-8 staining was adopted to detect the effect the puerarin of different concentrations on the survival rate of MPP(+)-induced SH-SYSY cells. The autophagosome formation was observed under transmission electron microscope. The AO staining showed the changes in the lysosome activity. RT-PCR was used to detect the changes in Lamp2a and Hsc70 mRNA expressions. The western blotting was adopted to test the expressions of Lamp2a, Hsc70 and alpha-synuclein protein in cells.
RESULTWithin the concentration range of 12. 5-50.0 micromol x L(-1), the pretreatment with puerain for 30 minutes could protect the injury of MPP+ in SH-SY5Y cells, and showed a certain dose-effect relationship. The AO staining and electron microscope showed the effect of puerain within the concentration range of 12.5-50.0 micromol x L(-1) on 1 mmol x L(-1) MPP(+)-induced SH-SY5Y cells; autophagosomes emerged in cells, and increased along with the rise in the puerarin dose. The results of the flow cytometry revealed that 50.0 micromol x L(-1) of puerarin could protect against the increase of the ROS level in 1 mmol x L(-1) MPP(+) -induced SH-SY5Y cells and prevent the oxidative injury. The results of RT-PCR and western blotting indicated that puerain within the concentration range of 12.5-50.0 micromol x L(-1) alleviated the MPP(+)-induced SH-SY5Y cell injury, and inhibited the accumulation of alpha-synuclein proteins in MPP(+) -induced SH-SY5Y cells by up-regulating Hsc70, Lamp2a mRNA and protein level.
CONCLUSIONPuerarin could protect against the MPP(+) -induced cell injury, whose protective mechanism may be related to the chaperone-mediated autophagy pathway of interventional molecules.

7.Clinical significance of serum parameters detection in the diagnosis of hypertension disorders complicating pregnancy

Objective To explore the diagnostic value of combined detection of serum calcium ion (Ca2+ ) ,C‐reaction protein (CRP) ,uric acid(UA) ,homocysteine(Hcy) and complement C3 ,C4 in hypertension disorders complicating pregnancy(HDCP) . Methods Patients with HDCP were enrolled as HDCP group ,and pregnant women without HDCP were enrolled as control group . Serum parameters ,mentioned above ,were detected and compared between the two groups .Diagnostic performance of the parameters was also analyzed .Results Serum levels of Ca2+ ,CRP ,UA ,Hcy and complement C3 ,C4 in HDCP group were significantly differ‐ent from those in control group(P<0 .05) .Sensitivity ,coincidence rate and correct diagnosis rate of combined detection of these six parameters were higher than single detection(P<0 .05) ,but the specificity was without significant difference(P>0 .05) .Serum lev‐els of CRP ,UA and Hcy increased with the progress of HDCP disease condition ,but serum levels of Ca2+ ,C3 and C4 decreased .Be‐tween different sub‐group of HDCP patients ,serum levels of the six parameters were with significant differences(P<0 .05) .Conclu‐sion Early combined detection of serum Ca2+ ,CRP ,UA ,Hcy and complement C3 ,C4 could be with significance for the early diag‐nosis and treatment of HDCP ,and also for the condition assessment of HDCP .
8.Quantitative electroencephalography effects of topiramate: a nonlinear analysis

ObjectiveTo investigate the effects of topiramate (TPM) on complexity of electroencephalography (EEG) by means of quantitative pharmacoelectroencephalography (QPEEG) and nonlinear analysis methods.MethodsOne dose of TPM was administrated to epileptics and healthy adults. The EEG samples were obtained prior to and at regular intervals (at 0.5, 1, 2, 4, 6, 8, 12, 24 hours after the administration) within 24 hours following the administration of TPM. The EEG activity was processed with the nonlinear analysis methods. The complexity measure, approximate entropy, Lyapunov exponent and fractional dimension of the whole scalp areas were calculated through 60 seconds epochs without artifacts after each recording. The statistical difference between baseline predrug assessment and each postdrug control was described by analysis of variance.ResultsThe Lyapunov exponent was increased first, then decreased, and then increased finally (the maximum value was 0.44 in both centrals in the healthy people). The approximate entropy and fractional dimension showed the tendency of descent, the complexity measure decreased in healthy adults (the least value was 0.34 on right parietal), but increased in patients (the maximum value was 0.34 in right posterior temporal).ConclusionTPM might change the complexity of EEG.
9.Possibility of no recurrent laryngeal nerve injury in thyroid surgery

OBJECTIVE To explore the possibility of no recurrent laryngeal nerve injury in thyroid surgery. METHODS A total of 659 consecutive patients with thyroid disease undergoing thyroidectomy byotolaryngologists from March 2001 to March 2005 were retrospectively analyzed. The operative mode and incidence of complications, particularly postoperative RLN palsy, hypoparathyroidism and postoperative recurrence were evaluated. Routine dissection and identification of the RLN was performed during all operative procedures and parathyroid with the blood supply was preserved. RESULTS Unilateral total thyroidectomy with contralateral partial lobectomy was performed in 376 cases, unilateral total thyroidectomy with isthmectomy in 87, bilateral subtotal thyroidectomy with the remnant left at the upper pole in 76, total thyroidectomy in 73. The operations on 47 patients with substernal goiter have been successfully performed via cervical collar incision. None of our patients incurred unilateral or bilateral vocal cord paralysis and permanent hypocalcaemia. Of these patients, the incidence of temporary postoperative hypocalcemia was 1.67 %(11/659). Postoperative hemorrhage requiring reoperation occurred in 4 cases (0.60 %) and 5 patients developed wound haematomas (0.76 %).Postoperative hypothyroidism was found in 3 patients(0.45 %).The incidence of postoperative recurrence was 0.15 %(1/659). No patients had incision infection. CONCLUSION With knowledge of the anatomy of the RLN and routinely complete identification the RLN in performing capsular dissection high on the surface of the thyroid gland, RLN injury may be avoided in thyroid surgery. 10.Semantic Processing in Patients with Epilepsy Studied by fMRI

Objective To reveal the cerebral activation areas related to semantic processing of language functions in patients with temporal lobe epilepsy(TLE).Methods 6 patients with left-TLE,6 patients with right-TLE,and 6 controls,all right-handed,were studied using functional magnetic resonance imaging(fMRI)during a reversible words decision task.Their responses were recorded by key-press.The image data was analyzed by statistical parametric mapping 2(SPM2)including the individual statistical analysis and group analysis.Results There was no difference between the right-TLE patients and controls in cerebral activation,which included left inferior frontal gyrus,left middle frontal gyrus,left superior temporal gyrus,left middle temporal gyrus and bilateral cingulate gyrus,bilateral fusiform gyrus,bilateral basal ganglia,bilateral cerebellum(especially right).The activation in patients with left-TLE was atypically distributed relative to the controls,there were more activation in right inferior frontal gyrus,right precentral gyrus and left cuneus,left precuneus.Conclusion There is atypical distribution of the cerebral activation area related to semantic processing of language functions in patients with left-TLE,and enantiomorphous transformation is found in the contrelateral side of cerebrum.
