2.Nonmyeloablative stem cell transplantation followed by donor stem cell infusion for the treatment of nonmalignant haematologic diseases

Objective To evaluate the efficacy and complications of nonmyeloablative stem cell transplantation(NST) for the treatment of nonmalignant haematologic diseases with high-risk rejection.Methods NST was performed for two patients with severe aplastic anemia(SAA) and one with beta-thalassemia major(TM).The protocal was designed on transplantation of granulocyte colony-stimulating factor(G-CSF) primed allogeneic bone marrow cells combined with perpheral blood stem cells(PBSCs) for two SAA patients,with conditioning regimen based on anti-lymphocyte globulin and reduced dose of cyclophosphamide(CTX).One TM patient was performed transplantation of PBSCs with conditioning regimen of anti-T-lymphocyte globulin(ATG),fludarabine and reduced dose of busulfan.Cyclosporin A combined with methylprednisone was used for graft-versus-host disease(GVHD) prophylaxis.Donor stem cells infusion(DSI) were underwent for three patients at 78,99 and 44 days post transplant respectively.Results Three patients achieved engraftment successfully with mixed chimera and the lowest white blood cell(WBC) of 0.26?10~9/L,0.5?10~9/L and 1.26?10~9/L respectively.The absolute neutrophil count achieved more than 0.5?10~9/L and platelet count achieved more than 20?10~9/L at days of 12d,3d,0d and 1d,5d,0d post transplant in three patients,respectively.The haematopoiesis and chimera were improved after DSI without complications of infection and GVHD in three patients.Conclusion The stem cells engraftment is achieved successfully with donor stem cell infusion followed NST for the treatment of nonmalignant haematologic disease patients with high-risk rejection.
3.Application of ultra-pulse CO2 laser in blepharoplasty

Objective To explore the applied values of ultra-pulse CO2 laser in blepharoplasty by shortening procedure time, improving the surgical satisfactory result and alleviating the operative and postoperative hemorrhage. Methods 238 patients with bulging, redundant skin in the upper and lower eye lids, including brow lift (26 cases), double eye lid surgery (48 cases), and lower lash line incision (159 cases). Laser incision was implemented with the ultra-pulse CO2 laser system. Results Bleeding was scant in 238 cases and hence local swelling subsided quickly with excellent healing. No eetropion, he-matoma and diplopia ever occurred. 3 months to 3 years follow-up showed satisfactory for all the patients. The result was quite satisfied in clinical observation and postoperative follow-up. Conclusion Utilizing the ultra-pulse CO2 laser in blepharoplasty has confirmed the satifisfatory surgical results, decreasing the surgical time, and reducing the intraoperative and postoperative bleeding.
4.Expression of natriuretic peptide receptor A in the developing retina of the mouse

Objective Our purpose is to investigate the expression of natriuretic peptide receptor A (NPR-A) in the retina and to understand the NPR-A’ s functions during the mouse development .Methods Mice eyes were harvested from E16 ( embryonic day 16 ) to P90 ( postnatal day 90 ) . Total of 127 eyes were used in the study . Immunohistochemistries of NPR-A were carried out .Results During development , NPR-A was widely expressed in the retinal neurons .In the outer nuclear layer , NPR-A began to appear in the inner and outer projections of cone and rod cells at P7, but decreased at P14.From P30 afterward, it continued to express weakly .In the inner nuclear layer , NPR-A expressed in the dendrites of bipolar cells weakly from P 7 to adulthood , whereas no expression in horizontal cells .In the ganglion cell layer, NPR-A started highly to express in the ganglion cell bodies at E 16, and in the meantime, in the nerve fiber layer , ganglion cell axons , NPR-A was expressed highly from embryonic to adult .In the inner and outer plexiform layers, NPR-A was highly expressed at P14, but decreased gradually after P30.In addition, NPR-A also widely expressed in the inner protrusions of Müller cells.Conclusion NPR-A participates in the development of the retina , and may be the key molecule in the developing retinal neurons .Moreover, it plays an important regulatory role in the functional activity of Müller cells .
5.Development of the mouse spinal cord and neuroapoptosis

Objective To investigate the neural proliferation , differentiation and apoptosis of the developing spinal cord of the mouse and to discuss the mechanism of spinal cord ’ s development .Methods 5-Bromodeoxyuridine ( BrdU) assay was used to mark the proliferative neural stem cells , and the immunofluorescent stainings ( DCX, NeuN and Caspase8) were carried out to visualize the newborn neurons , mature cells and apoptotic cells in the spinal cord with 173 mice arrange from E18 to P90.Results BrdU positive neural stem cells appeared evenly in the spinal cord at early days . With age increasing , the neural stem cells differentiated into neuroglial cells and neurons .The newborn neurons in the subventricular zone migrated toward the intermediate zone ( putative gray matter ) and differentiated into mature neurons gradually .With neurons ’ concentrating towards the center , the gray matter formed an “H” shape .In the meantime , with neural differentiation , some apoptotic neurons appeared among the newborn neurons and mature neurons . Double immunostaining showed that most apoptotic neurons were newborn neurons , suggesting the neuroapoptosis more likely occurred in newborn neurons .The statistical data showed that the number of DCX , NeuN and Caspase-8 positive cells reduced with age increasing , suggesting neural differentiation and neuroapoptosis decreased during spinal cord ’ s development .Conclusion Neural proliferation , neural differentiation and neuroapoptosis occur in developing spinal cord . They work together to regulate the formation and development of the spinal cord .
7.Expressions of nitric oxide synthase in neonatal rat after hypoxia-ischemia and their changes while ganglioside GM_1 administrated

AIM: To study the neuroprotective effect and possible mechanism of ganglioside GM 1 on neonatal hypoxic-ischemic-encephalopathy(HIE). METHODS: A rat model of neonatal HIE was established, then the pathological changes and expressions of nitric oxide synthase (NOS) in the brain tissues were investigated in different periods after hypoxia-ischemia (HI) and the subseqent changes of the above results after GM 1 administrated. RESULTS: The damage of the brain exposed to HI were alleviated significantly after GM 1 administrated. The levels of NOS expressions in the brain tissue increased after HI. GM 1 could inhibit NOS expressions induced by HI. CONCLUSION: GM 1 may have some protective effects on neonatal HIE, and the possible mechanism is related to the partial inhibition of NOS expression.
8.Radiotherapy sensitization role of curcumin in Hela cells and its possible mechanisms

Objective To investigate the mechanisms of radiotherapy sensitization role of curcumin in cervical carcinoma Hela cells.Methods Western blot was used to detect expressions of p53, p21, Bax,and Bcl-2 proteins after cervical cancer Hela cells were treated.Results The expressions of p53 and p21 proteins were increased.The ratio of Bcl-2/Bax was decreased when curcumin and irradiation were applied to cells alone (P < 0.05).This change were much more obvious when irradiation and curcumin were combined than either of them was used alone(P <0.01).Conclusions The mechanisms of curcumin enhancing radiosensitivity on Hela cells might be due to upregulation of p53, p21, and Bax proteins and downregulation of Bcl-2 protein.
9.Novel Method for Determination of Lysozyme Based on Fluorescence Recovery of a Cationic Aluminum Phthalocyanine-Mucopolysaccharides Association Complex Used as a Red Emitting Fluorogenic Substrate

We developed a novel method for the rapid determination of lysozyme using a new fluorogenic substrate that consists of a cationic aluminum phthalocyanine ( tetra ( trimethylammonio ) aluminum phthalocyanine, TTMAAlPc ) , and an anionic mucopolysaccharide ( heparin, HP ) . We found that fluorescence from the cationic aluminum phthalocyanine, a red-region fluorescence probe, was quenched significantly in acidic media in the presence of low concentrations of anionic mucopolysaccharide heparin ( HP) bearing anionic sulfonic acid groups, because of induced aggregation. The practically non-fluorescent substrate degraded into small molecular fragments upon the hydrolysis of lysozyme, and thus the phthalocyanine molecules aggregated in HP were released, resulting in significant fluorescence recovery in the reaction system. This phenomenon forms the foundation of the proposed method. The reaction mechanism was determined using fluorescence spectroscopy and fluorescence anisotropy techniques. Factors that affected the determination were investigated. Under optimal conditions, the linear range was 0. 2-2 mg/L, and the detection limit was 0. 015 mg/L. The developed method is easy to operate and has good selectivity and sensitivity. This method was used in the analysis of practical samples of lysozyme, and the results were in agreement with those determined by a conventional turbidimetric method.
