1.Effects of type Ⅳ collagen,MMP-9 and TIMP-1 gene expression on the initiation of early phase radiation pulmonary fibrosis

Objective To investigate the gene expression profiles of type Ⅳ collagen, matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) at the onset of radiation pulmonary fibrosis, so as to explore their possible roles in the pathogenesis of pulmonary radiation injury. Methods The whole lung was irradiated with 20Gy of 60Co ?-ray in C57 mice. At 1, 2 and 4 weeks after irradiation, both irradiated mice and corresponding number of normal control mice were sacrificed. Alterations in mRNA transcription of type Ⅳ collagen, MMP-9 and TIMP-1 were observed by RT-PCR at different time points post-irradiation. The mean optical density of electrophoretic bands was measured and analyzed by image analyzer. Result The mRNA expression of type Ⅳ collagen was up-regulated gradually after irradiation with the passage of time. The mean optical density of electrophoretic band was 0.202 at 1 week post irradiation and reached its peak value (0.340) at 4 weeks post irradiation; the mRNA expression of MMP-9 was up-regulated significantly and reached its peak value (0.730) at 1 week post irradiation. It began to lower at 2 weeks (0.592). The mRNA transcription of TIMP-1 began to increase slightly 1 and 2 weeks post irradiation (0.987) and reached its peak value (2.027) at 4 weeks post irradiation. Conclusion 60Co ?-ray can stimulate mRNA expression of type Ⅳ collagen, MMP-9 and TIMP-1 with different profiles at different time points. Complex interactions among them may result in early tissue remodeling after pulmonary radiation injury, and it bears close relation with subsequent pulmonary fibrosis.
2.Evaluation and cell warning information for clinical application of five-part differential hematology analyzer

Objective To investigate the technical charactenstics ot SYSMEX XS1000i 5-part differential automated hematological analyzer and its clinical applications.Methods 209 samples were analyzed with the analyzer of XS1000i and compared to the results from Beckman-Coulter LH750 analyzer.The main parameters from XS1000i,such as precision within-run,day to day precision and carryover contamination rates etc,were recorded respectively in 509 samples to compare the difference between the instrumental and optical examination.Results All variation coefficients of precision from XSI000i were within the manufacturer.The carryover contamination rates of WBC,RBC,Hb,HCT and PLT were 0.19%,0.93%,0,-0.88%,-0.76%,respectively.The parameters of XS1000i were correlated with the results of LH750 except basophil granulocyte.The correlation coefficients of WBC,RBC, HGB and PLT were 0.994 5,0.996 8,0.997 0 and 0.974 6 ,respectively.The sensitivity of warning flags in immature leukocytes was 100% and the specificity was 69.7%,the sensitivity of warning flags in atypical lymphocyte was 100% and the specificity was 66.7%.Especially in 3 leukocytopenia that was induced by chemotherapy in patients with leukemia who had only few immature cells existed in the peripheral blood,the parameters of XS1000i were positive,and correlated with the results of detection of minimal residual disease with flow cytometry.Conclusions The warning system of XSIO00i provides more valuable information for manual microscopic examination.If it is combined with flow cytometry,the advantages in detection of residual leukemic cell will be fully displayed.
5.Characteristics and implication of MMP-12 expression during the course of development of radiation pulmonary injuries

Objective To observe the dynamic changes in matrix metalloproteinase-12(MMP-12) expression during the course of development of radiation pulmonary injuries in order to get an insight of the roles played by MMP-12 during early tissue remodeling after radiation pulmonary injuries.Methods Twenty-four Wistar rats were randomly assigned into four groups(6 each): normal control group and 1,2 and 4 weeks after irradiation groups.The rats in the last three groups received irradiation of the whole lung with 20Gy of 60Co ?-ray,and they were sacrificed 1,2 and 4 weeks post irradiation.The gene expression of MMP-12 was determined by RT-PCR;the protein expression was detected by Western blot;MMP-12 activity was examined by gelatin zymography;the tissue distribution of MMP-12 was observed by immunohistochemistry;the degradation and collapse of elastin were observed after tissue elastin specific staining.Results The gene expression of MMP-12 began to increase 1 week post irradiation,then decreased at 2 weeks,and it reached its peak value at 4 weeks,while its protein expression was elevated again 2 weeks post irradiation.MMP-12 was mainly expressed in macrophages and activated fibroblasts of the alveolar septum.With the elapse of time,the lung injury aggravated gradually,with thickening of alveolar wall and septum,breakdown of the structure of alveoli,collapss of a part of the alveolar space,extensive breakdown of elastin in the basement membrane of lung,appearing as irregular fibrils distributed in the lung.Conclusion 60Co ?-ray irradiation can up-regulate the expression of gene and protein of MMP-12 significantly,implying that it may play a very important role in early remodeling process in of radiation pulmonary injuries,and initiate the process of pulmonary remodeling by degrading elastin and destroying the normal structure of basement membrane.
8.Expression of apoptosis inhibitor gene survivin in bladder carcinoma and its clinical implication

Objective To study the expression of survivin, an apoptosis inhibitor gene,in bladder carcinoma,and to investigate its clinical and pathological implications. Methods We detected the expression of survivin in 60 cases (48 men and 12 women) of bladder carcinoma and 10 cases of non-tumor bladder tissues (controls) by immunohistochemistry (SP) method.The mean age of the 60 cases at diagnosis was 59 years.The pathological grading showed Grade Ⅰ in 16 cases,Grade Ⅱ in 24 and Grade Ⅲ in 20.The clinical staging showed T 1 in 13 cases,T 2 in 15,T 3 in 21 and T 4 in 11.Of the 60 cases,21 developed relapse. Results The overall positive rate of survivin in 60 cases of bladder carcinoma was 60.0% (n=36).No survivin was found in 10 cases of non-tumor bladder tissues.The positive rate of survivin in Grade Ⅰ cases of bladder carcinoma was 37.5%(6/16),in Grade Ⅱ and Ⅲ cases of bladder carcinoma was 66.7%(16/24) and 70.0%(14/20);the difference of positive rates between Grade Ⅰ and Grade Ⅱ,Grade Ⅲ cases was significant (P0.05).The positive rate of survivin in cases of relapse was 80.9% (17/21). Conclusions Survivin may play an essential role in bladder carcinogenesis and serve as a marker for prognosis of bladder cancer.
9.GMP_(140) Expression in IgA Nephropathy

Objective In order to study the relationship between the platelet activation sign:P-selectin(GMP 140 ) expression and human IgA nephropathy.Methods Expression of platelet membrane glucoprotein Ⅱb/Ⅲa(GP Ⅱb ,GP Ⅲa ) in blood for IgA nephropathy patients(n=46) was investigated by flow cytometry methods.Results In IgA nephropathy the up-regulated GMP 140 expression in blood was significantly higher than that in normal humans.Conclusions GMP 140 might play an important role in pathogenesis of IgA nephropahty,and the level of GMP 140 in blood might predict renal outcome in IgA nephropathy.
10.The Effect of immunomodulatory therapy On CD4+CD25+ tregs in peripheral blood of patients with Sepsis

Objective To investigate the percentage of CD4 + C125 +Tregs in peripheral blood of patients with sepsis and its effect on cell immunity so as to unravel the effect of immunomodulatory therapy on it. Method Fourty patients with sepsis in ICU were randomly (random number) divided into experimental group and control group . The patients of experimental group were treated with Ulinastatin and immunoregulation agent (Thymosin αl) as well. The blood specimens were collected just before treatment, 3 days and 8 days after treatment. The percentages of CD4 + CD25 + Tregs and lymphocyte subsets were detected by using FCM (flow cytometry), and TNF-α, IL-6 and IL-10 assayed by using ELISA, and APACHE Ⅱ scores were calculated. Results Before treatment, the percentage of CD4 + CD25 + Tregs increased, and the number of lymphocytes and the percentage of T lymphocytes decreased, especially the CD4 + T lymphocytes and CD4+/CD8+ decreased more markedly, and the levels of IL-6 and TNF-α increased. After treatment,the percentage of CD4+ CD25 + Tregs was decreased, the number of lymphocytes and CD4 +/CD8 + increased, and the levels of APACHE Ⅱ score, IL-6 and TNF-α decreased especially in the experimental group decreased more significantly (P < 0. 05). Conclusions The percentage of CD4 + CD25+ Tregs in peripheral blood can reflect the immune status of patients with sepsis and become a novel indicator to estimate the progress of sepsis, and the immunity and prognosis of patients. Treating the patients with Thymosin αl and Ulinastatin can raise their immunity, decrease the levels of IL-6, TNF-α and APACHE Ⅱ score and improve their prognosis.
