The pharmacokinetics of ketamine were studied in 8 preschool children,aged 3.5～6 years. The ketamine was administrated by I. V. infusion at a dose of 3mg?kg~(-1) in one hour constantly. The plasma concentrations of ketamine were measured by high performance liquid chromatography. The disposition of intravenous infusion with ketamine could be described by two-compartment open model, and the variables of pharmacokinetics of ketamine were as follows: T_(1/2?)=0.15?0.04 hours,T_(1/2?)=2.84?0.44 hours,Cl=0.34?0.06L?kg~(-1)?h~(-1), AUC=9.21?1.00mg/L?h, and V_d=0.66?0.29 L?kg~(-1). This suggests that intravenous infusion of ketamine at 3mg?kg~(-1)?h~(-1) is safe for the maintenance of general anesthesia in preschool children, attention must he paid to the children anesthetized with long period of ketamine infusion because recovery may be delayed due to the prolonged elimination half-life.

Objective: To study the expressions of CD147and MMP9 in the lung metastasis model of breast cancer in BCML-TAII99 mice and their correlation with tumor growth and metastasis thereof. Methods: The breast cancer model with lung metastasis was established in BCML-TAII99 mice. The immunohistochemical staining method(SP method) was used to analyze the expressions of CD147 and MMP9 in mice lung metastasis tissue. The relationship between the tumor growth and the metastasis was analyzed. Results: The positive rates of CD147 and MMP9 were 63.33%（19/30）and 53.33%（16/30）respectively in mouse models. There was significant difference in positive rate of expressions between the metastasis group and the non-metastasis group（χ~2= 6.238， P ＝ 0．013； χ~2＝ ＝5．129， P ＝ 0．024). There was a statistically significant positive correlation between the expression of CD147 and MMP9（r = 0．786， P ＝ 0.025). Conclusion: The expression of CD147 may play a crucial role in infiltration and metastasis of breast cancer by inducing the expression of MMP9.

Objective: To determine the influence of ?-hydroxybutyric sodium (7-OH)on the pharmacokinetics of ketamine infusion in preschool children. Method: Sixteen patients were randomly assigned to group Ⅱ (Ketamine) and group Ⅱ (Ketamine plus ?-OH). After trachea intubation,0.05% ketamine was infused in both groups at 3mg?kg~(-1)?h~(-1) and ?-OH 125mg?kg~(-1) only in group Ⅱ. Objective To investigate the expressions of estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER-2) and relationships of these expressions with the clinicopathology in breast cancer patients aged 70 years old and over. Methods The expressions of ER, PR and HER-2 were detected by immunohistochemical staining from 124 elderly breast cancer patients, and the relationships of these expressions with pathological classification, tumor size, lymph node metastasis and TNM stage were analyzed. Results Among 124 patients, the positive expression rate of ER and PR was 75.0% (93/124) and 71.0% (88/124),respectively. They were not associated with pathological classification, tumor size, lymph node metastasis or TNM stage (all P＞0. 05). The positive expression rate of Her-2(+++) and Her-2 (++) was 11.3% (14/124) and 24.2% (30/124), respectively. And the expression of Her-2 was significantly associated with tumor size, lymph node metastasis and TNM stage (P＜0. 05) but not with pathological classification (P＞0. 05). Conclusions The positive expressions of ER and PR are not correlated with the clinicopathology in elderly breast cancer patients. Her-2 overexpression is positively associated with tumor size, lymph node metastasis and TNM stage.

Objective:To establish an HPLC method for the determination of norisoboldine in Suoquan capsules. Methods: The chromatographic procedure with acetonitrile-water containing 0. 5% folic acid and 0. 1% triethylamine (10∶90) as the mobile phase was carried out on an Ecosil C18 HPLC(150 mm ×4.6 mm,5 μm). The flow rate was at 1.0 ml·min-1, the detection wavelength was 280nm and the column temperature was 30 ℃. Results: The linear range of norisoboldine was within 38. 56-578. 40 ng ( r =0. 999 2), the average recovery of norisoboldine was 99. 9%(RSD=1. 7%, n=6). Conclusion:The method is highly sensitive, re-liable and accurate, and can be applied in the determination of Suoquan capsules.

Objective To evaluate the therapeutic effect of narrow band-ultraviolet B (NB-UVB) alone or in combination with interferon-alpha-2b (INF-alpha-2b) for mycosis fungoides (MF),and to assess the correlation between the therapeutic effect and peripheral blood regulatory T (Treg)/T helper type 17 (Th 17) cells.Methods Thirty-three patients with stage ⅠA to ⅡA MF were randomly divided into two groups:NB-UVB group (n =15) receiving NB-UVB radiation alone,combined group (n =18) treated with NB-UVB radiation and intramuscular injection of INF-alpha-2b.Ten healthy volunteers were selected as the control group.Peripheral blood samples were collected before and 9 months after the start of treatment.Flow cytometry was performed to determine the percentages of Treg cells and Th17 cells.Statistical analysis was carried out by using t test,one-way analysis of variance,and Fisher's exact test.Results The average treatment duration was 9 months among these patients.Therapeutic outcomes were significantly better in the combined group than in the NB-UVB group (P =0.023).Among the 15 patients in the NB-UVB group,6 achieved complete remission,3 partial remission,6 showed no response;of the 18 patients in the combined group,12 experienced complete remission,5 partial remission,and 1 showed no response.Before the treatment,the percentages of both Treg and Th17 cells in peripheral blood were significantly higher in the NB-UVB group and combined group than in the control group (both P ＜ 0.05),but similar between the NB-UVB group and combined group (both P ＞ 0.05).After the treatment,the percentages of both Treg and Th17 cells in the NB-UVB group and combined group significantly decreased compared with those before the treatment,but were still higher than those in the control group (both P ＜ 0.05).Additionally,the degree of decrease in the percentages of Treg and Th17 cells was significantly greater in the combined group than in the NB-UVB group (both P＜ 0.05).The seven patients with no response also showed a significant decrease in the percentage of Treg cells (P ＜ 0.05),but no obvious changes in that of Th 17 cells (P ＞ 0.05) after the treatment.Conclusions The therapeutic effect of NB-UVB radiation combined with intramuscular INF-alpha-2b is superior to that of NB-UVB radiation alone for MF,which may be associated with the degree of decrease in peripheral blood Treg and Th 17 cells.

Using Vibrio fluvialis Ⅱ as host bacteria, 19 bacteriophages have been isolated from the 76 samples which were collected from Haliotis discus hannai ～growing seawater in Dalian marine culture company Dalian, liaoning province from May in 1996 to August I 1997. Ultrastructure of 19 bacteriophages were observed with electron to Bradley the results showed that of these bacteriophages belonged to Bradley A type, they have hexagonal heads of bacteriophages were identified with VP1,VP2,VP4,VP8 as representatives respectively. The phages remain stable at pH6. 0～10. 0, moreover VP2,VP4 and VP8 are rather stable at basic pHs. Although the characterization of heat inactivation course of VP4 is different from others, four phages are sensitive to heat and inactivated at 80℃ in 5 minutes. One step growth experiment showed that the eclipse period of VP1,VP2,VP4,VP8 are sensitive to heat and the eclipse period of VP1, VP2, VP4, VP8 are 42, 30, 46, 28 minutes. In this experiment we have isolated at least four different types phages, it suggest that in fact there is a population of phages in the seawater environment. The result of this study provided a way to find the potential value of phages as an indicator of pathogenic microorganisms Vibrio fluvilis Ⅱ in marine environment.

Objective To investigate the neural proliferation , differentiation and apoptosis of the developing spinal cord of the mouse and to discuss the mechanism of spinal cord ’ s development .Methods 5-Bromodeoxyuridine ( BrdU) assay was used to mark the proliferative neural stem cells , and the immunofluorescent stainings ( DCX, NeuN and Caspase8) were carried out to visualize the newborn neurons , mature cells and apoptotic cells in the spinal cord with 173 mice arrange from E18 to P90.Results BrdU positive neural stem cells appeared evenly in the spinal cord at early days . With age increasing , the neural stem cells differentiated into neuroglial cells and neurons .The newborn neurons in the subventricular zone migrated toward the intermediate zone ( putative gray matter ) and differentiated into mature neurons gradually .With neurons ’ concentrating towards the center , the gray matter formed an “H” shape .In the meantime , with neural differentiation , some apoptotic neurons appeared among the newborn neurons and mature neurons . Double immunostaining showed that most apoptotic neurons were newborn neurons , suggesting the neuroapoptosis more likely occurred in newborn neurons .The statistical data showed that the number of DCX , NeuN and Caspase-8 positive cells reduced with age increasing , suggesting neural differentiation and neuroapoptosis decreased during spinal cord ’ s development .Conclusion Neural proliferation , neural differentiation and neuroapoptosis occur in developing spinal cord . They work together to regulate the formation and development of the spinal cord .

Objective To assess the effect of different treatment complex on portoazygous shunt in portal hypertensive patients. Methods Patients (115 cases) with esophageal varices were randomized to receive either endoscopic variceal ligation ( EVL) alone (54 cases ) , pericardial devascularization procedure (PDP) alone(30 cases) or a combination of EVL and PDP(31 cases) for variceal eradication. Esophageal vascular structures were examined with miniature ultrasonic probe and azygos blood flow (ABF) was measured with color Doppler ultrasonography. Results Esophageal varices were obliterated and collateral veins remained unchanged in patients treated by EVL, esophageal varices were diminished in size and collateral veins were obliterated by PDP and both esophageal varices and collateral veins were obliterated by the combination of EVL and PDP. EVL,PDP alone and a combination of the two modalities decreased ABF by 31% ,32% and 43% respectively. Conclusions The combination of EVL and PDP most effectively blocks esophageal collateral shunts.

Objective To investigate the relationship between gene polymorphism of transcripion factor 7-like 2 (TCF7L2) at positions rs290487, rs11196205, rs11196218 and gestational diabetes mellitus (GDM) in Chinese women.Methods In 1140 unrelated pregnant Northern Chinese women (335 women with GDM, 158 gestational cases with impaired glucose tolerance and 647 pregnant non-diabetic controls) ,three single nucleotide polymorphisms (rs290487, rs11196205, and rs11196218) in the TCF7L2 gene were genotyped using ligase detection reaction (LDR).In the present study, cases with GDM and impaired glucose tolerance (IGT) were indistinguishable clinically and biochemically, and were combined into case group.Results The frequency of C allele of rs290487 was 41.6% in case group, being significantly higher than that in control group (36.3%, P=0.012).There was significant difference in the frequency of CC genotype between case group and control group (18.7% vs 14.0%, P=0.033).Compared with T allele carriers, CC genotype carriers had a 1.418-fold increased risk of GDM (95% CI 1.028-1.955).After adjusting for age, body mass index, family history of diabetes,systolic blood pressure,and diastolic blood pressure, pregnant women with CC genotype carriers of rs290487 were more prone to hyperglycemia compared with the T allele carriers (OR 1.518, 95% CI 1.064-2.166).Conclusions The TCF7L2 rs290487 variant may contribute to the genetic predisposition to GDM.CC genotype is likely to be associated with an increased risk of GDM in the pregnant Chinese women.
