1.Effects of halofuginone on maturation of dendritic cells and immune tolerance during homograft heart transplantation of rats

Objective To explore the role of halofuginone in immune regulation,observe the effects of halofuginone on maturation of dendritic cells and immune tolerance during homograft heart transplantation of rats,in order to provide new ways and means to treat transplantation tolerance and to explore its possible mechanism.Methods Rat bone marrow derived dendritic cells were isolated and the effects of halofuginone in LPS-treated DCs were investigated by flow cytometry;heart transplantation model of rats were established and the effect of halofuginone stimulated dendritic cells in the immune tolerance by pathological analysis and mixed lymphocyte reaction were observed.Results Flow cytometry results show that the expression of cell surface CD80 CD86,OX62 antigens were not high in control group,their expression increased significantly when stimulated by LPS for 24h,and decreased when stimulated by both Changshan ketone and LPS for 24h,and even lower than their expression levels in blank group,which showed that Changshan ketone has the ability to inhibit the maturation of dendritic cells induced by LPS.In rat heart transplantation model,dendritic cells stimulated with Changshan were injected into recipient rats in vivo,and receptor of rat transplantation rejection decreased significantly,which showed that dendritic cells stimulated with Changshan ketone may be used as a new method for cell therapy in treatment of transplant rejection.Conclusion This study systematically observes that halofuginone can inhibit the maturation of rat bone marrow-derived dendritic cells and lymphocyte proliferation for the first time in vivo and in vitro.
2.Clinical investigation of neonate food allergy in 10 cases

Objective To find out the clinical characteristics related to neonate food allergy.Methods Ten subjects were diagnosed as neonate food allergy at the Third Hospital of Peking University from January 201 3 to January 201 5.The following clinical data:the gestational age,genter,age of onset,feeding,clinical manifestations,white blood cell count,eosinophilic granulocyte count,C -reactive protein(CRP),routine stool test and occult blood test,blood spe-cific IgE of cow′s milk and clinical outcomes were investigated.Results There were 4 term infants,and 6 preterm in-fants,including 2 very -preterm subjects.The gender ratio between male and female cases was 73.The age of onset from 1 day after birth to 43 days after birth,clinical investigation of neonate food allergy in 1 0 cases,and the average time was 20.3 d,the median age was 1 8.5 d,and the SD was 1 3.38 d.Their rectified gestational ages were from 30 to 37 weeks in 6 cases at onset.Four infants received breast feeding,3 infants received formula milk feeding,and the other 3 infants received mixed feeding.At onset,the symptoms were almost involved in the digestive system.The white blood cell counts of lab test were almost normal in 1 0 subjects.And eosinophilic granulocytes were elevated in 8 subjects.CRP elevated in 1 case,and those of the other 9 subjects were normal.The cow′s milk specific IgE antibody was elevated in 1 subject,and that of the other 9 subjects was normal.Two subjects were performed opened food challenge test and the finding were positive.Conclusions Both the term infants and the preterm infants can have food allergy,but the age of onset ranged widely.Breast feeding infants can also be attacked.The symptoms of the digestive tract were the main clinical manifestations of such neonates.The open -food challenge test is beneficial to diagnose but difficult to practice in the neonates.Dietetic treatment is the only method can therapy the disease.And mostly the clinical outcomes are desirable.

4.Effect of Danhong injection combined with metoprolol tartrate tablets in menopausal women with arrhythmia

Objective To investigate the therapeutic effect of Danhong injection combined with metoprolol tartrate tablets on cardiac arrhythmia in climacteric women.Methods Retrospective a total of 115 patients with arrhythmia in The First People’s Hospital of Guiyang from May 2014 to May 2016 were divided into the western medicine treatment group and the traditional Chinese and western medicine treatment group.Western medicine treatment group were treated by metoprolol tartrate tablets, the traditional Chinese and western medicine treatment group were treated by Danhong injection combined with metoprolol tartrate tablets.The effect of two groups of patients after two months of treatment were observed,the heart rate variability,heart function index,sleep quality and the incidence of adverse reaction rate were compared between two groups of patients before and after treatment .Results After treatment,the effective rate of the traditional Chinese and western medicine treatment group was 98.33%, higher than the western medicine treatment group (P<0.05);the levels of SDNN,SDANN,RMSSD and PNN50 (%) were significantly higher in the the traditional Chinese and western medicine treatment group than those in the western medicine group (P<0.05);the levels of SV,LVPS and LVEF in the traditional Chinese and western medicine treatment group were higher than that in the western medicine treatment group ( P <0.05 );the sleep quality of the traditional Chinese and western medicine treatment group was significantly improved compared with the western medicine treatment group(P<0.05);the abdominal distension, nausea and vomiting,fatigue and cough incidence were no difference between the two groups.Conclusion Danhong injection combined with metoprolol tartrate tablets arrhythmia in menopausal women has a better therapeutic effect ,can obviously improve the patients with heart rate variability and cardiac function and adverse reactions occurred rate is low.
5.Expression and clinical significance of telomerase and hTERT in ovarian neoplasm tissues

Objective To discuss the expression and clinical significance of telomerase and hTERT in ovarian neoplasm tissues . Methods The activities of telomerase and the expression levels of hTERT mRNA were detected in the tissues of malignant ovarian neoplasm ,benign ovarian neoplasm and normal ovarian respectively ,and the results were analyzed .Results Comparing with the tis‐sues of benign ovarian neoplasm and normal ovarian ,the activity of telomerase and the expression level of hTERT mRNA increased in malignant ovarian neoplasm tissues ,and the differences were statistically significant(P< 0 .05) .Conclusion The telomerase plays an important role in the occurrence and development of malignant ovarian neoplasm .The detection of telomerase activity can help in early diagnosis of malignant ovarian neoplasm .

7.Protection against hydrogen peroxide-induced cytotoxicity in PC12 cells by guggulsterone

The present study is to investigate the protective actions of guggulsterone against the cytotoxicity produced by exposure to hydrogen peroxide (H2O2) in PC12 cells. It was evaluated by MTT [3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl-tetrazolium bromide] reduction assay, lactate dehydrogenase (LDH) release assay, and the release of nitric oxide (NO). ROS and Ca2+ in cells were evaluated by DCFH and Fura 2-AM, respectively. Mitochondrial membrane potential (MMP) was assessed by the retention of rhodamine 123 (Rh 123). Apoptosis and morphological alteration in PC12 cells were monitored with flow cytometry and electric microscope. Vitamin E, a potent antioxidant, was employed as a comparative agent. The results showed that preincubation of PC12 cells with guggulsterone (0.1-10 μmol·L-1) prevented cytotoxicity induced by H2O2. Extracellular accumulation of LDH, NO and intracellular accumulation of ROS, Ca2+ resulting from H2O2 were significantly reduced by guggulsterone. Incubation of cells with H2O2 caused a marked decrease in MMP, which was significantly inhibited by guggulsterone. The percentage of H2O2-induced apoptosis in PC12 cells was 24.3%, and decreased in the presence of guggulsterone (0.1-10 μmol·L-1) by .8.4%, 15 .9%, 11.8%, respectively. Guggulsterone exhibited comparable potency against oxidative stress induced by H2O2 in PC12 cells as that of vitamin E. The present findings showed that guggulsterone attenuated H2O2-induced cytotoxicity, extracellular accumulation of LDH and NO, intracellular accumulation of ROS and Ca2+, loss of MMP, and apoptosis, which may represent the cellular mechanisms for its neuroprotective action.
8.The relationship of angiotension-Ⅱ to portal hypertension in patients with liver cirrhosis

Objective:To study the relationships of portal hypertensions to vasoactive substances-angiotensin(AT-Ⅱ)in cirrhosis patients.Methods:48 patients with liver cirrhosis(LC)(including 18 patients with compensated LC and 30 patients with decompensated LC) and 32 normal controls(NC) were studied. Color duplex doppler ultrasonography was used to study portal venous and splenic venous blood flow(PVBF and SVBF).Also,the blood concentration of AT-Ⅱ was detected.Then,analyze the correlation between PVBF and SVBF to AT-Ⅱ.Results:Portal hyperdynamic exists in cirrhosis patients from beginning to the end. Low level of AT-Ⅱ in compensated LC patient has no relation with portal hyperdynamic, high level of AT-Ⅱ in decompensated LC has positive relation with portal hyperdynamic.Conclusion:This study has directed significance of clinical treatment and diagnosis.
9.Mental Health Status in 123 Patients with Hepatitis and Cirrhosis

Objective: To study mental health status in patients with hepatitis and cirrhosis. Methods: Symptom checklist 90 (SCL-90) were assessed to 123 patients with acute and chronic hepatitis and cirrhosis. Results: The factor scores of SCL-90 in those patients were significantly higher than those in Chinese norms (P﹤0.01), especially for somatization, depression, anxiety and hostility factors in patients with chronic hepatitis and cirrhosis. The factor scores existed elevatory tendency according to the severity of disease among the three groups, but the significant difference was only founded in psychotic factor. Multiple factor analysis showed that the type of liver diseases, age and marriage influenced the factor scores. Conclusion: Mental health problems exist in varying degrees in most patients with hepatitis and cirrhosis, and the type of liver diseases, age and marriage play an importance role in the factor scores of SCL-90.
