1.Clinical Study on Large Dose Intravenous Immunoglobulin in Decreasing Incidence of Asthma Following Bronchiolitis

Objective To study the efficiency of large dose intravenous immunoglobulin(IVIG) in decreasing incidence of asthma following bronchiolitis.Methods One hundred and sixty-eight children with bronchiolitis were chosen to use different treatment plans,81 children receiving large dose of IVIG[ 0.4 g/(kg?d)] daily,on the basis of routine treatment for 5 days,while other 87 children were reserved as control group receiving routine treatment only,which were followed up 5 to 10 years.Results After following up 5 to 10 years,only 17 cases in 81 patients with bronchiolitis receiving large dose IVIG developed into asthma.However,46 cases in control group developed into asthma.The incidence of asthma was 20.99% and(52.87%) in two groups,respectively,showing significant(differe-)nce between them(P

3.Research on the methods for inter-class distinctive feature selection for leucocyte recognition based on attribute hierarchical relationship.

To increase efficiency of automated leucocyte pattern recognition using lower feature dimensions, a novel inter-class distinctive feature selection method for chromatic leucocyte images was proposed based on attribute hierarchical relationship. According to the attribute constraints in formal concept analysis, we established a knowledge representation and discovery method based on the hierarchical optimal diagram by defining attribute value and visual representation of optimized hierarchical relationship. It was applied to human peripheral blood leucocytes classification and 12 distinctive attributes were simplified from 60 inter-class attributes, which contributes significantly to reduced feature dimensions and efficient inter-class feature classification. Compared with the classical experimental data, the inter-class distinctive feature selection method based on hierarchical optimal diagram was proved to be usable and effective for six leucocyte pattern recognition.

6.Proliferation capacity of rabbit adipose tissues-derived mesenchymal stem cells at early stage in vitro

BACKGROUND:Surface marker molecules and differential ability of human adipose tissues-derived mesenchymal stem cells can change with the prolonged culture time.OBJECTIVE:To analyze the morphological characteristics and colony forming capacity of early cultured rabbit adipose tissues-derived mesenchymal stem cells in vitro.DESIGN,TIME AND SETTING:The cytological in vitro study was performed at the Institute of Ophthalmology,and Central Laboratory,Eye and ENT Hospital,Fudan University from January to March 2008.MATERIALS:Nine female New Zealand rabbits aged three months were obtained from the Shanghai Yingenyang Rabbit Room.METHODS:Adipose tissue was harvested from the nape fat pad of the New Zealand rabbits following anesthesia.Primary cultured cells were established using type I collagenase and cell cultures were maintained with DMEM containing 0.1 volume fraction of fetal bovine serum.Cells were passaged when 80% was confluent.The second,third and fourth passages of cells were utilized for the study.MAIN OUTCOME MEASURES:The morphology of adipose tissues-derived mesenchymal stem cells was observed using inverted microscope.Cell phenotype was detected utilizing flow cytometry.The cloning capacity was determined.RESULTS:Adipose tissues-derived mesenchymal stem cells were fibroblast-like and proliferated rapidly in vitro at the second,third and fourth passage.Flow cytometry showed that adipose tissues-derived mesenchymal stem cells expressed CD29 and PCNA.The colony-forming rates were respectively(8.0?0.6)%,(6.7?0.4)% and(4.6?0.5)%(F=12.18,P

8.Effects of Zhennao-Ning capsule on cerebral microcirculation

Objective To observe the effects of Zhennao-Ning capsule on cerebral microcirculation.Methods Cerebral hypoperfusion model of mices was induced by right common carotid arteries ligation method,The cerebral blood flow for 5min was recorded under normal circumstances comand for 30min when modeled,and then calculate the rate of blood flow changes.Microcirculatory impairment model of rats was induced by intravenous injection of high molecular dextran(HMD).The time of the emergence of microcirculatory impairment was recorded.Vascular flow,blood flow pattern and the number of outlets in the same range were recorded.Results The blood flow was significant decrease in the cerebral hypoperfusion model mice,when compared with the control mice.Zhennao-Ning capsules at the dosage of 1500 mg/kg、750 mg/kg treatment could improve the cerebral blood flow of rats at 10,20,and 30 min after modeled (56%,51%,50% and 42%,49%,51%,respectively,P<0.01).Zhennao-Ning capsules at the dosage of 600 mg/kg,300 mg/kg could significantly reduce the change of blood flow pattern(P<0.01),improve vascular flow rate (26%,36%,P<0.05 or P<0.01),extend the time of the emergence in the microcirculatory impairment model rat (53%,34%,P<0.01).Conclusion Zhcnnao-Ning capsules could improve microcirculatory impairment and partly cure the clinical migraine disease.

OBJECTIVE:To promote rational clinical use of drugs by the aid of computer.METHODS:The databases for basic data of drugs,irrational use of commonly-used drugs,compatibility of agents for injection and consultation for drug use in periods of prequancy and lactation were established.Modules for inquiry,addition,revision,deletion and maintaince were designed.The computer real time monitoring system for rational clinical use of drugs was thus developed.RESULTS&CONCLUSION:This system is simple,flexible and quick,and can check the prescription and provide consultation for compatibility between drugs.It is suitable for extensive use in clinical practice.
10.Effect of Picroside II on ERK1/2 Signal Pathway in Cerebral lschemic Injury Rats.

OBJECTIVETo explore the neuroprotective effect and mechanism of picroside II on extracellular regulated protein kinases1/2 (ERK1/2) signal transduction pathway in cerebral ischemia injuryrats. METHODS The middle cerebral artery occlusion (MCAO) model was established by inserting a monofilament into middle cerebral artery. Totally 96 successfully modeled Wistar rats were divided into the modelgroup, the treatment (picroside II) group, the Lipopolysachcaride (LPS) group, and the U0126 group according to random digit table. Each group was further divided into 3 subgroups, i.e. 6, 12, and 24 h sub-groups. Picroside II (20 mg/kg) was peritoneally injected to rats in the treatment group 2 h after ischemia.LPS (20 mg/kg) and Picroside II (20 mg/kg) were peritoneally injected to rats in the LPS group 2 h after ischemia. U0126-EtOH (20 mg/kg)and Picroside II (20 mg/kg) were peritoneally injected to rats in the U0126group 2 h after ischemia. Equal volume of normal saline was peritoneally injected to rats in the control groupand the model group. The neurobehavioral function was evaluated by modified neurological severity score(mNSS) test. The structure of neurons was observed using hematoxylin-eosinstaining (HE) staining. Theapoptotic cells were detected using terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. The expression of phosphorylated extracellular signal-regulated protein kinase1,2 (pERK1,2) in cortex was detected using immunohistochemistry (IHC) and Western blot.
RESULTSAfter cerebral ischemia injury neurological impairment score increased, the damage of neuron in the cortical area was aggravated, apoptotic cells increased in the model group as time went by. The expression of pERK1/2 increased more significantly in the model group than in the control group (P <0.05). The damage of neuron in the cortical area was milder, while apoptotic cells decreased, the expression of pERK1f2 obviously decreased more in the treatment group and the U0126 group (P < 0.05). The early damage of neuron in the cortical area was more severe, apoptotic cells and the expression of pERK12 were comparatively higher in early stage of the LPS group, but the expression of pERK1/2 was somewhat decreased in late stage.
CONCLUSIONSActivating ERK12 pathway could mediate apoptosis and inflammatory reactions of neurons after cerebral ischemia injury. Picroside II could protect the nerve system possibly through reducing activation of ERKI2 pathway, inhibiting apoptosis of neurons and inflammation reaction.
