3.An analysis on influencing factors of elders' medical consumption behavior under the back-ground of pension combined with medical service:A case study of Nanjing

Objective: Under the background of pension combined with medical service, for the elders to reasonably, effectively and fully utilize the medical resources, it is of vital significance to establish good medical con-sumptive behavior and consumption intention, to promote healthy old age and improve elders' health and happiness to help them cultivate a fine habit in their practical life. Methods: Based on the micro-survey data of investigation in Nanjing, this paper systematically analyzed the factors influencing elders' medical consumption behavior by using the theory of planned behavior and structural equation model. Results:Different factors were classified into behavioral at-titudes, subjective norms and perceived behavior control, and significant variables were found out through behavior model as well as though measuring their relative effects. Conclusion: There is no significant casual relationship be-tween the perceived behavior control and medical consumption behavioral intention. The elders' medical consumption behavior intention is affected by various social network relationships. The belief strength, normative belief, behavioral attitudes and subject norms can affect elders' medical consumption behavior.
4.Down-regulation of interphotoreceptor retinoid-binding protein and up-regulation of recoverin expression in rats with N-methyl-N-nitrosourea-induced retinal damages

Objective To investigate the effect of N-methyl-N-nitrosourea (MNU) on expressions of interphotoreceptor retinoid-binding protein (IRBP) and recoverin in rat retinal.Methods Thirty female Sprague-Dawley (SD) rats were randomly divided into five groups,including normal control group,and MNU treatment one day,three days,seven days and 10 days groups,each group had six rats.Rats in MNU-treatment groups received a single intraperitoneal injection of 40 mg /kg MNU,while rats in the normal control group received intraperitoneal injection of saline 5 ml/kg.Reverse transcription-polymerase chain reaction (RT-PCR) and immunofluorescence were used to detect the retinal expression of IRBP and recoverin.Results RT-PCR results indicated that retinal IRBP levels decreased after MNU injection compared with normal control (P<0.01),while recoverin levels increased (one day:P>0.05; three days:P<0.05; seven days:P<0.5; 10 days:P<0.05).Immunofluorescence assays demonstrated that normally IRBP protein is mainly in the inner and outer segments of photoreceptors.After MNU treatment,IRBP protein was detected in all retinal layers but much faint.Recoverin was expressed in the inner nuclear layer,inner plexiform layer and ganglion cell layer,and its expression was increased after MNU injection.Conclusion MNU suppresses IRBP expression and promotes recoverin expression in rat retina.
5.Clinical correlation of plasma N -terminal probrain natriuretic peptide and 6 min walk test performance in patients with chronic heart failure

Objective To explore the clinical effect and prognosis of plasma N -terminal probrain natriuret-ic peptide (NT -proBNP)and 6 min walk test performance (6 -MWT)in patients with chronic heart failure (CHF).Methods 100 patients with CHF were selected as observation group,and 50 healthy people were selected as control group.They were grouped by NYHA degree,28 cases were NYHA Ⅱ,56 cases were NYHA Ⅲ and 16 cases were NYHA Ⅳ.The plasma level of NT -proBNP,6 -MWT,left ventricular ejection fraction (LVEF),early diastolic peak mitral flow velocity to late diastolic peak mitral flow velocity (E /A)were measured.Results The plasma NT -proBNP level of CHF patients was positively correlated with NYHA degree (r =0.683,P <0.01);6 -MWT was neg-atively correlated with NYHA degree(r =-0.518,P <0.01).For CHF patients,significant correlation was observed between NT -proBNP and 6 -MWT (r =-0.789,P <0.05).Plasma NT -proBNP level was correlated with dias-tolic HF,systolic HF and mixed type heart failure (r =-0.678,P <0.01;r =-0.845,P <0.01;r =-0.759,P <0.01).Conclusion Plasma NT -proBNP,6 -MWT can serve as a marker for the detection and evaluation of heart failure.Meanwhile,the higher the NT -proBNP level,the shorter the 6 -MWT,the more serious the heart failure,the poorer the prognosis.
6.Effect of linoleic acid on lipopolysaccharide-induced release of inflammatory factors in macrophages of mice

Objective To evaluate the effect of linoleic acid on lipopolysaccharide (LPS)-induced release of inflammatory factors in the macrophages of mice.Methods The peritoneal macrophages obtained from C57BL/C mice were seeded in 24-well plates at a density of 4× 105 cells/well and in 6-well plates at a density of 2× 106cells/well.The cells were incubated and attached to the wall overnight in a 5% CO2 incubator in humidity at 37 ℃.The experiment was performed in 2 parts.Part Ⅰ The cells in 24-well plates were randomly divided into 5 groups (n =8 each) using a random number table:control group (group C);LPS group;3 different concentrations of linoleic acid groups (LA1-3 groups).The sterile anhydrous alcohol 1 μl was added in group LPS,0.1,0.5 and 1.0 mol/ml linoleic acid 1 μl were added in LA1-3 groups,respectively,and 30 min later 100 μg/ml LPS 1 μ,l was added in LPS and LA1-3 groups.The culture medium was collected at 6 h after LPS administration to measure the concentrations of tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6) in the supernatant by enzyme-linked immunosorbent assay.PartⅡ The cells in 6-well plates were randomly divided into 3 groups (n =6 each) using a random number table:control group (group C);LPS group;0.5 mol/ml linoleic acid group (group LA).The sterile anhydrous alcohol 1 μl was added in group LPS,0.5 mol/ml linoleic acid 1 μl was added in group LA,and 30 min later 100 μg/ml LPS 1 μl was added in LPS and LA groups.At 1 h after administration of LPS,the expression of Toll-like receptor 4 (TLR4) was determined by flow cytometry,and the expression of phosphorylated nuclear factor kappa B (NF-κB) p65 (p-NF-κB p65),phosphorylated extracellular signal-regulated protein kinase (p-ERK) and phosphorylated p38 mitogen-activated protein kinase (p-p38 MAPK) was determined by Western blot.Results Part Ⅰ Compared with group C,TNF-α and IL-6 concentrations in the supernatant were significantly increased in LPS and LA1 3 groups (P < 0.05).Compared with group LPS,TNF-α and IL-6 concentrations in the supernatant were significantly decreased in LA1 3 groups (P<0.05).Compared with group LA1,TNF-α and IL-6 concentrations in the supernatant were significantly decreased in LA2 and LA3 groups (P<0.05).Compared with group LA2,TNF-α and IL-6 concentrations in the supernatant were significantly decreased in group LA3 (P < 0.05).Part Ⅱ Compared with group C,the expression of TLR4,p-NF-κB p65,p-ERK and p-p38 MAPK in macrophages was significantly up-regulated in LPS and LA groups (P<0.05).Compared with group LPS,the expression of TLR4,p-NF-κB p65,p-ERK and p-p38 MAPK in macrophages was significantly down-regulated in group LA (P<0.05).Conclusion Linoleic acid can inhibit LPS-induced release of inflammatory factors in the macrophages of mice,and the mechanism may be related to the inhibition of TLR4 signaling pathway activation.
7.The effects of comprehensive control measures on intelligence of school-age children in coal-burning-borne endemic fluorosis areas

Objective To investigate the incidence of dental fluorosis,urinary fluoride level and intelligence of children who lived in the coal-burning-borne endemic fluorosis areas and to reveal the effects of comprehensive control measures on intelligence of children in this area.Methods Children aged 8-12 who lived in coal-burning-borne endemic fluorosis areas in Bijie City of Guizhou Province were selected and divided into two groups according to the duration of comprehensive treatments given:long treatment group (Xiaba Village and Zhongtun Village,furnace stovewas changed and comprehensive control measure of health education was carried out for more than 3 years) and short treatment group(Chadi Village and Maoliping Village,stoves were improved and health education time < 1 year).The children who lived in a non-fluorosis area were selected as controls in 2012.Dental fluorosis was diagnosed by the method of Dean; urinary fluoride was analyzed by the method of fluoride-ion selective electrode; and the intelligence quotient (IQ) was measured by Raven's Standard Progressive Matrices Test.Results The number of children surveyed in control group was 104,long treatment group was 298,short treatment group was 339,and the incidence rates of dental fluorosis were 0 (0/104),725%(216/298) and 85.2% (289/339),respectively,and the incidence rates of dental fluorosis in children lived in the endemic fluorosis areas were significantly increased compared with that of control group; the difference of incidence rates between long treatment group and short treatment group was statistically significantly(x2 =15.736,P < 0.01).Urinary fluoride content were (2.33 ± 0.18) and (3.03 ± 0.16)mg/L,respectively,compared with the control group[(1.34 ± 0.64) mg/L],the values in endemic fluorosis areas were significantly higher(F =306.53,P < 0.01).Above average IQ of children in the control group was 97.1% (101/104),which was significantly higher than that of long and short treatment groups; after a lengthy treatment,mental retardation detection rate was significantly lower in the low-age group,8-10 year-old ehildren(x2 =7.542,P < 0.01).Urinary fluoride content was negatively correlated with the level of IQ (r =-0.553,P < 0.01).Conclusions The intelligence development of children in coal-burning-borne endemic fluorosis area is significantly delayed.After a certain period of comprehensive treatment,the decreased level of cognition is inhibited and the mental retardation in the low-age group is improved.
8.Establishment of RP-HPLC detection method of N-isopropyl oxamate in the serum of plateau pikas.

OBJECTIVETo explore the intergrating of N-isopropyl oxamate and serum protein and establish a high performance liquid chromatography (HPLC) detection method of N-isopropyl oxamate (specific inhibitor of testis-specific lactate dehydrogenase (LDH-C4)) in the blood of plateau pikas.
METHODSTwenty highland pika 150-200 g, were randomly divided into two groups (n = 10): control group and inhibitor group. Different concentrations of N-isopropyl oxamate were added to examine the intergrating of N-isopropyl oxamate and serum protein. In order to determine its concentration in the pika blood accurately, we used the method of adding trypsin to incubate the serum first, followed by trichloroacetic acid treatment and detecting by HPLC. Results: When the concentrations of N-isopropyl oxamate in the pika serum were added to 0.05 mmol/L, 0.1 mmol/L, 1 mmol/L, 10 mmol/L, 16.7 mmol/L, 33.3 mmol/L and 100 mmol/L, the intergrating rates between N-isopropyl oxamate and plateau pika serum were 100%, 100%, 100%, 86.84%, 54.11%, 40.10% and 20.18%, respectively. The method established in this paper was good on recovery rates, precision and stability. A good linearity was obtained in the range of 0.0125-0.25 mmol/L. When the concentrations of N-isopropyl oxamate in the serum were added to 0. 15 mmol/L,0.3 mmol/L and 1 mmol/L, the recovery rates were 98.05%, 98.98% and 98.12%, respectively; the precision relative standard deviation( RSD) of concentrations were 1.17%, 0.92% and 0.83%, respectively; the stability relative standard deviation (RSD) of concentrations were 1.38%, 1.40% and 0.88%, respectively. The repeatability RSD of the method was 1.76%. Quantitative limit was 0.0125 mmol/L.
CONCLUSIONN-isopropyl oxamate has a strong affinity with plateau pika serum protein that can't be accurately determined with common HIPLC method. It can be accurately determined in the blood by adding trypsinto digest the serum protein first, followed by adding trichloroacetic acid to precipitate the protein.

9.Assessment of the quality of life of oral cancer patients after reconstruction with free anterolateral thigh perforator flaps.

OBJECTIVEThis study aims to evaluate the quality of life (QOL) of patients who underwent resection of oral cancer and reconstruction by free anierolateral thigh perforator flaps (ALTF).
METHODSA total of 32 patients with oral and maxillofacial malignancies who had undergone the resection of oral cancer and reconstruction by ALTF were retrospectively analyzed. At 12 months postoperatively, the QOL of these patients was assessed by using the 14-item oral health impact profile (OHIP-14) and the medical outcome study short form-36 (SF-36) questionnaires.
RESULTSA total of 32 questionnaires were collected. In SF-36, the highest scoring domains were bodily pain (78.58 ± 14.82), physical functioning (72.08 ± 27.86), and the role of physical (60.00 ± 42.63), whereas the lowest scoring domains were role-emotional (41.67 ± 39.62), followed by mental health (50.75 ± 13.07) and health transition (54.17 ± 21.75). In OHIP-14, the lowest scoring domains were social disability (34.50 ± 11.32) and handicap (36.04 ± 12.05), indicating the functional recovery was better; and the highest scoring domains were physical pain (73.50 ± 18.96) and psychological discomfort (60.17 ± 20.66), indicating the functional recovery was worse.
CONCLUSIONThe ALTF is an ideal selection for the reconstruction of oral defects after cancer resection. In using this flap, the basic social need of patients after surgery can be satisfied. Moreover, the appearance and the functions of chewing, deglutition, and speech can be restored in varying degrees. Thus, ALTF can improve the patients' QOL.

10.Effect of integrin beta1 on adhesion and migration of human trophoblast cells.

Although the mechanism by which migratory trophoblasts reach the spiral arteries is currently obscure, yet the process has been noted to involve the attachment, adhesion and migration of trophoblasts on the blood vessel walls. To test this, micropipette and flow chamber were used to measure quantitatively the adhesion forces and migration of early gestation human trophoblast cells (TCs) cultured on the glass slides coated with type I rat collagen or cultured with human umbilical vein endothelial cells (HUVECs). The results showed that the interdiction of integrin beta1 interaction remarkably reduced the adhesion forces of TCs to type I rat collagen or endothelial cells, and remarkably resisted the displacement of TCs induced by shear stress. By contact between TCs and endothelial cells, the TCs' adhesion force and TCs' resistance to shear stress were significantly enhanced. The results indicated that the contacts of TCs with endothelial cells enhanced the adhesion forces of human TCs, and regulated the migration of human TCs by shear stress.
