1.The cause of atherosclerosis:secretory dysfunction in vascular endothelial cells and proliferation of smooth muscle cells
Chinese Pharmacological Bulletin 2010;26(2):155-158
Vascular endothelial cells and smooth muscle cells, structurally and functionally closely related, are two main cells of vascular wall. While the interaction between cycling factors and vascular wall cells can lead to the occurrence of atherosclerosis. The injury of endothelial cells results in secretory dysfunction and abnormal proliferation of smooth muscle cells which narrows the cavity of blood vessel and cause spasm. This is the common pathological basis of the occurrence and development of atherosclerosis and other vascular diseases. In the pathological state of atherosclerosis, the proliferation of smooth muscle cell will be affected by what the vascular endothelial cells secrete, like growth factors, cytokines, and vasoactive substances. This paper gives a comprehensive overview of the latest research advances in this field.
2.Advances in research on relationship between Hedgehog signal pathway and gastroenteric tumor
China Oncology 2006;0(11):-
Hedgehog(HH),a vital endodermal signal, plays a critical role in the embryonic development of the GI system.During mammalian embryonic development, deficiency of HH signaling leads to gastrointestinal malformations.Nevertheless, normal presence of HH family is also found in the adult gut,and HH signaling now may be important for homeostasis of the gut in such a case. However, many researches revealed that the HH signal's involved in gastrointestinal carcinogenesis and its role is altered in various tumors due to various mechanisms. This review focused on the progress of HH signal pathway in gastrointestinal tumors.

Objective To summarize the preliminary experience of coils, Onyx and covered stents for the treatment of carotid-cavernous sinus fistula (CCF). Methods Of the 25patients with CCF, 22 were traumatic CCF, and 3 were spontaneous CCF; the fistula orifices of 14 patients were located in the cavernous segment of right internal carotid artery (ICA), and 11were located in the cavernous segment of left ICA. According to the Barrow classification,Types A, C, and D were 22,2, and 1, respectively. Eighteen patients were treated with Guglielmi detachable coils alone, 1 was treated with Guglielmi detachable coils and Onyx, 2 were treated with Onyx alone, and 4 were treated with covered stents. Results Intracranial bruit disappeared immediately in all patients after the procedures. Postoperative angiography showed complete occlusion of the fistula orifices, the symptoms disappeared after 1 to 20 days, and there were no death and procedure-related complications. The recanalized ICAs were patent in patients (12/18) treated with coils, and the ICAs were patent in 4 patients treated with the covered stents. The ICAs were patent in 2 patients treated with Onyx, the ICA was patent in 1 patent treated with detachable coils and Onyx. One patient with traumatic CCF treated with coils had clinical recurrence, and was cured after re-embolization with coils. The remaining patients were followed-up for 6 to 24 months, and no recurrence of clinical symptoms was found. The followup cerebral angiography of 6 patients was obtained, and no CCF recurrence was observed. Conclusion Endovascular treatment with Guglielmi detachable coils, Onyx, and covered stents is a safe and useful method for closing CCFs.

Objective To evaluate the dose distribution of target volume and normal tissues with different treatment planning such as three dimensional conformal radiotherapy(3DCRT) ,simplified intensity modulated radiotherapy(sIMRT) ,and intensity modulated radiotherapy (IMRT) for patients with radically resected rectal cancer. Methods Ten male patients with stage Ⅱ and Ⅲ rectal cancer after radical resection (Dixon surgery) were enrolled in this study. 3-field or 5-field 3DCRT,slMRT and 5-field or 7-field IMRT plans were performed for each patient. The dose distributions of target volume and normal tissues, conformal index(CI) and heterogeneous index(HI) were analyzed using the dose-volume histogram(Dvit). The prescription dose was 50 Gy in 25 fractions. Results The CI for PTV of IMRT and sIMRT was superior to 3DCRT. Conversely, the HI for PTV of 3DCRT was superior to sIMRT and IMRT. sIMRT and IMRT can protect the organs at risk better than 3DCRT. The mean of total MU for 3DCRT3f,3DCRT5f, sIMRT, IMRT5f and IMRTT7f was 482±13,504±11,455±42,841±36 and 884±46, respectively. Conclusions Comparing with 3DCRT plans and IMRT plans, sIMRT plan was the optimal plan for clinical practice. All of the three radiotherapy techniques can protect the rectal stump and anal canal well with the prescription dose of 50 Gy.

Objective:To investigate the numbers and distributions of dendritic cells and the expressive intensity of CD1c,S 100 in different human ovarian carcinoma tissues, and to discuss the relationship between them and the mechanism of development. The study can provide the experimental date of biological therapy of ovarian carcinoma. Methods:The samples of 31 patients with ovarian carcinoma were analyzed by ABC immunocytochemistry and computer imaging techniques. Results:The results showed that the number of positive dendritic cells in ovarian carcinoma reduced greatly with significant and the expressive intensity of CD1c,S 100 was weaker in ovarian carcinoma than in normal ovarian tissues. Conclusion:The total expression of dendritic cells phenotypic antigens was reduced in ovarian epithelial carcinoma, which may play an important role in initiating immunity against ovarian carcinoma.

Objective To investigate the expression of interleukin 18 (IL-18) in liver tissues of patients with chronic hepatitis B (CHB) and its clinical significance. Methods Totally 172 CHB patients admitted in the Second Affiliated Hospital of Anhui Medical University from January 2012 to December 2012 were enrolled. The tissue specimens were obtained by liver biopsy from all patients, and 22 normal liver specimens were also collected as controls.Liver function, serum markers of HBV and HBV DNA load were measured and liver inflammation grades and fibrosis stages were assessed.Immunohistochemical method was used to detect the expression of IL-18 in liver tissues.Spearman rank correlation was used to analyze the correlations between IL-18 expression and the above indicators. Results IL-18 was mainly expressed in cytoplasm of liver cells. It was positive or strong positive in liver cells of healthy people, but weakly positive in liver cells of CHB patients. The difference of IL-18 expression between CHB patients and healthy people was of statistical significance (x2 =42.124,P < 0.05).In CHB patients, the expression of IL-18 was gradually enhanced with the increasing of ALT,inflammation grades and fibrosis stages (r =0.182,0.173 and 0.068,P <0.05),but it was not correlated with HBV DNA load and HBeAg (r =-0.05 and 0.041,P >0.05).Conclusion In CHB patients, the expression of IL-18 may be involved in liver injury and fibrosis formation.

Objective The study aimed to investigate the effect of lipoxin A4 (LXA4) on lipopolysaccharide (LPS)?induced oxidant stress in human renal tubular epithelial cells (HK2 cells) and possible underlying mecha?nisms. MethodsHK2 cells were divided into three groups: Control ,LPS and LPS+LXA4 groups. After cells were treated with indicated conditions, morphological changes were observed. The expressions of Nrf2 were detected by immunofluorescence and cells were collected for RT?PCR experiments. Results HK2 cells seemed disrupted and necrotic with the administration of LPS. However ,LXA4 could prevent cells from injury induced by LPS. LPS decreased Nrf2 expression and promoted it to translocate to cytoplasm ,while LXA4 could increase its expression and promote it to translocate to nucleus. Moreover ,LPS could decrease Nrf2 and its downstream molecule mRNA expressions, but LXA4 could reverse this effect. Conclusion Our results demonstrated that LXA4 effectively inhibit?ed HK2 cell oxidant stress via Nrf2 pathway.
