1.Latest progress of chimeric antigen receptor modified T cells in the treatment of hematological malignancies

BACKGROUND:In recent years, the use of chimeric antigen receptor modified T cells (CAR-T cells) has achieved good results in the treatment of hematological malignancies. OBJECTIVE:To summarize the CAR-T technology, and its anti-tumor mechamism, progress in the treatment of hematological malignancies as well as its safety and coping strategies. METHODS:A search of PubMed, CNKI, Wanfang by computer was performed for articles related to CAR-T published from January 2010 to January 2016, using the keywords of CAR-T in English and Chinese, respectively. RESULTS AND CONCLUSION:The main principles that CAR-T cells fight against tumors are as follows: (1) resistance to immune escape through down-regulation of MHC; (2) production of interleukin-6, -10 indirectly influences the growth of tumor cells; and (3) tumor microenvironment changes inhibit tumor growth. The use of CAR-T in the treatment of hematological malignancies has been developed by leaps and bounds in recent years. Current studies concerning B cell lymphoma mainly focus on anti-CD19 CAR-T cells that have certain therapeutic effects on acute myeloid leukemia and acute lymphoblastic leukemia. However, its safety and effectiveness have yet to be studied.
2.Effects of Sodium Arsenite on mRNA Expression of GTP Cyclohydrolase and 6-pyruvoyl-tetrahydropterin Synthase in Chang Liver Cells

Objective To study the effects of sodium arsenite on the mRNA expression of GTP cyclohydrolase (GTPCH) and 6-pyruvoyl-tetrahydropterin synthase (PTPS) in Chang liver cells. Methods Human Chang liver cells were cultured with sodium arsenite at doses of 0, 50, 200 and 400 ?mol/L for 12 hours. Cell viability was tested by MTT assay, and the expression of mRNA of GTPCH and PTPS was detected by RT-PCR. Results Cell viability in 50 ?mol/L group was almost the same as that in control, while the viability in the other two groups were significantly lower than that in control. All of the mRNA expression levels in the three groups were significantly lower than that in control with dose-dependent manner. Conclusion The down-regulated expression of GTPCH and PTPS, the main enzymes of the synthesis of BH4, induced by sodium arsenite exposure may be related with arsenic related skin depigmentation.

Objective To evaluate the efficacy of PTCRA+ PTCA in patients with complicated coronary disease Methods PTCRA+PTCA were performed on 16 lesions in 14 patients in our hospital Results In 14 lesions, PTCA was performed after direct PTCRA in order to achieve opital angigraphic result; PTCRA was performed in 5 coronary artery lesions after failed PTCA 14 patients (100%) had successful result Conclusion The combined use of PTCRA+PTCA can improve the success rate during revasculrization for patients with complicated coronary artery disease

5.Study on the specific autoantibody in nonimmune thrombocytopenia in malignant tumor

Objective To study the possibility of the exist of specific antibody against platelet in tumor patients and to evaluate the value of MAIPA in measure of specific antibody against platelet in malignant tumor patients with nonimmune thrombocytopenic purpura. Methods Anti- GPⅡb/Ⅲa and anti- GPⅠb/Ⅸ autoantibodies were detected by a modified monoclonal antibody immobilization of platelet antigens assay(modified MAIPA). Results The positive rate of the platelet antibody in nonimmune thrombocytopenic purpura is very low with MAIPA. Conclusions The platelet GP- specific autoantibody detection is considerably important in discriminating immune from nonimmune thrombocytopenia.
6.Studies on the lymphatic tropism and lymph cells apoptosis of cisplatin-nano carbon suspension in the rats

Objective To research the lymphatic tropism and lymph cell apoptosis of cisplatin-nano carbon suspension in rats with the aim of proposing a new way for chemotherapy.Methods A total of 72 Wistar rats were randomly divided into two groups.For the experimental group,cisplatin-nano carbon suspension 0.3 ml (4 mg/ml) was injected subcutaneously into Wistar rats' plantar.For the control group,cisplatin 0.3 ml (4 mg/ml) was injected intravenously.Cisplatin concentration in the inguinal lymphatic tissue and plasma was determined by high performance liquid chromatography (HPLC) at 1,2,3,4,5 and 6 hours after drug administration.The apoptosis of lymph cell was detected by terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end labeling assay (TUNEL).Targeting ability were evaluated and compared by targeting index (TI),selecting index (SI) and relative extraction efficiency (RE).SPSS 17.0 statistical software was used to analyze the differentiation of the cisplatin concentration and apoptosis index (AI) among various groups.DAS software was used to evaluate the lymphatic tropism.Results The cisplatin concentration of lymphatic tissue in experimental group were respectively (1.03±0.32),(3.00±0.91),(2.20±0.73),(1.56±0.38),(1.30±0.74) and (0.78±0.34) μg/g after administration 1,2,3,4,5 and 6 hours,while in control group were (0.49±0.21),(1.02±0.70),(0.59±0.50),(0.56±0.21),(0.47±O.18) and (0.36±0.13) μg/g,in which there were significant difference at every times (all P<0.05) except at 1 hour (P=0.173).The cisplatin concentration in plasma were significant higher in the experimental group than those in the control group at various times (all P<0.05),the former were respectively (0.57±0.28),(1.22±0.45),(0.61 ±0.18),(0.51 ±0.13),(0.45 ±0.13) and (0.40±0.07) μg/ml,while the latter were respectively (3.12± 0.33),(4.09± 0.48),(2.56 ± 0.38),(2.05 ± 0.13),(1.81 ± 0.28) and (1.44± 0.40) μg/ml.Values of TI were respectively 2.12,2.93,3.73,2.78,2.76 and 2.19 and SI were 1.80,2.45,3.63,3.07,2.86 and 1.93.Value of RE was 2.86.The AI in experimental group were respectively (16.5±5.2)%,(30.2±2.8)%,(51.7±4.3)%,(69.8±3.2)%,(80.1 ±4.3)% and (89.7±8.5)%,while in control group were respectively(1.3±0.8)%,(2.4±1.7)%,(3.2±1.1)%,(3.9±2.6)%,(5.1±2.1)% and (6.3±2.3)%,in which there were significant difference at every points (all P<0.05).Conclusions The nano carbon has the character of lymphatic tropism,and could send cisplatin to lymphatic tissue to achieve a higher concentration.The trait may break a new way for chemotherapy targeting lymph metastasis.
7.Change in brain neurosteroid level of rats in morphine addiction and stress-induced addiction relapse condition

AIM To investigate if morphine addiction and relapse to morphine-seeking is related to the change in neurosteroid levels in the brain of rats. METHODS Rats were injected ip morphine (5 mg·kg-1·d-1, 18:00-20:00) and trained in conditioned place preference (CPP) box, once daily for 10 d. CPP test was performed 24 h after the last training. After discontinuation of training for 7 d for CPP extinction, then intermittent and inescapable foot-shock (FS, 0.5 mA, 0.5 s on, 40 s off, 15 min) was applied to rats as the priming stimuli for relapse. CPP test was performed 2 h after FS. When CPP test finished, rats were decapitated and the levels of neurosteroids were analyzed using gas chromatography/mass spectrometry. RESULTS CPP was established when rats were injected morphine and trained for 10 d. At the same time, the levels of pregnenolone and allopregnanolone in the brain tissues of rats were significantly increased. When CPP was reinstated in morphine-treated rats by FS-stress after 7 d CPP extinction, the levels of dehydroepiandrosterone and dehydroepiandrosterone sulfate were significantly increased. CONCLUSIONThe development of morphine addiction and relapse may be related to endogenous neurosteroids in rat brain tissues.
8.Brief introduction of acupuncture needling and teaching keypoint.

We summarized our accumulated clinical and teaching experiences and explored the regularity of acupuncture needling and teaching. It is of great importance in pressing hand during inserting needle. Stroking and pressing are two crucial parts which deserve more attention, and seldom useage of pressing hand should be abolished. Operating hand needs practice before inserting needle, while it should fully relaxed during inserting. Blending "touching", "stretch" "gathering" "erupting" and "advancing" in single moment, applying appropriate dynamic mode of inserting needle such as "join 3 forces as one" "3 points in a line" expertly and naturally. In addition, enough attention should be paid on "altering direction" and "shifting point". Inserting deftly and powerfully, no/slight sensation, deqi when inserting needle are the highest reflection as an acupuncturist.

9.Correlation of TFRC polymorphism with the susceptibility and clinicopathologic phenotypes of IgA nephropathy

Objective:To explore the association of its polymorphism of TFRC with the susceptibility, clinical and pathologic phenotypes of IgA nephropathy. Methods: A total of 380 patients with IgA nephropathy and 250 normal controls were enrolled in the study. The regions with 424G/A and -5184C/T polymorphism sites of TFRC were amplified by PCR from genomic DNA and then the PCR-RFLP were performed by restriction enzymes, BanⅠ and BsmA Ⅰ, respectively. The genetic association of genotypes with the clinical and pathologic phenotypes was analyzed. Results: The distribution of frequency in TFRC was consistent with Hardy-Weinberg equilibrium; however, we found no significant difference in genotypes distribution between patients and controls. There were no differences between genotypes in age, blood pressure, 24 h urine protein excretion, serum creatinine, creatinine clearance and serum IgA. 424G/A and -5184C/T polymorphisms were associated with immunofluorescent intensity of IgA deposit in mesangial area, though there was no difference in pathological lesions evaluated by HAAS grade. Conclusion: The polymorphisms of TFRC in 424G/A and -5184C/T sites were not associated with susceptibility to IgA nephropathy, but associated with density of immunofluorescence of IgA in mesangium in our large population based Chinese patients. The association of IgA nephropathy and other polymorphism sites, as well as interaction between TFRC polymorphism and other genes' polymorphisms, neededs to be further investigated.
10.AngRem104 potentially regulates the transcription of fibronectin through the activation of promoter

AIM: To investigate the mechanism of AngRem104-mediated regulation of fibronectin gene in human mesangial cells. METHODS: A series of deleted FN promoter sequences was constructed, which fuse to a luciferase reporter gene, and then the potential active regions that respond to AngRem104 in the upstream regulatory sequence of human FN gene was screened by detecting the luciferase activity of promoter-reporter gene. RESULTS: The detection of relative luciferase activity revealed that there was no significant differences when the HMC were transfected with FN122-reporter gene together with sense AngRem104 construct, but the luciferase activity significantly increased when transfection of FN507-reporter gene construct together with sense AngRem104 construct. However, no increase in luciferase activity was observed when transfection of FN1280-reporter gene construct together with sense AngRem104 construct. The potential regulatory region responds to AngRem104 is in the upstream sequence (-122 to -507) of human FN gene. CONCLUSION: Our preliminary study provided the evidence that AngRem104 may mediate the transcription of the FN gene via the activation of its promoter.
