1.Effect of tumor necrosis factor-α and asymmetric dimethylarginine on left ventricle of patients with primary hypertension

Objective To understand the level of tulnor necrosis factor-α(TNF-α)and asymmetric dimethylarginine (ADMA) in serum of healthy adults and primary hypertension patients with different left ventricle,and to explore the role of TNF-α and ADMA in the pathophysiological mechanism of heart damage due to hypertension.Methods Thirty health adults were recruited as control.Based on the left ventricular mass index(LVMI)and relative wall thickness(RWT),66 patients of hypertension were divided into 4 groups:normal left ventricle group with normal LVMI and RWT,concentric remolding group with normal LVMI and incremental RWT,typical concentric hypertrophy group with incremental LVMI and RWT,eccentric left ventricular group incremental LVMI and normal RWT.TNF-α was measured by radioimmunoassay and ADMA was measured through reversed phase high-performance liquid chromatography.Resuits TNF-α and ADMA increased with the severity of the left ventricle.Each left ventricle group was significantly higher than control group (P<0.01).The differences between left ventricle groups were statistically significant (P<0.01).Conclusions TNF-α and ADMA may participate in the development of left ventricle.

4.Preparation of HER2 radioligand 99Tcm-ABH2 and its in vivo imaging in breast cancer-xenografted mice

Objective To prepare 99Tcm-human epidermal growth factor receptor 2 (HER2) affibody (ABH2) and explore its feasibility as an imaging agent in HER2-positive breast cancer.Methods Sodium glucoheptonate and SnCl2 · 2H2O were used to label ABH2 with 99Tcm.The labeling yield and radiochemical purity of 99Tcm-ABH2 were determined.The stability of 99Tcm-ABH2 was tested in PBS and serum.The equilibrium disassociation constant (Kd) of 99Tcm-ABH2 was measured with MBA-MD-361 breast cancer cells.SPECT/CT imaging was carried out at 1.0 h and 4.5 h after injection of 37 MBq 99Tcm-ABH2 in nude mice (n=4) xenografted with MBA-MD-361 breast cancer.The T/NT (liver,brain,lung,heart,bone and muscle) ratios were deduced from SPECT/CT acquired data.For the blocking experiment,200 μg ABH2 was injected intravenously before 99Tcm-ABH2 injection.SPECT/CT imaging was performed in the same way.The T/NT ratios were compared between non-blocked and blocked groups by one-way analysis of variance.Results Fhe labeling yield of 99Tcm-ABH2 was above 99%.99Tcm-ABH2 was substantially stable in PBS and serum;the radiochemical purity was (95.0± 1.0)% after incubation with serum at 37 ℃ for 6.0 h.The Kd of 99Tcm-ABH2 was 1.7 nmol/L.The radioactive uptake in cancer was visualized at 1.0 h and 4.5 h after injection of 99Tcm-ABH2 in HER2 positive MDA-MB-361 breast cancer.99Tcm-ABH2 was cleared out mainly through urinary system.The ratios of tumor to liver,lung,brain,heart,muscle and bone were 1.81± 0.60,8.95±1.13,20.08±6.12,7.61±0.56,10.62±1.78,11.42±2.07,respectively at 4.5 h after 99Tcm-ABH2 injection.After ABH2 blocking,the corresponding T/NT ratios were 0.60±0.23,3.05± 1.38,5.24±2.17,2.42±1.02,8.16±2.66,2.76±0.48 (F=29.38,P<0.05) respectively.Conclusion 99Tcm-ABH2 could be synthesized with high purity,and this new agent could be used to image HER2-positive breast cancer specifically.
5.MicroPET imaging with 2-[18F] fluoropropionic acid in Lewis lung carcinoma-bearing mice

Objective To prepare 2-[18F]fiuoropropionic acid (18F-FPA) and evaluate its biodistribution and imaging capacity in Lewis lung carcinoma-bearing mice.Methods 18F-FPA was prepared by nucleophilic substitution reaction,and its hydrophilicity was analyzed.18F-FPA (7.4-11.1 MBq) was injected into Lewis lung carcinoma-bearing mice via tail vein.MicroPET imaging was performed at 20,80 min after the injection.The biodistribution of 18F-FPA in organs was analyzed.The blocking effects of sodium propionate and dichloroacetate to 18F-FPA were tested in vivo.Data were analyzed by two-sample t test using GraphPad Prism software.Results The synthesis of 18F-FPA took 40 min.18F-FPA had high radiochemical purity (>99%) and hydrophilicity.18F-FPA was mainly distributed in the carcinoma,the urinary bladder and the caecum.The radioactive uptakes in muscles,brown fat and bones were relatively low.Quantitative analysis showed that the uptake of 18F-FPA in Lewis lung carcinoma from 20 min to 80 min was slightly increased ((17.03±2.87) %ID/g vs (19.33±2.45) %ID/g) without significant difference (t=1.100,P>0.05).Neither sodium propionate nor dichloroacetate could block the uptake of 18F-FPA in Lewis lung carcinoma (t=1.544,0.894;both P>0.05).Conclusions 18F-FPA can be quickly synthesized and has good physicochemical properties.It can be used as a tracer to visualize Lewis lung carcinoma in mice,and its tumor uptaking can not be blocked by propionate and dichloacetate.18 F-FPA PET has the potential to detect lung cancer noninvasively in clinic.
6.Bibiometric analysis on papers produced by Peking university hospital

Objective To evaluate the academic status of the researchers in pediatric department of Peking university first hospital, we conducted bibliometric analysis of research on the scientific publications. Methods The total number, first authors, and the citing and cited conditions of these papers published from 1999 to 2000 were analyzed by bibliometric methods. Results There were total 513 papers including 275 original articles published in 59 journals from 1999 to 2003.There were 238 papers written in Chinese, of which 188 papers (79 %) were from the core journals. In the 37 original articles written in English, 31 papers(83.8 %) were select-ed by science citation index(SCI).The average of impact factors (IF) of sourcing journals was 1.447(0.107 -7.717). The averages of IF per year ranged from 0. 768 to 2.206.The original articles selected by SCI were totally cited 38 times. Every article was cited 1.41 times in average.Of all 275 papers,each had an average of 9.8 citing reference, increasing from 8.1 in 1999 to 12.0 in 2003.About 80.7 % of those references were written in foreign languages.The Price's index of those papers was 43.2%.Conclusions The number of papers kept improving in the past 5 years, and had a significant improvement in 2003.The authors showes great talents to use English articles as their main information sources. But the utilization of the last published papers in this subject should be im-proved.
7.Clinical analysis of non-Hodgkin lymphoma of palatine tonsil-Report of 70 cases

Objective To investigate the treatment method and prognostic factors of patients with non-Hodgkin lymphoma(NHL)of palatine tonsil.Methods 70 patients with NHL of palatine tonsil were re- viewed.According to Ann Arbor staging classification,12 patients had stageⅠ,39 stageⅡ,15 stageⅢand 4 stageⅣ.Working formulation was used in pathologic classification which was low grade 8 cases,intermedi- ate grade 28 cases and high grade 34 cases.30 cases were treated by chemotherapy alone,3 cases by radio- therapy alone,37 cases by chemotherapy and radiotherapy.Results The overall survival rate was 56.3%. Objective To explore the correlation between the level of serum uric acid (UA) and high-sensitivity C-reactive protein(hs-CRP) with sudden cardiac death of coronary artery disease.Methods Clinical data of 80 patients with coronary artery disease were collected,and according to whether occurred coronary heart sudden cardiac death,the patients were divided into the observation group(coronary heart disease with sudden cardiac death group) and the control group(coronary heart disease with non-sudden death group),each group had 40 cases.The levels of serum UA,low-density hpoprotein cholesterol (LDL-C),blood sugar,hs-CRP of the two groups were detected and compared.Results The UA and hs-CRP levels of male and female in the observation group were (444.06 ±36.44.) μmol/L,(7.29 ± 0.57) mg/L,(399.25 ± 9.91) μmol/L,(6.56 ± 0.38) mg/L,respectively,which were significantly higher than those in the control group [(289.40 ± 25.78) μmol/L,(1.04 ± 0.11) mg/L,(209.90 ±29.87) μmol/L,(1.22 ± 0.31) mg/L] (t =-15.49,-47.71,-26.90,-47.92,all P < 0.01).There were no statistically significant differences in the levels of LDL-C and blood glucose between the same sex of the two groups(t =-1.34,-1.54,-1.06,-1.33,all P > 0.05).Conclusion The increased levels of serum UA and hs-CRP may be associate with sudden cardiac death of coronary artery disease.
9.Mucoraceae infection lead to the pseudoaneurysms following renal transplantation: a report of 2 recipients with the same donor after cardiac death

Objective To analyze the diagnosis and treatment of iliac pseudoaneurysms following renal transplantation.Method The data of two patients with pseudoaneurysm who underwent kidney transplantation were retrospective analysied.Result One case with pseudoaneurysm received transplant nephrectomy,and pathological examination diagnosed mucoraceae infection; the other patient received endovascular treatment and amphotericin B therapy,endly died of hemorrhagic shock.Conclusion The development of pseudoaneurysms at the kidney transplantation recipients with the same donor results in high rates of mucormycosis.They should take anti-fungus therapy and operation as early as possible.
10.Detection of causative allergens of cosmetic allergic contact dermatitis

Objective To make a survey on common cosmetic allergens, and to provide epidemiological data and clinical evidence for cosmetic allergy. Methods Patch test was performed by using 49cosmetic allergens from a European cosmetic series and 5 Chinese standard screening allergens on 89patients with suspected cosmetic allergic contact dermatitis. Test results were determined according to the International Contact Dermatitis Research Group (ICDRG) recommendation. Results Of the 89 patients, 61(68.5%) showed positive reactions to one or more cosmetic allergens. The most common allergens were fragrances (33.7%), followed by preservatives (30.3%), para-phenylenediamine (25.8%) and amerchol L 101(10.1%). Conclusion Fragrances, preservatives, para-phenylenediamine and amerchol L 101 are dominant causative allergens in patients with cosmetic allergic contact dermatitis.
