Objective To study the effect of Bcl-2 gene transfer on apoptosis of myocardium in mice heart transplantation rejection. Methods Models of mice neck heterotopic heart transplantation were set up, and were randomly divided into transplant group, Bcl-2 group and control group. Bcl-2 was transfected into isolated donor heart, and after 1 hour the donor heart was transplanted to the recipient. At 1st,3rd,5th and 7th day after transplantation, apoptotic index was evalulated by the percentage of mycardial cells with TUNEL positive staining and the protein expression of Bcl-2 was measured by immunohistochemical method. Results The apoptotic myocardial cells significantly increased at 3rd day and the apoptotic peak was at 7th day after transplantation in transplant group.The protein expression of Bcl-2 in Bcl-2 group significantly increased at 3rd day, the expression peak was at 5th day after transplantation, and high level expression remained at 7th day. Objective:To study the dissolution behavior of panax notoginseng saponins R1 , ginseng saponin Rg1 and ginseng sapo-nin Rb1 in Xuesaitong soft capsules in two different dissolution media. Methods:The dissolution test conditions were confirmed accord-ing to“quality of medical drugs information set” of Japan and the dissolution media were chosen in accordance with the physicochemi-cal properties of Xuesaitong soft capsules, the in vitro dissolution of Xuesaitong soft capsules was inspected respectively in pH 6. 8 phosphate buffer and water. The dissolution rate was determined by HPLC-ELSD, and the similarity of the release curves for the three components was compared with similarity factors ( f2 ) method. Results: The assay displayed a good linearity over the concentration range of 0. 8-16. 0 mg·L-1(r=0. 999 6), 3. 0-60. 0 mg·L-1(r=0. 999 8) and 4. 0-80. 0 mg·L-1(r=0. 999 7) for panax notog-inseng saponins R1 , ginseng saponin Rg1 and ginseng saponin Rb1 , respectively. In pH 6. 8 phosphate buffer and water, f2 of ginseng saponin Rg1 and ginseng saponin Rb1 were both above 50 when compared with panax notoginseng saponins R1 . Conclusion:The meth-od is simple, accurate and reproducible in the dissolution determination of the soft capsules. The dissolution of ginseng saponin Rg1 and ginseng saponin Rb1 has similar characteristics with that of panax notoginseng saponins R1 in Xuesaitong soft capsules.

Objective:By observing effects of Dahuang Zhechong Pills on CD44,VEGF and the metastasis suppressor in mice with Lewis Lung carcinoma,to discuss its mechanism of anti-tumor metastasis preliminarily.Methods:The mice were divided randomly into five groups the day after modeling and each group included ten mice.According to the pre-test,high-dose,medium-dose and low-dose were quantitated with 0.18g/ml,0.09 g/ml,0.045g/ml.The model group was intragastric administrated with NS 0.2ml for ten days;the chemotherapy group was Intraperitoneal administrated 50mg/kg CTX on the first day.Observing and numerating the pulmonary metastases with nodules by pulmonary pressure slide film on the 12th day.Based on immunohistochemical method to detect the expression of VEGF and the activity detection by flow cytometry CD44,Its anti-tumor mechanism and the role of target were preliminarily studied.Results:The inhibition rates of lung spontaneous metastasis in experimental groups of high(H),medium(M),low(L)dosages and the chemotherapy group were respectively 57.44 percent, 55.32 percent,38.72 percent,60.00 percent.CD44,respectively,values were 0.59?0.86,0.21?0.28,0.33?0.31,1.36?0.52; the experimental group(M)compared with model group,VEGF expression was significantly reduced,there was a significant difference(P0.05).Conclusion:Dahuang Zhechong Pills reduced the lung metastases incidence of C57BL mice with Lewis lung carcinoma and prolonged their lives.These results were obvious in the medium-dose experimental group.Medium-dose of Dahuang Zhechong Pills had effects on C57BL mice with Lewis lung carcinoma,whose CD44 activity were inhibited and the expression of VEGF was reduced.

Objective To observe protective effects of four active liposoluble alkaloids of a Chinese herb, lotusine (Lot), liensinine (Lien), isoliensinine (Iso) and neferine (Nef) of embryo loti (the green embryo), against H2 O2-induced oxidative damage on human umbilical vascular endothelial cell ECV-304.Methods The protective effects of Lot, Lien, Iso and Nef on the survival of normal and oxidatively damaged ECV 304 cells were studied by cell morphology observation and 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium (MTT) assay.The levels of nitric oxide (NO) and nitric oxide synthase ( NOS) were measured using colorimetric assay.Results Lot, Lien, Iso and Nef did not affect cell morphology and cell viability of normal ECV 304 cells.The survival of oxidative damaged vascular endothelial cells was rescued by incubating with Lot at 100μmol/L, and Lien, Iso and Nef at 0.1 μmol/L.The proliferative activity of medicated groups increased to 112.8%, 129.3%, 125.6 and 118.2%, respectively (P<0.01 or 0.05), relative to that of the group with H2O2 induced oxidative damage.The four alkaloids restrained oxidative injury of endothelial cells induced by H2 O2 and the protective influences were similar with captopril, which served as a positive control.Each alkaloid except Lot reduced intercellular space and increased the connections of oxidative damaged cells, concomitant with more recognizable cell borders.Lien, Iso and Nef also increased the concentration of NO ( P<0.05 ) .Besides, all of the four alkaloids activated NOS in damaged vascular endothelial cells ( P<0.05 ) . Conclusion The four alkaloids of embryo loti, especially Lien, Iso and Nef, have certain protective effects against H2 O2-induced oxidative damage on vascular endothelial cells.The protective mechanism may be promotion of NO release through the increase of NOS production.

Objective To study and contruct competency characteristic evaluation models of nursing supervisors in secondly hospitals.Methods Using the matron semi-structured interviews,literature review,questionnaire survey and expert questionnaires to discuss the competency of nursing supervisors in secodary hospitals.Results We found that the competency model of nursing supervisors in secondary hospitals could better reflect the competency of nursing supervisors.The model had 16 competencies which included knewleage skills.management and individual characters.Conclusions Through the study of the model of competency nursing supervisors in secondary hospitals,we form a set of standard for nursing supervisor selection based on their competency.The standard can help to select the most suited nursing supervisors who have great potentials.

Comparing the Psychiatry teaching among Jinan University,Sum-Yan-San University and The Chinese University of Hong Kong,we found the time,contents and methods of teaching were significantly different among them,so we suggested that Jinan University should reconstruct teaching mode,raise teaching time,optimize teaching contents,change practice teaching methods and examination way in psychiatry teaching.

Objective:To investivate the relationship of serum estradiol and oxidative stress with microcirculation resistance in women with syndrome X.Methods:A total of 120 patients with syndrome X who were hospitalized in the Department of Cardiology in Guangzhou First People's Hospital from January 2015 to January 2019 were enrolled. All patients underwent coronary angiography and pressure wire examination and were divided into two groups according to the index of microcirculation resistance (IMR). Forty healthy people in the medical examination center were used as controls in the same period. The level of serum estradiol, interleukin-1 (IL-1), tumor necrosis factor-α (TNF-α), superoxide dismutase (SOD) were measured among three groups.Results:The rate of diabetes mellitus in high IMR group was higher than that in control group and low IMR group ( P<0.05). There was no significant difference between low IMR group and control group ( P>0.05). The levels of serum estradiol and SOD were significantly lower in high IMR group than those in low IMR group and control group. The levels of IL-1 and TNF-α were significantly higher in high IMR group than those in low IMR group and control group ( P<0.05). These indexes have the same relationship between low-IMR group and control group ( P<0.05). The level of serum estradiol was negatively correlated with the levels of IL-1 and TNF-α in high and low IMR groups and positively correlated with the level of SOD in these groups. Multivariate logistic regression analysis showed that diabetes, low serum estradiol level, low SOD level, high IL-1 level, high TNF-α level were the independent risk factors for microcirculation resistance in women with syndrome X ( P<0.05). Conclusions:The decreased serum estradiol is an important factor for coronary microcirculation disorders in women with syndrome X. The decrease of serum estradiol level leads to the loss of the corresponding antagonistic effect in oxidative stress state, which may be one of the important mechanisms of the formation and progress of coronary microcirculation disorder.

OBJECTIVETo explore activity laws of mitochondrial complex II in patients of deficiency-cold syndrome (DCS) and deficiency-heat syndrome (DHS) under various ambient temperatures.

METHODSSubjects were recruited by questionnaire and expert diagnosis from grade 1 - 3 undergraduates at Henan College of Traditional Chinese Medicine in November 2012, and assigned to a normal control group, the DCS group, and the DHS group, 20 in each group. Their venous blood samples were collected at two different temperature conditions. Activities of mitochondrial complex II were measured by spectrophotometry.

RESULTS(1) Comparison of mitochondrial complex It under various ambient temperatures: Compared with room temperature in the same group, activity values were all increased in the normal control group at cold temperature with significant difference (P <0.05), but there was no significant difference in the DCS group and the DHS group (P >0. 05). Compared with the normal control group, activity values of complex H were reduced in the DCS group at cold and room temperatures with significant difference (P <0.05). Compared with the DCS group, activity values of complex It were increased in the DHS group with significant difference (P <0. 05). (2) Changes of adjustment rates: Compared with room temperature, the adjustment rate all rose at cold temperature in the normal control group and the DHS group with significant difference (P <0.05), but with no significant difference found in the DCS group (P >0. 05). Compared with the normal control group at the same temperature, the adjustment rate in the DHS group and the DCS group was all reduced at cold and room temperatures with significant difference (P <0. 05). There were no significant difference in the adjustment rate between the DHS group and the DCS group (P > 0. 05).

CONCLUSIONSEnvironment temperature can affect the activity of mitochondrial complex II with different influence degrees on different syndrome types of people, but its change trend are basically identical.
