1.Comparative analysis of the MRI features between cholangiocarcinomas associated with clonorchiasis and with hepatolithiasis

Objective To explore the relationships between cholangiocarcinoma and clonorochiasis or hepatolithiasis,and to compare their MRI features.Methods The MRI features in 37 patients with cholangiocarcinoma confirmed by surgery and pathology were retrospectively analyzed,including clonorchiasis with cholangiocarcinoma in 21 (group A)and hepatolithiasis with this tumor in 16 (group B).The analyzed parameters included the location,size,shape,signal intensity and enhancement pattern of the tumor and the characteristics of bile duct dilatation.Results In group A,the tumors were located in the right lobe in 12 (57.2%),left lobe in 4 (19%),and porta hepatic in 5 (23.8%).In group B,those were located in the right lobe in 3 (18.7%),left lobe in 9 (56.3%),and porta hepatic in 4 (25%).There was a significant difference in the left/right lobe between two groups (P<0.05),however,no significant difference was found in porta hepatic (P>0.05).The MRI showed the tumors with low signal on T1 WI and high or heterogeneous signal on T2 WI,and with peripheral enhancement in early stage and delayed centric enhancement in 34 patients.Diffuse saccular dilatations of terminal intrahepatic bile ducts were showed in 1 6 (7 6 .2%)in group A and 1 (6 .2%)in group B,meanwhile the dilatations of bile ducts within tumors were showed in 2 (9.5%)in group A and 13 (81.3%)in group B,exhibiting significant differences between the two groups (P<0.05).Conclusion Clonorochiasis and hepatolithiasis are related with the cholangiocarcinoma.No differences in MRI features of cholangiocarcinoma are found between the two groups,however significant differences in location and the features of bile duct dilatation are observed.
2.Expression of promyelocytic leukemia gene in peripheral blood lymphocytes from patients with psoriasis vulgaris

Objective To detect the expression of promyelocytic leukemia(PML)gene in peripheral blood lymphocytes from patients with psoriasis vulgaris,and to investigate the relationship between PML gene and psoriasis.Methods This study included 50 patients with vulgaris psoriasis who were aged between 18 and 65 years and had never received systemic treatment,as well as 45 healthy controls.Peripheral blood samples were collected from these subjects,and lymphocytes were separated by density-gradient technique.Then,RNA was extracted from the lymphocytes by using RNAprep pure blood kit followed by real time fluorescence-based quantitative reverse transcription-PCR for the detection of PML mRNA expression.Flow cytometry was performed to quantify the expression of PML protein in the lymphocytes.Independent samples t test was carried out to compare the differences in PML expression between the patients and controls.Results The relative quantity (RQ)of PML mRNA was 14.98 ± 3.64 in peripheral blood lymphocytes from the patients,significantly higher than that in the healthy controls(5.50 ± 1.10,t =16.79,P < 0.05).Similarly,a significant increment was observed in the fluorescence intensity of PML protein in peripheral blood lymphocytes from the patients compared with the healthy controls(3.13 ± 0.27 vs.2.43 ± 0.21,t =6.93,P < 0.05).Conclusions PML is overexpressed at the protein and mRNA levels in peripheral blood lymphocytes from patients with psoriasis vulgaris,hinting that PML may play a certain role in the development of psoriasis.
3.Target points: a discussion on acupuncture treatment of primary trigeminal neuralgia.

In this paper, issues of acupuncture using target points and depth of needling were discussed based on the theory of traditional Chinese medicine acupoints and combined with anatomical and neurological research of Western medicine. The theoretical evidence of acupuncture on nerve foramen and ganglion and the reasons for being nonstandard operating on acupoints were analyzed. This study summarized the method of using acupuncture at Xiaguan (ST7), Cuanzhu (BL2), Sibai (ST2) and Jiachengjiang acupoints to align with the spheno-palatine ganglion and additional nerve foramen (supraorbital, infraorbital and mental foramina) to treat primary trigeminal neuralgia. This study adhered to the Standards for Reporting Interventions in Clinical Trials of Acupuncture and helped in development of study standardization for acupuncture processes. The authors hoped that should help in significantly diminishing risk and improving therapeutic efficacy in clinic.

5.The effects of pioglitazone on the Kupffer cells of nonalcoholic fatty liver disease in rats

Objective To explore the preventive effect and mechanism of pioglitazone on nonalcoholic fatty liver disease in rats.Methods Thirty six male Sprague-Dawley ( SD) rats were randomly divided into normal control group , high-fat diet group , and pioglitazone intervention group , with 12 rats in each group .Normal control group fed with standard diet for eight weeks , and the rest two groups fed with high fat diet .Pioglitazone intervention group fed with pioglitazone in the four to eight weeks , the rest two groups in the same period fed with distilled water .Fasting serum alanine aminotransferase , aspartate aminotransferase , total cholesterol , triglyc-erides, fasting glucose , and fasting blood insulin levels were determined .The fasting insulin resistance index was calculated .Routine histological features of hepatic section were observed by hematoxylin eosin (HE) staining.The shape change of Kupffer cells in the liv-er was detected .The levels of tumor necrosis factor ( TNF)-αand nitric oxide ( NO) secreted by Kupffer cells were measured .Results The fasting insulin resistance index , triglycerides , and total cholesterol in rats of high-fat diet group were significantly higher than in normal control group ( P <0.05 ) .Histopathological examination showed hepatocellular macrove sicular steatosis , lobular inflammatory cell infiltration, and inflammatory cell infiltration .Hepatic Kupffer cell morphology change , its produced TNF, NO and liver tissue pa-thology changes were positively correlated ( P <0.05 ) .The fasting insulin resistance index , triglycerides , total cholesterol was signifi-cantly lower in rats of pioglitazone intervention group than the high-fat diet group ( P <0.05 ) .The liver tissue structure was obviously improved pioglitazone intervention group rats , morphology and function of liver Kupffer cells were normal .Conclusions Pioglitazone can prevent the nonalcoholic fatty liver disease induced by high fat diet .The mechanism may be related to improve insulin resistance , reduce blood fat , and regulate function of Kupffer cells .
6.Determination of Stilbence Glucoside in Polygonum Multiflorum by HPLC

Objective:To determine the content of stilbene glucoside in polygonum multiflorum by HPLC. Methods: An Agilent Eclipse Plus C18(100 mm ×4.6 mm,3.5 μm)column was adopted with acetonitrile-water(18∶82) as the mobile phase at a flow rate of 1. 0 ml·min-1 . The detection wavelength was set at 320nm and the injection volume was 8 μl. Results:The linear range of stilbene glucoside was 38-237. 5 μg·ml-1(r=0. 999 4)with the average recovery of 98. 32%(RSD=1. 59%). Conclusion: The method is simple, sensitive and specific in the content determination of stilbene glucoside in polygonum multiflorum.
7.The role of Kupffer cells disorder on the occurrence and development of Nonalcoholic Fatty Liver Disease in rats

Objective To explore the role of Kupffer cells( KCs)disorder on the occurrence and development of Nonalcoholic Fatty Liver Disease ( NAFLD) by means of rat model fed with high-fat diet. Methods Twenty-four male SD rats were random divided into model group ( n =12) and normal group( n = 12) , with a high-fat diet and standard diet for 12 weeks. The levels of body weight, liver weight, alanine aminotransferase( ALT), aspartate aminotransferase( AST) , triglyceride(TG) , and total cholesterol( TC) were measured. Routine histological features of hepatic section were observed by H. E staining. The shape changes of KCs in the liver were detected, and the levels of tumor necrosis factor-α(TNF-α) and nitric oxide (NO) secreted by KCs were measured. Results The weight, liver index, the levels of ALT, AST, TG, TC, and the levels of TNF-α and NO secreted by KCs in model group were higher than that in the normal group ( P < 0. 05). Histopathological examination showed hepatocellular macrovesicular steatosis, lobular inflammatory cell infiltration and necrosis. Compared with normal group, the shape and function of KCs in the liver changed largely. Furthermore, these changes of KCs were in accordance with the degree of steatosis, inflammation and necrosis in the liver of the model group. Conclusions The shape and function of KCs changed significantly in NAFLD induced by high-fat diet, and KCs disorder might be involved in the pathogenesis of NAFLD.

Objective To explore the effect of medicated serum of Sangleng and Eshu on the expression of vascular endothelial growth factor and vascular endothelial cell proliferation induced by VEGF in vitro. Methods Medicated serum of Sangleng and Eshu was used to culture human umbilical vein endothelial cell (HUVEC-1) induced by VEGF. The morphologic changes of HUVEC-1 were observed with phase contrast microscope, and cell proliferation was detected by MTT method, and the expression of vascular endothelial growth factor protein and mRNA in endothelial cells was detected by Western blotting and RT-PCR. Results The medicated serum of 5.0, 2.5 g?kg~ -1 ?d~ -1 Sangleng and Eshu could cause arrangement disorder in the normal umbilical vein endothelial cells. Objective To observe the iodine state of patients with autoimmunity thyroid disease(AITD) in Shaanxi province,and to discuss the relationship between the metabolism of iodine and the occurrance,development and prognosis of AITD.Methods A total of 388 cases were collected,among whom 283 were patients with AITD,and 105 were patients with non-AITD.Questionnaires were designed to determine the general state of the patients,the size,shape and texture of the thyroid,the urinary concentration of iodine,the function of thyroid and the autoantibody of thyroid.The data were processed by SPSS12.0.Results The mean urinary concentrations of iodine in patients with GD,with HT,and the control group were 212?g/L,245.4?g/L,and 197.8?g/L respectively,which were statistically different.In the HT group,wich different urinary concentrations of iodine,there were no statistical difference of TGAb,TMAb,and HS-TSH.Conclusion Excessive in-take of dietary iodine may be related with AITD.
