1.Research progress on cancer therapy targeted at myeloid derived suppressor cells

Myeloid-derived suppressor cells (MDSC) are a heterogeneous population of cells of myeloid origin that comprises myeloid progenitor cells and immature macrophages, which expand in tumorbearing mice and patients' bone marrow, spleen and peripheral blood and recruit to the tumor site. MDSC express high levels of arginase 1 (ARG1), inducible-nitric oxide syntheses (iNOS), reactive oxygen species (ROS) and peroxynitrite. They could suppress T-cell functions by cell contact or not, and induce regulatory T cells (Treg), all of the above are its weapons to defend individuals' immune system. Anti-tumor strategies targeted at MDSC develop rapidly now. In this review, we briefly introduce the strategies that targeted at MDSC and their mechanisms.

3.Imaging Evaluation of Gastrointestinal Stromal Tumor

Objective To analyze the imaging features of gastrointestinal stromal tumor(GIST).Methods Imaging features and differential diagnosis of GIST proved surgically and pathologically in 21 cases were studied retrospectively.Results The lesions occurred in esophagus in 1,stomach in 15,small intestine in 3 and colon in 2.The findings of barium meal examination included flattened or shifed mucosal folds(6/21),filling defect(8/21) and extralumially oppressed sign(2/21).CT showed extralumial or intraluminal mass with homogeneous density in 12 cases,liquid-dense necrosis in 6 cases and calcification within tumor in 2 cases.Remarkable enhancement was noted in all tumors,heterogeneous and homogeneous enhancement were showed in 17 and 3 cases respectively.Conclusion GIST is one of the most common gastrointestinal mesenchymal tumors.CT is an effective and useful modality in the diagnosis of GIST.
4.Legal Regulation on the Financial Conflicts of Interests of Researchers in Drug Clinical Trials

OBJECTIVE:To put forward suggestions for the legal regulation on the financial conflict of interest of researchers in drug clinical trials.METHODS:The definition and harm of the financial conflict of interest of the researchers in clinical trials were analyzed,and the legal measures of FDA on regulating and controlling the financial conflict of interest of the researchers were evaluated.RESULTS & CONCLUSIONS:The Chinese legislature should draw useful legal experiences from USA to establish suitable legal measures to regulate and control the financial conflict of interest of researchers in clinical trials,clarify the definition of the financial interest of researchers,require the applicant for clinical trial to disclose his financial interest and the applicants for drug registration to submit the statement about the financial conflict of interest of researchers to the drug administration department.
5.Effects of Fu Shen ke li on myocardial cell apoptosis and endoplasmic reticulum stress-related protein expressions in rats with adriamycin induced heart failure

Objective To explore the contribution of endoplasmic reticulum stress to cardiac myocyte apoptosis in mouse congestive heart failure induced by Doxorubicin.Methods Doxorubicin (DOX)was administered intraperitoneally to rats,2 mg/kg doses over 8 weeks.Echocardiographic and hemodynamics measurements were obtained at 8 weeks after treatment.Then left ventricular (LV) samples were collected.The successfully established rats were divided into 3 groups (Fu Shen Ke Li 40 mg/kg,60 mg/kg,and 80 mg/kg).After 5 weeks post the operation,an echocardiographic was performed on all the rats under narcotism,Western blot to test the B-cell lymphoma-2 (Bcl-2),myeloid cell leukemia-1 (Mcl-1),Bcl-2-Associated X (Bax),Caspase-3,C/EBP homologous protein (CHOP),and 78-kDa glucose-regulated protein(GRP78),and reverse transcription-polymerase chain reaction (RT-PCR) to test the mRNA expression levels of Mcl-1,and NOXA.Results The left ventricular end-diastolic diameter (LVEDD),left ventricular end systolic diameter (LVESD),left ventricular ejection fraction (LVEF) prompted modeling success The left ventricular end-diastolic diameter (LVEDD),left ventricular ejection fraction (LVEF),and fractional shortening (FS) have been improved in high and middle groups of Fu Shen Ke Li.Compared to the model group,higher expression levels of Bcl-2,and Mcl-1 were,lower expressions of Bax,NOXA,CHOP,and GRP78 were (P < O.05) in 3 groups of Fu Shen Ke Li.Compared to the model group,there were higher mRNA expression level of Mcl-1 was,lower mRNA expression of NOXA was (P < 0.05) in 3 groups of Fu Shen Ke Li.Conclusions Fu Shen Ke Li depresses endoplasmic reticulum stress,which can reduce the apoptosis of myocardial cells,and ultimately achieve the purpose of treatment of heart failure.

Drug transporters are functional membrane proteins located in various tissues, which play vital roles in absorption, distribution and excretion of drugs, especially those located in intestine, liver and kidney. The expression and function of transporters will alter in diseases state, which affects the therapeutic effects of drugs by altering their pharmacokinetics. In this review, we focus on the alterations in related transporters and the effect on the drug therapy in common intestinal diseases, liver diseases, kidney diseases and diabetes mellitus.

Objective To observe the effects of sodium arsenite (NaAsO2) on the expression of transcription factor nrf2 (Nrf2) and heme oxygenase 1 (HO-1) in Chang liver cells, and to explore the possible mechanism by using buthionine sulfoximine ( BSO), a GSH synthesis inhibitor. Methods Chang liver cells were treated with NaAsO2 at the doses of 5, 10 and 20 ?mol/L, alone, for 24 h, or pretreated with BSO (3 mmol/L, 12 h).
