3.Isolation purification and characterization of NV-PP-1 from Nidus vespae

OBJECTIVES:To isolate and purify a new peptide named as NV-PP-1,from anti-inflammatory protein extracts of Nidus Vespae and to characterize it.METHODS:It was purified by ammonium sulfate precipitation,DEAE-Sephadex A50 ionexchange chromatography,Sephadex G-50 gel filtration and HPLC,respectively.Then it was characterized by HPLC on Protein-PakTM-60 column,IEF-HPCE(isoelectric focusing-high performance capillary electrophoresis),UV spectrum,amino acid analysis,and the N-terminal analysis with DNS-Cl.RESULT:Both HPLC on Protein-PakTM-60 column and IEF-HPCE showed its homogeneity.Its molecular weight was estimated to be 7.079 kD by HPSEC.UV spectrum indicated its maximum absorption at 274nm.Its PI was determined to be 4.08 according to IEF-HPCE.The terminal analysis showed its N-terminal amino acid was blocked.Amino acid analysis of NV-PP-1 indicated that it was rich in Ser,Glu,Gly.CONCLUSION:NV-PP-1 is the first peptide isolated from the anti-inflammatory protein extracts of Nidus vespae.
5.Rhabdomyolysis Associated with Viral Encephalitis

Viral encephalitis is infectious diseases caused by many kinds virus which infects the brain and produces a series of clinical manifestations,it is an common disease in children.Rhabdomyolysis is a sort of syndrome characterized by striated muscle necrosis and the release of intracellular muscle constituents into the circulation,most notably high level of creatine phosphokinase and myoglobin,and myoglobi-nuria,which is often complicated with acute renal failure.It was reported that rhabdomyolysis could be associated with many viral encephalidities,the clinician should pay attention to this condition because it was not rare especially,when hematuria appearred in the course of viral encephalidities,serum creatine phosphate kinase,serum and urine myohemoglobin should be detected to identify whether rhabdomyolysis was existed.Once the diagnosis was confirmed,it should be treated with fluid administration and basification in order to avoid acute renal failure,and to improve prognosis on these patients.This study reviewed the literature on viral encephalidities and rhabdomyolysis.
6.Central Nervous System Damage in Children with Mycoplasma Pneumoniae Pneumonia and Its Mechanism

Objective To observe the central nervous system(CNS)damage in children with mycoplasma pneumoniae pneumonia(MPP)and explore its mechanism.Methods A total of 325 cases of MPP were selected to investigate the incidence of CNS damage,and the levels of IL-8,IL-18 and soluble interleukin-2 receptor(sIL-2R)in both serum and cerebrospinal fluid(CSF)were detected in those patients with CNS complications.Children with viral encephalitis at hospital during the same period were taken as control group.Use SPSS 12.0 software to analyze the data.Results There were 22 cases(6.8%)of MPP who had CNS complications,presented mainly with fever,vomiting,headache and convulsion.The levels of IL-8,IL-18 and sIL-2R were higher in MPP patients who had CNS complications than those in control group,especially the level of IL-18 and sIL-2R in CSF(Pa

9.A study of the positioning errors of head and neck in the process of intensity modulation radiated therapy of nasopharyngeal carcinoma

Objective To investigate the positioning errors of head and neck during intensity-modulated radiation therapy of nasopharyngeal carcinoma.Methods Nineteen patients with middle-advanced nasopharyngeal carcinoma (T2-4N1-3M0), treated by intensity-modulated radiation therapy, underwent repeated CT during their 6-week treatment course.All the patients were immobilized by head-neck-shoulder thermoplastic mask.We evaluated their anatomic landmark coordinated in a total of 66 repeated CT data sets and respective x, y, z shifts relative to their position in the planning CT.ResultsThe positioning error of the neck was 2.44 mm±2.24 mm,2.05 mm±1.42 mm,1.83 mm±1.53 mm in x, y, z respectively.And that of the head was 1.05 mm±0.87 mm,1.23 mm±1.05 mm,1.17 mm±1.55 mm respectively.The positioning error between neck and head have respectively statistical difference (t=-6.58,-5.28,-3.42,P=0.000,0.000,0.001).The system error of the neck was 2.33,1.67 and 1.56 higher than that of the head, respectively in left-right, vertical and head-foot directions;and the random error of neck was 2.57,1.34 and 0.99 higher than that of head respectively.Conclusions In the process of the intensity-modulated radiation therapy of nasopharyngeal carcinoma, with the immobilization by head-neck-shoulder thermoplastic mask, the positioning error of neck is higher than that of head.
10.Potassium channel blocker(4-AP)enhances the radiosensitivity of human cervical cancer cell line HeLa to ~(252)Cf neutron ray

Objective To investigate the potential effect of 4-amino-pynidine(4-AP)on human cervical carcinoma cell line HeLa to 252Cf neutron ray.Methods HeLa cells were treated with 4-AP(0,1,5,10,20 mmol/L,respectively)and then exposed to 0.67 Gy 252Cf neutron ray.The effect of 4-AP on proliferation of HeLa cells was assessed by MTT assay.Cell apoptosis were detected by flow cytometry.Reverse transcription polymerase chain reaction(RT-PCR)and Western blotting were used to investigate the levels of HIF-1? mRNA and protein of HeLa cells treated with 4-AP(0,20 mmol/L)and 0.67 Gy 252Cf neutron ray.Results After HeLa cells treated with 0,1,5,10,20 mmol/L 4-AP were exposed to 0.67 Gy 252Cf neutron ray for 48 h,their inhibitory rate was 0,(38.81 ?3.45)%,(46.63?3.88)%,(63.58?6.19)%,(77.51?8.87)%,and their apoptotic rate was(3.15?0.85)%,(8.01?1.19)%,(16.00?1.58)%,(47.20?3.18)%,(62.56?4.27)%,respectively,suggesting 4-AP may inhibit the proliferation of HeLa cells and induce their apoptosis in a dose-dependent manner(P0.05).However,after treated with 20 mmol/L 4-AP and exposed to 0.67 Gy 252Cf neutron ray for 24,48 and 72 h,the optical density ratio of HIF-1? mRNA/?-actin of HeLa cells was(0.401?0.121),(0.364?0.142),(0.257?0.137),(0.165?0.099),while the optical density ratio of HIF-1? protain/?-actin were(0.879?0.117),(0.645?0.115),(0.312?0.114),(0.130?0.118),showing that the levels of HIF-1? mRNA and protein both declined(P
