1.ADR Reports of Our Hospital in 2008: Analysis of 113 Cases

OBJECTIVE: To investigate the characteristics and general pattern of the adverse drug reactions (ADR) occurred in our hospital. METHODS: A total of 113 ADR cases reported by our hospital in 2008 were analyzed statistically in respect of patients’age and sex, route of administration, dosage form, category of drug, organs and systems involved in ADR and clinical manifestations, etc. RESULTS: The ADR were mainly induced by venous administration (59.29%). Anti-infective drugs dominated the first place on the list of ADR incidence rate (46.90%) followed by TCM preparations (23.01%). The most common manifestation of the ADR was lesion of skin and its appendants(37.17%). CONCLUSION: Great importance should be attached to ADR monitoring to reduce or avoid the occurrence of ADR.

4.Role of glucose transporters in apoptosis induced by sodium butyrate

Objective The regulation on glucose transporters(GLUT1~GLUT5)by sodium butyrate was observed,and the regulation on bax and bcl-x/l by sodium butyrate and glucose was studied.This helped to learn about the possible mechanism of apoptosis of cancer cells induced by sodium butyrate.Methods The expression of GLUT1~GLUT5 mRNA was detected by RT-PCR.The expression of bax and bcl-x/l was detected by immuocytochemistry.Cell apoptosis was detected by TUNEL assay.Results In HT-29 cell line,sodium butyrate downregulated the expression of GLUT1 mRNA and the expression of bcl-x/l and induced cancer cell apoptosis,but did not regulate the expression of GLUT2,GLUT3,GLUT5 and bax.GLUT4 was not detected in HT-29 cell line.And when treated with sodium butyrate,the increase of concentration of glucose could increase the expression of bcl-x/l and reduce the effect of apoptosis induced by sodium butyrate.Without treatment with sodium butyrate,the change of glucose concentration only had a feeble regulation on apoptosis.Conclusion Sodium butyrate can obviously downregulate the expression of GLUT1 and downregulation of the expression of GLUT1 is associated with the apoptosis induced by sodium butyrate in HT-29 cell line.
5.Flavonoids and spiro-lactones from Hypericum hookerianum

Objective To study the chemical constituents in plants of Hypericum hookerianum and their activities by pharmacological experiment. Methods Cytotoxicity screening of the extracts from H. hookerianum were carried to isolate and purify the chemical constituents. The chemical structures were (identified) by physicochemical properties and spectral data analysis. Results Seven compounds were isola-(ted) and (established) as quercetin (Ⅰ), luteolin (Ⅱ), apigenin (Ⅲ), hyperin (Ⅳ), astragalin (Ⅴ), hyper-(olactone) A (Ⅵ), and hyperolactone C (Ⅶ). Conclusion All the compounds are isolated from H. hookerianum for the first time. Compounds Ⅲ and Ⅴ are isolated from the plants of Hypericum Linn. for the first time. In addition, the fractions by chlorform and ethylacete extracting, quercetin, and luteolin possess the cytotoxicity.
6.Using mimicry epitopes from phage displayed peptide library as substitute antigen for cyclophilin A in ELISA for detection of anti-CyPA autoantibody

Objective To use mimicry epitopes from phage displayed peptide library as substitute for recombinant human cyclophilin A (rhCyPA) in detection of CyPA autoantibody in sera of patients with systemic lupus erythematosus (SLE).Methods Monoclonal antibody D4 and E6 were used as the capture antibody; the Ph.D.-12 p hage display peptide library was screened by biopanning. Ten phage clones with h igh affinity for McAb D4 and E6 (five for each) were chosen. Their inserts were sequenced. The amino sequence of mimicry epitope was subsequently reasoned out. Selected 12 peptide and previously obtained 7 peptide mimicry epitopes were used as coated antigen and compared with rhCyPA in ELISA for anti-CyPA autoantibody .Results Nine of ten phage clones with high affinity for McAb D4 and E6 (five for D4, four for E6) shared a consensus amino sequence HW EYTTSPHPRL. In comparative study of ELISA, 56 serum samples from SLE patients we re tested for anti-CyPA autoantibody, 28 samples were positive (50%) with 12 pe ptide mimicry antigen and 24 were positive (43%) with rhCyPA, 4 of 34 (11%) samp les from those with other autoimmune disorders were positive with both antigens, the coincidence rate between two methods was 95 6%. The research also showed t hat 12 peptide mimicry antigen has the advantages of rhCyPA and 7 peptide mimicr y antigen in detecting anti-CyPA autoantibody. Conclusions The specific 12 peptide screened from phage-displayed peptide library by ant i-CyPA monoclonal antibody can mimic epitope information of cyclophilin A well and be used as substitute for rhCyPA in ELISA.

The criterion of injury severity is an important part of evaluation,which can affect the treatment for closed fracture.The reaction in soft tissue healing process contains microvascular responses and inflammation.Take soft tissue with low immunity as a incision will lead to wound dehiscence and deep infection.Therefore,realization of injury severity lays a foundation for effective treating with closed fracture.Many treatment methods,such as splint fixation,cryotherapy,compression or delayed operation,can prevent soft tissue from a second injury,and promote self-repair ability prior to operation.The newly development surgical tech-nique can improve efficacy of therapy,meantime,decrease the incidence of complications.
