1.Mucosal administration of altered CⅡ263-272 peptide inhibits collagen-induced arthritis

Objective:To evaluate the effect of mucosal administration of altered collagen Ⅱ(CⅡ)263-272 peptide(267Q→A,270K→A and 271G→A) on collagen induced arthritis(CIA),and to explore the mechanism of the inhibitory effect of the altered CⅡ263-272 peptide on CIA.Methods:CIA was induced in Lewis rats by immunization with bovine CⅡ.Altered CⅡ263-272 peptide was given intranasally beginning from the onset of arthritis(100 ?g/dose,daily for 5 doses and continuing every other day for other 7 doses).Wild CⅡ263-272 peptide(100 ?g/dose) or PBS was administered as controls with the same procedure.Therapeutic effects were evaluated by arthritis scores,body weight change,and joint pathologic scores.The anti-CⅡ antibody and its subtypes were measured with ELISA.The cytokines of IFN-? and IL-10 were measured with ELISA.The induction of regulatory T cells was assessed by FACS analysis of percentage of peripheral CD4+CD25+ T cells,and by real-time PCR analysis of the expression of Foxp3 and TGF-? mRNA.Results:(1) Following treatment with the altered CⅡ263-272 peptide,arthiritis scores were reduced and body weight was increased.The mean arthritis scores of rats treated with altered peptide,wild peptide and PBS were 2.50?2.43,4.50?2.23 and 6.33?2.73,respectively.The altered peptide could retard the histologic lesion of the joints.(2) The titers of anti-CⅡ antibodies IgG and IgG1 in the three groups were similar,but the IgG2a in altered peptide-treated rats decreased markedly as compared with PBS-treated rats(0.56?0.19 vs 0.95?0.29,P

Objective To detect PAH gene mutations in classical PKU patients by HRM analysis. MethodsMutation scanning of PAH gene were performed in 17 classical PKU patients by HRM analysis ( LightScanner), covering the 13 exons and exon-intron boundaries. The HRM results were further confirmed by DNA sequencing, and the sensitivity and specificity of HRM method in PKU diagnosis were also evaluated. In addition, prenatal diagnosis was performed in two fetuses at risk for classical PKU. Results In the 17 patients, two mutations were identified in 16 patients, three mutations were identified in 1 patient.In this subject, a total of 22 different pathogenic mutations : 194V( c. 280A > G), IVS4nt-1 G > A( c. 442-1G > A), R158Q( c. 4736 > A), Q160X( c. 478C > T), W187X( c. 561G > A), E6nt-96A > G( c. 611A >G), G239D( c. 716G > A), R241 C( c. 721C > T), R243Q( c. 728G > A), G247R (c. 739G > C), G247V (c. 740G>T), R261X(c. 781C >T), PR261Q(c. 782G > A), H264R (c. 791A > G), F302fsX39 (c. 904delT), E305K( c. 913G > A), G312V( c. 935G > T), Y356X( c. 1068C > A ), V399V ( c. 1197A >T), R408Q(c. 1223G > A), T418P(c. 1252A > C) , A434D(c. 1301C > A), 3 silent mutations Q232Q (c. 696G > A), V245V(c. 735G > A), L385L(c. 1155C > G), and one single nucleotide polymorphism rs2280615 ( c. 402A > C) were identified, of which 194V ( c. 280A > G), Q160X ( c. 478C > T), H264R (c. 791A > G), G312V( c. 935G > T) and E305K ( c. 913G > A) were novel mutations identified in PAH gene. The prenatal diagnosis results of the two fetuses : one was diagnosed as normal, the other was diagnosed as a carrier. In this study, the sensitivity and specificity for mutation detection by HRM were 100%, and the HRM results were consistent with DNA sequencing results. Conclusions HRM analysis is a simple,accurate, rapid, high-throughput and low-cost genetic analysis approach. It could be applied to mutation scanning of classical PKU of PAH gene and rapid prenatal diagnosis in parents with known mutations.
3.Effect of X-ray irradiation on the neurites growth of primary hippocampal neurons

Objective To investigate the effect of X-ray irradiation on neurites growth of primary hippocampal neurons in vitro.Methods Primary hippocampal neuron culture in vitro were irradiated with 0,2,4,8,10,12 Gy of X-rays.In the first day and the third day after irradiation,the cell death of primary hippocampal neurons was detected by MTT method,and the morphological changes of primary hippocampal neuronal neurites were detected with immunofluorescence staining method.Results In the first day and third day after irradiation,the cell death of hippocampal neurons increased significantly (F =123.068,43.370,P <0.05),but there were no significant difference among 4,8,10,and 12 Gy irradiation groups.Immunofluorescence staining showed that,in the first day after irradiation,the neurite length and total dendritic branch length (TDBL) were significantly changed (F =9.169,7.856,P <0.05),and in the third day after radiation,the neurite length,TDBL and total dendritic branch tip number (TDBTN) were also altered (F =23.797,6.565,6.021,P < 0.05).Conclusion X-ray irradiation can inhibit the growth of neurites in the primary hippocampal neurons in vitro.
4.Asymmetric Morita-Baylis-Hillman Reaction of Conjugated Nitroalkene with Activated Aldehyde Catalyzed by Cinchona Alkaloids

Objective:To develop a catalytic system for the asymmetric Morita-Baylis-Hillman ( MBH) reaction of conjugated ni-troalkene with activated aldehyde, and screen out the chiral catalysts with high activity and enantioselectivity. Methods: Totally 21 chiral organocatalysts were applied in the asymmetric MBH reaction ofβ-nitrostyrene with ethyl glyoxylate, and the ee value was deter-mined by chiral HPLC. The effects of temperature, solvent and substrate ratio on the catalytic reaction were investigated. Results: In the presence of cinchona alkaloid catalyst (DHQ)2AQN, β-nitrostyrene reacted with ethyl glyoxylate in toluene at 0℃ affording the MBH adduct in 60% yield with good enantioselectivity (up to 56.9% ee). Conclusion: The bis-cinchona alkaloids with aromatic bridging group are the efficient catalysts for the asymmetric MBH reaction ofβ-nitrostyrene with ethyl glyoxylate, and moderate isolated yield and enantioselectivity are obtained.
5.Comparison of characteristics between indocyanine green and fundus fluorescence angiography in patients with age-related macular degeneration

Objective To compare the characteristics between indocyanine green angiography (ICGA) and fundus fluorescence angiography ( FFA ) in patients with age-related macular degeneration ( ARMD ). Methods Twenty-five cases ( 35 eyes ) were diagnosed as exudative ARMD by ocular examination. Their age ranged from 60 to 81 years. The visual acuities were 1.0 at FC/30 cm before eye. We analyzed and compared the characteristics of FFA and ICGA in these patients. Results 5 eyes ( 14.3% ) with classic choroidal neovascularization ( CNV ) were found by FFA examination. 15 eyes ( 42.9% ) with classic CNV were found by ICGA examination. Of 20 eyes with occult CNV considered by FFA, 7 eyes had classic CNV by ICGA. Of 3 eyes considered as serous retinal pigment epithelial detachment ( PED ) without classic CNV by FFA, 1 eye had serous PED with classic CNV and 2 eyes had classic CNV by ICGA.1 eye was diagnosed as serous PED with CNV by FFA, and as occult CNV by ICGA. Conclusions ICGA raise detectability and accuracy of CNV in ARMD compared with FFA. It is contributive to early treatment in ARMD.
6.Analysis of the characteristics of juvenile primary Sj(?)gren's syndrome with renal damage

Objective To study the renal damage of patients with juvenile primary Sjgren's syndrome (pSS)and its clinical manifestations,pathologic characteristics with biopsy,treatment and prognosis.Methods Ten patients with juvenile pSS complicated with renal impairments were retrospectively analyzed.Data of these 10 patients were compared with those without renal impairments.Results Ten patients complicated with renal impairments in 24 patients with juvenile pSS,9 of them presented with type I renal tubular acidosis(RTA), 5 with hypokalemia paralysis,3 with calcification of the renal tissue,3 with positive urine protein.1 with dia- betes insipidus.There was no significant difference between patients with renal impairments and those without. Three patients underwent kidney biopsy that showed chronic interstitial nephritis(CIN)with extensive lymp- hoplasmie cell infiltration.Two patients had glomerular lesions and one of them was diagnosed as pSS over- laped with systemic lupus erythematosus(SLE).Steroid and immunosuppressive agents had significantly alle- viated symptoms and the hypergammaglobulinemia was significantly improved.Conclusion Renal impairment may be the major complication in juvenile pSS.The major clinical manifestations are RTA and the glomeruli are involved occasionally.Treatment with steroid anti immunnsuppressive agents should be given to those who have evidence of systemic involvement.
7.Significance of detection of the autoantibodies in patients with rheumatoid Arthritis

Objective To explore the sensitivity of antibodies against RF,APF,AKA,anti-CCP in rheumatoid arthritis (RA) and analyze the relationship between these autoantibodies in RA.MethodsFive hundreds of RA patients were studied.Immunoturbidmetry was used to measure the RF-IgM.The enzyme-linked immunosorbent assay (ELISA) was used to measure the HRF-IgG,HRF-IgA and anti-CCP.Indirect immunofluorescence was used to measure the APF and AKA.ResultsThe seropositive rates of RF-IgM,anti-CCP,APF,HRF-IgG,AKA and HRF-IgA were 78.0%,70.8%,51.8%,48.4%,45.0% and 20.0% respectively.RF-IgM could not be detected in 110 patients while the seropositive rates of anti-CCP,APF,AKA,HRF-IgG and HRF-IgA were 41.8%,27.3%,22.7%,15.5% and 1.8% respectively.Anti-CCP could not be detected in 146 patients while the seropositive rates of RF-IgM,HRF-IgG,APF,AKA and HRF-IgA were 56.8%,24.7%,16.4%,8.9% and 7.5% respectively.The combined sensitivities of RF-IgM anti-CCP and APF were higher than others.The seropositive rates were 88.6%.ConclusionsRF-IgM and anti-CCP are sensitive antibodies for diagnosis of RA.Combinations RF-IgM with anti-CCP can significantly improve the sensitivity of diagnosis of RA.
9.Identification of liposoluble constituents in Yushu tablets by UPLC-ESI-IT-TOF/MS.

In order to identify the chemical constituents of Yushu tablets comprehensively, we studied the chemical constituents of CHCl3 extract from Yushu tablets by the ultra performance liquid chromatography-electrospray ionization-ion trap-time of flight mass spectrometry (UPLC-ESI-IT-TOF/MS). It showed that there were more than 100 compounds separated, and forty-nine peaks among these were identified on the basis of high resolution mass spectrometry data and literature data reported. Determination of twelve peaks was further confirmed by standard substances. These components assigned to the different plant sources mainly included phenylpropanoids, triterpenoids, quinones and m-trihydroxybenzene compounds. By analyzing the chemical components of CHCl3 extract from compound Chinese medicine Yushu tablets comprehensively, this study provided the foundation for studying chemical components, pharmacodynamic substance and quality control of Yushu tablets.

10.The clinical value of neutrophil to lymphocyte ratio in postmenopausal patients with type 2 diabetes

Objective To investigate the clinical value of neutrophil-lymphocyte ratio (NLR) in postmenopausal patients with type 2 diabetes mellitus (T2DM). Methods A total of 128 newly diagnosed T2DM female patients were randomly recruited from the Zhujiang Hospital of Southern Medical University from March to September 2015. According to the physiological state, the patients were divided into menopausal group (n=66) and non-menopausal group (n=62). Fifty-six healthy females were chosen as the control group. The values of blood pressure, blood lipid, fasting plasma glucose (FPG), fasting insulin (FINS) and white blood count (WBC), neutrophil count, percentage of neutrophils,lymphocytes count, and percentage of lymphocytes were detected and compared between three groups. The NLR and the HOMA insulin resistance index (HOMA-IR) were calculated. Results Age, systolic blood pressure (SBP), triglyceride (TG), FINS, percentage of neutrophil, NLR, HOMA-IR were significantly higher in menopause group than those of control group and non-menopause group (P<0.05). There were no significant differences in diastolic blood pressure (DBP) and FPG between menopause group and non-menopause group, but both were significantly higher than those of control group (P < 0.05). There were no significant differences in total cholesterol (TC), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), WBC and HbA1c between three groups (P>0.05). There was no linear correlation between NLR and HOMA-IR (rs=0.015, P > 0.05), HbA1c (r=0.030, P > 0.05). Conclusion NLR is of a certain significance in the assessment of inflammatory status and IR levels in postmenopausal women with T2DM, which may be related to the changes of autonomic nervous system.
