Chinese Journal of Cancer Biotherapy 1995;0(03):-
Human tumor antigen elicit a cell-mediate immune response in the host have been identified. In this study, a pancreatic cancer cell line SW1990 associated apomucin was examined for the induction of tumor draining lymph node cells(TDLN) anti-tumor immune responses. The TDLN of patients with breast cancer(4 cases), gastric cancer(3 cases), colon cancer(3 cases) were isolated, cocultured with apomucin 50 ?g/ml and low dose rhIL-2 50U/ml in vitro, the cytolytic effects were measured by MTT assay. The results showed that: the proliferated lymphocytes can lysis LS174T colon cancer cells, KATO-Ⅲ gastric cancer cells, SW1990 pancreatic cancer cells and K562 leukemia cells. The most cytolytic effects were higher than LAK. This results might provide a rational basis for future attempts to elicit and manipulate apomucin vaccine for active specific immunotherapy or adoptive immunotherapy for adenocarcinoma.

Chinese Journal of Experimental Ophthalmology 2017;35(1):47-52
Background Optic neuritis is one of the common clinical neuro-ophthalmic diseases.Spectraldomain OCT (SD-OCT) is a valuable tool in assessing the thickness changes of retina, while enhanced depth imaging (EDI) OCT can further quantitatively and morphologically evaluate the changes of retina and choroid. The pathological mechanism of optic neuritis is unclear now. Objective This study was to quantitatively measure the retinal and choroidal thickness in early optic neuritis eyes by SD-OCT and EDI OCT.Methods A prospective cohort study was carried out in Tianjin Eye Hospital from July 2015 to May 2016.Twenty eyes of 20 patients with acute optic neuritis were enrolled as optic neuritis group and 22 eyes of 20 healthy subjects with matched age and gender were included in the normal control group. The mean thickness of retinal nerve fiber layer (RNFL) and choriod in superior, inferior, nasal and temporal quadrants at 3.4 mm around optic disc was measured, and the mean thickness of RNFL,ganglion cell layer (GCL),inner plexiform layer (IPL),inner nuclear layer (INL),outer plexiform layer (OPL),outer nuclear layer (ONL) and photoreceptor layer also was measured by EDI OCT.Pattern visual evoked potential (P-VEP) and visual field were examined in all the individuals, the correlations of mean defect (MD) with the thickness of RNFL,choroid and the thickness of RNFL,GCL,IPL,INL,OPL,ONL,photoreceptor layer at macular area were evaluated. Results The RNFL thickness values were (424.00±160.30),(428.40±169.83) and (108.15 ±50.66) μm in superior, inferior, nasal quadrants at 3.4 mm arear around optic disc in the optic neuritis group, which were significantly higher than (265.68±26.25),(283.27±52.81) and (72.68± 12.01) μm in the normal control group (t=4.571,3.814,3.190,all at P<0.01),and there was no significant difference between the two groups (t =0.849,P =0.401),and no significant differences were found in choroidal thickness of 4 quadrants between the two groups (all at P>0.05).The thickness of RNFL,GCL and IPL at 1 mm area around macula and the thickness of GCL,IPL,INL at 3 mm area around macula were evidently thining in the optic neuritis group compared with the normal control group (all at P<0.05).The latency of P100 wave was (133.15±11.11) seconds in the optic neuritis group and that in the control group was (94.59 ±4.38) seconds, showing a significnat difference between them (t =15.058,P<0.05).Positive correlations were found between MD and the RNFL thickess in superior, inferior, nasal quadrants at 3.4 mm arear around optic disc (r =0.649,0.649,0.635,all at P<0.05),however,no evidentlylinear correlations were found between MD and choroidal thickness in 4 quadrands (r =-0.120,-0.102,-0.415,0.120,all at P>0.05),and the thickness of RNFL,GCL,IPL,INL,OPL,ONL,photoreceptor layer at macular area was significantly linear correlated with MD.Conclusions EDI OCT can reflect the RNFL edema around optic disc and thining of various layers of retina at macular area in acuter optic neuritis eyes, however, the choroidal thickess is unchanged.EDI OCT is a useful tool in quantitative evaluation of retinal and choroidal thickness of early optic neuritis.

Objective To analyze the correlation between the expression of Abl interactor 1 (ABI1) and the clinicopathologic characteristics in colorectal adenocarcinoma tissues. Methods Immunohistochemistry was used to determine ABI1 expression in human colorectal adenocarcinoma tissues and matched adjacent tissues.A statistical analysis was used to determine the potential correlation between ABI1 expression and clinicopathological characteristics and prognosis. Results ABI1 is up-regulated in colorectal adenocarcinoma tissues compared with matched adjacent tissues (P =0.000),ABI1 expression is significantly correlated with infiltration (P =0.043) and differentiation (P =0.040),but not with sex, age, tumor location, clinical stage, lymph node metastasis and distant metastasis (P >0.05);It was also shown that ABI1 expression had a significant influence on prognosis (x2 =11.090,P =0.001).Multivariate analyses showed that high ABI1 expression is not an independent poor prognostic factor for overall survival (P =0.119).Conclusion ABI1 overexpression might act as pro-oncogene for patients with colorectal adenocarcinoma.

To compare the chemical change of Paeoniae Radix Alba (PRA) after vinegar-baking processing, as well as the effect of vinegar types exerted on the processing, 1H NMR-based metabolomic approach combined with multivariate statistical analysis was used to investigate the different metabolites between the raw and two vinegar-baked PRA. More than thirty metabolites were identified in the 1H NMR spectrum of PRA, and the multivariate statistical analysis showed that raw and two vinegar-baked PRA could be separated obviously. After vinegar-baking, the contents of isoleucine, lactate, alanine, arginine, albiflorin, and 5-hydroxymethyl furfural (5-HMF) elevated, while those of sucrose, paeoniflorin and its analogues (calculated by benzoate) decreased. The chemical compositions of two vinegar-baked PRA were also different. Shanxi vinegar- baked PRA showed higher levels of leucine, isoleucine, valine, and albiflorin, while rice vinegar-baked PRA contained more sucrose and paeoniflorin's analogues (calculated by benzoate). And the chemical changes in Shanxi vinegar-baked PRA were greater than those of rice vinegar-baked PRA. The results revealed the chemical differences between raw and vinegar-baked PRA, as well as the influence of vinegar type on processing, in a holistic manner, the results obtained suggested that the correlations between the chemical change and the drug action after processing, as well as the vinegar type used in processing, should be further studied.

Chinese Traditional and Herbal Drugs 1994;0(11):-
Objective To analyze the DNA methylation status of polyploid complex of Pinellia ternata. Methods Methylation sensitive amplified polymorphism(MSAP) technique was carried out to analyze the methylation status of polyploid complex of P.ternata. Thirty-four selective primer amplifications were used to check the status of cytosine methylation DNA samples. Results A total of 7 708 bands were obtained.Among them 5 636 bands, each representing a recognition site cleaved by one or both of the isoschizomers(Hpa Ⅱ and Msp Ⅰ),were amplified.Furthermore, methylation patterns varied among the four polyploids:heptaploid, octoploid, nonuploid, and decaploid.Total and full methylation levels in P.ternata were 54%-58% and 24.1%-24.3%.All types of MSAP patterns detected in the study belonged to two classes, type Ⅰ and Ⅱ. 52.5% of detected bands belonged to Type Ⅰ; Another 47.5% were type Ⅱ,which showed the methylation differences among the four polyploids. Conclusion The results demonstrate DNA methylation events occur in P.ternata and the general methylation levels are higher.
