Tumor 2007;27(12):985-989
Objective: To prepare anti-hnRNP A2/B1 polyclonal antibody and determine its clinical significance in diagnosis and prediction of non-small cell lung cancer and analyze the relationship between the expression of hnRNP A2/B1 and pathological features. Methods: HnRNP A2/B1 expression was induced by IPTG. The rabbit-anti-human hnRNP A2/B1 polyclonal antibodies were prepared and purified. The specimens were isolated from 45 cases of non small cell lung cancer tissues and paracancerous tissues and 16 cases of lung inflammatory pseudotumor tissues and embedded in paraffin. hnRNP A2/B1 expression was detected by immunohistochemical method using rabbit anti-human hnRNP A2/B1 polyclonal antibodies. The results were compared with imported anti-hnRNPA2/B1 monoclonal antibody to observe the clinical value of self-made polyclonal antibodies. Results: The rabbit anti-human hnRNPA2/B1 polyclonal antibody is successfully generated. The positive rate of hnRNP A2/B1 expression detected by anti-hnRNP A2/B1 polyclonal antibody was significantly higher in lung cancer tissues than that in paracancerous tissues (62.2% vs 40.0%, P = 0.035) and lung inflammatory pseudotumor tissues(62.2% vs 31.3%, P = 0.033). The overexpression of hnRNP A2/B1 was positively related with age and smoking and negatively related with differentiation degree. But it was not related to tumor histological classification, gender, lymph node metastasis, TNM stage, and distant metastasis. (4) The difference in survival time was significant between the two groups. hnRNP A2/B1-positive patients had shorter survival time than hnRNP A2/ B1-negative patients (P = 0.048). Conclusions: The rabbit anti-human hnRNP A2/B1 polyclonal antibody is successfully generated. Overexpression of hnRNP A2/B1 in lung cancer tissues has clinical values to a certain degree for diagnosis and prediction of the prognosis of lung cancer.

BACKGROUND:Polylactic acid(PLA)surface is hydrophobic and there are no nataral recognition sites,so its application is limited. Many different strategies have been studied such as composition and chemical Drafting,to produce hydrophilic groups. However, these traditional methods are always involved in complex process and many organic reagents, resulting in decrease of biocompatibility.OBJECTIVE:To conduct the surfacr modification of PL.A and improve its hydrophilicity with low temperature plasma treatment.DESIGN:Controlled observation.SETTING:Department of Materials Science and Engineering in Jinan University.MATERIAIS:The experiment was processed from October 2004 to October 2005 at Guangzhou Research Center of Artificial Organs and Materials Engineering(Ministry of Education). The mainly used materials were:Poly-D,L-lactic acid(Mr 2.9×104,Shandong Medical Instrument Institute),N-vinyl-pyrrolidone(NVP,Acros Company, purified before use).METHODS:The materials were treated in the plasma reactor under different conditions(power=150 W,t=3,2,1 minutes;power=30 W,t=10,8,5,3,1 minutes)to choose the optimal condition, and then PLA were grafted with NVP by gas phase polymerization and liquid polymerization, respectively.MAIN OUTCOME MEASURES:THe surface morphology of the films was observed with scanning electron microscopy and atomic force microscopy. The hydrophilicity of native and treated material surfaces was measured using a water contact angle meter.Surface composition was detected with Fourier transform infrared spectrometer.Mass changesmeasurement was applied to characterize the grafting efficiency.RESULTS:Scanning electron microscopy and atomic force microscopy showed that the plasma medified materials possessed coarse surface with pores and notches.Water contact angles decreased obviously from 78°to 50°after grafting,Fourier transform infrared spectrometer showed a new absorption peak at 1 637.19 cm-1,which corresponded to the carbonyl stretch vibration absorption of amide group in poly-N-vinyl-pyrrolidone.After the plasma treatmerlt at low temperature.the mass change ratio of PLA film material was-0.6.CONCLUSION:The plasma treatment and polymerization can improve the hydrophilicity and cause coarsr surface of PLA material, which will be helpful for the cell attachment.

Objective To evaluate KISS1 expression, and its significance in the prognosis of GIST patients. Methods In this study, 137 GIST cases and 73 non-GIST sarcoma cases were evaluated for clinicopathological characteristics and immunohistochemistry for KISS1 antibodies. Result The expression rate of KISS1 was 40.9% (56/137) in GISTs,which was significantly correlated with tumor size, disease extent, cellularity, presence of pseudocapaule, Fletcher's risk stratification and metastatic status after resection (P<0.05). Patients with positive KISS1 expression had significantly worse disease free survival and disease specific survival (P < 0.05 ). Conclusions KISS1 expression was associated with some clinicopathological characteristics as well as malignant behaviors in patients with GISTs. KISS1 might be a predictor in prognosis for GIST patients.

Objective To investigate the effect of a step-wise diversified teaching mode on cardiac electrophysiology (EP) education. Methods 64 young doctors (male:36, female:28) who studied in EP sub-specialty were enrolled. The atrioventricular nodal reentrant tachycardia (AVNRT) and atrioventricular reentrant tachycardia (AVRT) were selected as teaching content. All the doctors were randomly divided into traditional teaching group (TT) and step-wise diversified teaching group (DT) according to cross-over design protocol. In TT group, only teaching and answering the questions by teachers were given to students, for the DT group, in different stages of teaching, multiple teach-ing strategy were combined to complete teaching. T test and Chi square test were used to compare the effect of two teaching methods. Results There was no difference of basic features and time of self-studying between the two groups. In examinations, the students in DT group got higher score than TT group[DT:(92.3±9.8) and (93.1±7.8),TT:(88.3±8.6) and (87.1±10.0),P<0.05], and had better perfor-mance of learning interest, initiative, ability of finding and resolving problems, teamwork spirit and so on. Conclusion The step-wise diversified teaching mode contributes improving the quality of instruction of cardiac EP, moreover, arousing learning interest and initiative, enhancing ability of find-ing and resolving problems, teamwork spirit and so on, which is worthy of wide-popularizing in cardiac EP training.

We wished to select a cold-adapted genotype G1P[8] ZTR-68 rotavirus (China southwest strain) in MA104 cells for possible use as a live vaccine. ZTR-68 was recovered originally from children with diarrhea. The virus was cultivated at 37 degrees C at the first passage. Then, the cultivation temperature was decreased stepwise by 3 degrees C per eight passages. In total, the virus was passaged 32 times, and cultivation was terminated at 28 degrees C. Biological characteristics of the virus were analyzed during serial passages. There was no difference between the migration patterns of genomic dsRNA segments according to polyacrylamide gel electrophoresis of original and cold-adapted viruses. Infectious and red cell-agglutination titers of cold-adapted virus were lower than those of the parent virus. Also, the virus formed small-size plaques with irregular shapes at 31 degrees C and 28 degrees C. These results suggested that a genetically stable attenuated virus can be obtained through serial cold-adapted passages. Thus, an alternative strategy is provided by cold-adaption for development of attenuated live rotavirus vaccines.
